We propose a fully automated approach for multiclass skin lesion segmentation and classification by using the most discriminant deep features. First, the input images are initially enhanced using local color-controlled histogram intensity values (LCcHIV). Next, saliency is estimated using a novel Deep Saliency Segmentation method, which uses a custom convolutional neural network (CNN) of ten layers. The generated heat map is converted into a binary image using a thresholding function. Next, the segmented color lesion images are used for feature extraction by a deep pre-trained CNN model. To avoid the curse of dimensionality, we implement an improved moth flame optimization (IMFO) algorithm to select the most discriminant features. The resultant features are fused using a multiset maximum correlation analysis (MMCA) and classified using the Kernel Extreme Learning Machine (KELM) classifier. The segmentation performance of the proposed methodology is analyzed on ISBI 2016, ISBI 2017, ISIC 2018, and PH2 datasets, achieving an accuracy of 95.38%, 95.79%, 92.69%, and 98.70%, respectively. The classification performance is evaluated on the HAM10000 dataset and achieved an accuracy of 90.67%. To prove the effectiveness of the proposed methods, we present a comparison with the state-of-the-art techniques.There is no standardized or validated definition or measure of "child-appeal" used in food and beverage marketing policy or research, which can result in heterogeneous outcomes. Therefore, this pilot study aimed to develop and validate the child-appealing packaging (CAP) coding tool, which measures the presence, type, and power of child-appealing marketing on food packaging based on the marketing techniques displayed. Children (n = 15) participated in a mixed-methods validation study comprising a binary classification (child-appealing packaging? Yes/No) and ranking (order of preference/marketing power) activity using mock breakfast cereal packages (quantitative) and focus group discussions (qualitative). The percent agreement, Cohen's Kappa statistic, Spearman's Rank correlation, and cross-classification analyses tested the agreement between children's and the CAP tool's evaluation of packages' child-appeal and marketing power (criterion validity) and the content analysis tested the relevance of the CAP marketing techniques (content validity). There was an 80% agreement, and "moderate" pairwise agreement (κ [95% CI] 0.54 [0.35, 0.73]) between children/CAP binary classifications and "strong" correlation (rs [95% CI] 0.78 [0.63, 0.89]) between children/CAP rankings of packages, with 71.1% of packages ranked in the exact agreement. The marketing techniques included in the CAP tool corresponded to those children found pertinent. Pilot results suggest the criterion/content validity of the CAP tool for measuring child-appealing marketing on packaging in accordance with children's preferences.Cortisol, a stress hormone, plays key roles in mediating stress and anti-inflammatory responses. As abnormal cortisol levels can induce various adverse effects, screening cortisol and cortisol analogues is important for monitoring stress levels and for identifying drug candidates. A novel cell-based sensing system was adopted for rapid screening of cortisol and its functional analogues under complex cellular regulation. We used glucocorticoid receptor (GR) fused to a split intein which reconstituted with the counterpart to trigger conditional protein splicing (CPS) in the presence of targets. CPS generates functional signal peptides which promptly translocate the fluorescent cargo. The sensor cells exhibited exceptional performance in discriminating between the functional and structural analogues of cortisol with improved sensitivity. Essential oil extracts with stress relief activity were screened using the sensor cells to identify GR effectors. The sensor cells responded to peppermint oil, and L-limonene and L-menthol were identified as potential GR effectors from the major components of peppermint oil. Further analysis indicated L-limonene as a selective GR agonist (SEGRA) which is a potential anti-inflammatory agent as it attenuates proinflammatory responses without causing notable adverse effects of GR agonists.The rapid growth of nanoparticle-based therapeutics has underpinned significant developments in nanomedicine, which aim to overcome the limitations imposed by conventional therapies. Establishing the safety of new nanoparticle formulations is the first important step on the pathway to clinical translation. We have recently shown that plasma-polymerized nanoparticles (PPNs) are highly efficient nanocarriers and a viable, cost-effective alternative to conventional chemically synthesized nanoparticles. Here, we present the first comprehensive toxicity and biosafety study of PPNs using both established in vitro cell models and in vivo models. Overall, we show that PPNs were extremely well tolerated by all the cell types tested, significantly outperforming commercially available lipid-based nanoparticles (lipofectamine) used at the manufacturer's recommended dosage. Supporting the in vitro data, the systemic toxicity of PPNs was negligible in BALB/c mice following acute and repeated tail-vein intravenous injections. PPNs were remarkably well tolerated in mice without any evidence of behavioral changes, weight loss, significant changes to the hematological profile, or signs of histological damage in tissues. Alpelisib PPNs were tolerated at extremely high doses without animal mortality observed at 6000 mg/kg and 48,000 mg/kg for acute and repeated-injection regimens, respectively. Our findings demonstrate the safety of PPNs in biological systems, adding to their future potential in biomedical applications.
