Traditionally, there has been a trade-off in spectroscopic measurements between high resolution, broadband coverage, and acquisition time. Originally envisioned for precision spectroscopy of the hydrogen atom in the ultraviolet, optical frequency combs are now commonly used for probing molecular ro-vibrational transitions throughout broad spectral bands in the mid-infrared providing superior resolution, speed, and the capability of referencing to the primary frequency standards. Here we demonstrate the acquisition of 2.5 million spectral data points over the continuous wavelength range of 3.17-5.13 µm (frequency span 1200 cm-1, sampling point spacing 13-21 MHz), via interleaving comb-tooth-resolved spectra acquired with a highly-coherent broadband dual-frequency-comb system based on optical subharmonic generation. With the original comb-line spacing of 115 MHz, overlaying eight spectra with gradually shifted comb lines we fully resolve the amplitude and phase spectra of molecules with narrow Doppler lines, such as carbon disulfide (CS2) and its three isotopologues.This study evaluated the effects of different concentrations (1, 10, 25, 50, and 100 µM) of the antioxidant N-(2-mercaptopropionyl)-glycine (NMPG), during the culture of in vitro-fertilized porcine oocytes. While the highest concentrations of NMPG (50 and 100 µM) were toxic to the developing embryos during the first two days of culture, 25 µM NMPG achieved cleavage rates that were similar to those achieved by the control but did not sustain blastocyst production by Day 7 of culture. Compared to the control culture medium, the culture medium supplemented with 10 µM NMPG increased (P  less then  0.05) the rates of blastocyst formation, decreased (P  less then  0.05) the intracellular levels of reactive oxygen substances, and downregulated (P  less then  0.05) the expression of the oxidative stress related gene GPX1. In conclusion, these results demonstrated that supplementation of porcine embryo culture medium with 10 µM NMPG can attenuate oxidative stress and increase the yield of in vitro production of blastocysts.Antipsychotic drugs (AP) are used to treat a multitude of psychiatric conditions including schizophrenia and bipolar disorder. However, APs also have metabolic side effects including increased food intake and body weight, but the underlying mechanisms remain unknown. We previously reported that minocycline (MINO) co-treatment abrogates olanzapine (OLZ)-induced hyperphagia and weight gain in mice. Using this model, we investigated the changes in the pharmacometabolome in the plasma and hypothalamus associated with OLZ-induced hyperphagia and weight gain. Female C57BL/6 mice were divided into groups and fed either i) control, CON (45% fat diet) ii) CON + MINO, iii) OLZ (45% fat diet with OLZ), iv) OLZ + MINO. We identified one hypothalamic metabolite indoxylsulfuric acid and 389 plasma metabolites (including 19 known metabolites) that were specifically associated with AP-induced hyperphagia and weight gain in mice. We found that plasma citrulline, tricosenoic acid, docosadienoic acid and palmitoleic acid were increased while serine, asparagine and arachidonic acid and its derivatives were decreased in response to OLZ. These changes were specifically blocked by co-treatment with MINO. click here These pharmacometabolomic profiles associated with AP-induced hyperphagia and weight gain provide candidate biomarkers and mechanistic insights related to the metabolic side effects of these widely used drugs.Disulfide bonds link pairs of cysteine amino acids and their formation is assumed to be complete in the mature, functional protein. Here, we test this assumption by quantifying the redox state of disulfide bonds in the blood clotting protein fibrinogen. The disulfide status of fibrinogen from healthy human donor plasma and cultured human hepatocytes are measured using differential cysteine alkylation and mass spectrometry. This analysis identifies 13 disulfide bonds that are 10-50% reduced, indicating that fibrinogen is produced in multiple disulfide-bonded or covalent states. We further show that disulfides form upon fibrin polymerization and are required for a robust fibrin matrix that withstands the mechanical forces of flowing blood and resists premature fibrinolysis. The covalent states of fibrinogen are changed by fluid shear forces ex vivo and in vivo, indicating that the different states are dynamic. These findings demonstrate that fibrinogen exists and functions as multiple covalent forms.Manipulation of an object requires us to transport our hand towards the object (reach) and close our digits around that object (grasp). In current models, reach-related information is propagated in the dorso-medial stream from posterior parietal area V6A to medial intraparietal area, dorsal premotor cortex, and primary motor cortex. Grasp-related information is processed in the dorso-ventral stream from the anterior intraparietal area to ventral premotor cortex and the hand area of primary motor cortex. However, recent studies have cast doubt on the validity of this separation in separate processing streams. We investigated in 10 male rhesus macaques the whole-brain functional connectivity of these areas using resting state fMRI at 7-T. Although we found a clear separation between dorso-medial and dorso-ventral network connectivity in support of the two-stream hypothesis, we also found evidence of shared connectivity between these networks. The dorso-ventral network was distinctly correlated with high-order somatosensory areas and feeding related areas, whereas the dorso-medial network with visual areas and trunk/hindlimb motor areas. Shared connectivity was found in the superior frontal and precentral gyrus, central sulcus, intraparietal sulcus, precuneus, and insular cortex. These results suggest that while sensorimotor processing streams are functionally separated, they can access information through shared areas.The Madden Julian Oscillation (MJO), the dominant subseasonal variability in the tropics, is widely represented using the Real-time Multivariate MJO (RMM) index. The index is limited to the satellite era (post-1974) as its calculation relies on satellite-based observations. Oliver and Thompson (J Clim 251996-2019, 2012) extended the RMM index for the twentieth century, employing a multilinear regression on the sea level pressure (SLP) from the NOAA twentieth century reanalysis. They obtained an 82.5% correspondence with the index in the satellite era. In this study, we show that the historical MJO index can be successfully reconstructed using machine learning techniques and improved upon. We obtain a significant improvement of up to 4%, using the support vector regressor (SVR) and convolutional neural network (CNN) methods on the same set of predictors used by Oliver and Thompson. Based on the improved RMM indices, we explore the long-term changes in the intensity, phase occurrences, and frequency of the winter MJO events during 1905-2015.
