This cross-sectional study investigated menopause status in relation to hand grip strength, standing balance, and rapid foot tapping. A secondary aim was to examine the relationship between physical performance and urban/rural residence with a focus on habitual daily tasks.

Maya and non-Maya women (40-60 years) were drawn from urban and rural sites in Campeche, Mexico (n= 543). Demographic, reproductive, and lifestyle information was collected in face-to-face interviews along with anthropometric and physical function measures. Linear regression was used to evaluate menopause status in relation to strength, balance, and foot tapping speed while adjusting for residence, ethnicity, and other variables.

Hand grip strength was 22.5, 21.6, and 20.0 kg in pre-, peri-, and postmenopausal women, respectively, but menopause status was not significantly related to grip strength in models adjusted for age. Grip strength was negatively associated with age and socioeconomic index, and positively associated with height and weight, self-reported health, and hours/week spent grinding corn/making tortillas. Postural stability was 9.4, 6.9, and 5.6 s across menopause categories; and menopause status remained significant in adjusted models. The number of foot taps in 10 s was 35.7, 33.4, and 33.9 taps in pre-, peri-, and postmenopausal women. Parity was negatively associated with foot tapping in adjusted models.

While age is a key predictor of physical function in women aged 40-60 years, menopausal status appears to have additional influences on postural control beyond age alone. Hours spent grinding corn/making tortillas were significantly associated with grip strength among rural women.

Psoriasis is a chronic inflammatory disease of the skin with complex pathogenesis. Long non-coding RNAs (lncRNAs) play an important regulatory role in the occurrence and progression of many diseases, as well as psoriasis.

This study aimed to investigate the role and mechanism of the lncRNA, SPRR2C, in M5-induced psoriatic keratinocytes.

SPRR2C expression and subcellular localization was detected using FISH and qRT-PCR. Ker-CT and HaCaT cells stimulated by M5 (IL-17A, tumour necrosis factor-α, IL-1α, IL-22, and oncostatin-M) were used to establish a psoriatic cell model. CCK-8 assay, CFSE proliferation assay, flow cytometry, western blotting and ELISA were used to examine the effects of SPRR2C in the keratinocyte model.

SPRR2C was highly expressed in psoriatic samples and M5-induced psoriatic keratinocytes, and SPRR2C was mainly localised to the cytoplasm. In keratinocytes, SPRR2C regulated proliferation, cell cycle and apoptosis, and induced the expression of IL-1β, IL-6, IL-8, CXCL2 and CCL20. Moreover, SPRR2C cellular effects were shown to be mediated by the PI3K/AKT/mTOR signalling pathway, based on experiments with the AKT-specific inhibitor, MK-2206, which was also shown to suppress overexpression of SPRR2C.

Our results indicate that SPRR2C plays a regulatory role and is involved in the PI3K/AKT/mTOR signalling pathway in psoriatic keratinocytes, which may provide a potential diagnostic and therapeutic target for psoriasis.

The global, regional, and national burden of psoriasis was investigated based on the Global Burden of Disease (GBD) study.

To report the incidence of psoriasis in 204 countries and territories from 21 regions according to age, sex, region, country, and socialdemographic index (SDI) between 1990 and 2019.

Estimates from the GBD 2019 study were used to analyse the incidence of psoriasis at the global, regional, and national levels. The estimated annual percentage changes (EAPCs) in the age-standardized incidence rate (ASIR) were calculated to quantify trends between 1990 and 2019.

From 1990 to 2019, the global incidence of psoriasis increased by 26.53%, but the ASIR of psoriasis decreased, with an EAPC of -0.77 (95% confidence interval [CI] -0.78 to -0.76). In 2019, the highest ASIRs of psoriasis (112.58 per 100,000 population; 95% uncertainty interval, 108.89 to 116.07) were observed in high-SDI regions. The male-to-female ratio for psoriasis incidence peaked globally in the 50-54-year-old age group and peaked in the 75-79-year-old age group in high-SDI regions. Regionally, Central Sub-Saharan Africa (EAPC, -0.57; 95% CI -0.67 to -0.48) and Eastern Sub-Saharan Africa (EAPC, -0.36; 95% CI, -0.38 to -0.34) had the largest decrease in ASIR of psoriasis from 1990 to 2019. Nationally, increases in the ASIR of psoriasis was observed only in Japan (EAPC, 0.04; 95% CI 0.02 to 0.05).

Globally, the incidence of psoriasis showed an increasing trend, but the ASIR of psoriasis decreased significantly from 1990 to 2019. Only Japan showed an unfavourable increasing trend.

The Dermatology Quality of Life Index (DLQI) is the most commonly used quality of life questionnaire in dermatology.

The aim of this study was to compare the DLQI to measures of wellbeing and general quality of life in patients with chronic inflammatory skin diseases.

A cross-sectional study among patients with psoriasis and atopic dermatitis was conducted at a hospital in Munich, Germany (12/2017-04/2019). Participants filled in validated scales measuring happiness, quality of life, and DLQI.

In 102 patients with chronic inflammatory skin diseases, the DLQI was associated only with physical quality of life and negative emotions, and not psychological quality of life or positive affect. The DLQI alone accounted for 26% of variance in general quality of life. Combining DLQI and happiness accounted for a total of 73% of variance, with both variables contributing to the model.

The DLQI alone only partially reflected well-being. Assessing happiness in addition to the widely used DLQI can contribute to a more comprehensive evaluation of well-being.

Narrowband-UVB (NB-UVB) is a common and effective psoriasis treatment. It exerts its effect locally and is therefore a better model for exploring dynamics of serum biomarkers reflecting psoriasis skin disease activity compared to other treatments with systemic uptake.

To perform an exploratory study to assess potential roles of multiple disease mediators as biomarkers for psoriasis disease activity, and increase understanding of NB-UVB treatment effects in psoriatic skin.

Patients with plaque psoriasis were sampled (lesional, non-lesional skin, serum) before and after full NB-UVB treatment. read more Samples were assessed for 78 different mediators using Luminex assays. Correlation networks were analysed to explore interactions between lesional skin mediators before and after NB-UVB treatment.

None of the studied serum mediators were significantly affected by NB-UVB treatment after correction for multiple testing. Thirty mediators revealed a significant difference in lesional skin compared to non-lesional skin was observed in serum, thus challenging the possibility of a serum biomarker reflecting skin disease activity.
Treatment with baricitinib in combination with topical corticosteroids previously showed greater improvements in itch and sleep versus placebo in adults with moderate-to-severe AD.

To assess whether improvements in itch and sleep translate to greater quality of life (QoL), productivity and treatment benefit in AD.

In this post hoc analysis with data from BREEZE-AD7 (NCT03733301), itch and sleep improvements at Week 16 were defined by ≥4-point improvements in the Itch Numeric Rating Scale and ≥1.5 decreases in the number of night-time awakenings since baseline, respectively. Dermatology Life Quality Index, Work Productivity and Activity Impairment-AD and Patient Benefit Index (PBI) scores were compared in patients with and without improvements. Proportions were analysed using logistic regression with non-responder imputation. Changes from baseline were calculated using ANCOVA, with last observation carried forward. Least square mean PBI scores were assessed using ANOVA.

More patients with itch improvemments in well-being.
Sebaceous carcinoma and sweat gland carcinoma (malignant tumours with apocrine and eccrine differentiation) are rare malignant adnexal tumours that differentiate toward sebaceous glands and eccrine and apocrine glands, respectively. Because of the rarity of these malignancies, standard treatments for advanced disease have yet to be established. The outcomes of patients with systemic metastasis remain poor, highlighting the need for novel treatment strategies. Nectin cell adhesion molecule 4 (NECTIN4) and its antibody-drug conjugate, enfortumab vedotin, have attracted attention as potential treatments for solid tumours.

To examine the potential use of NECTIN4-target therapy for sebaceous and sweat gland carcinoma.

We immunohistochemically investigated NECTIN4 expression in 14 sebaceous carcinoma samples and 18 sweat gland carcinoma samples, and examined whether NECTIN4-targeted therapy could be applied to these cancers.

We found strong and frequent expression of NECTIN4 in both cancers. All tumours exhibited positive staining at least in a part of the lesion, and the mean H-score, a semiquantitative score ranging from 0 to 300, was 259.4 for sebaceous carcinoma and 253.1 for sweat gland carcinoma.

Our results suggest that both sebaceous carcinoma and sweat gland carcinoma could be potentially treated with NECTIN4-targeted antibody-drug conjugates, such as enfortumab vedotin.

Psoriasis is a systemic inflammatory disease. The autoimmune response plays a role in the pathogenesis of psoriasis. The long non-coding RNA (lncRNA) antisense non-coding RNA in the INK4 locus (ANRIL) can regulate the immune response and participate in the pathogenesis of immune-related diseases, however, the role of ANRIL in the pathogenesis of psoriasis requires further clarification.

To study the association between ANRIL polymorphisms and psoriasis in the northern Chinese population.

We genotyped six SNPs in ANRIL in 270 psoriasis vulgaris patients and 271 healthy controls in the northern Chinese population using an improved multiplexed ligation detection reaction method, in order to identify the role of ANRIL in psoriasis.

The C allele of rs3217992 and the T allele of rs2518723 were more prevalent in the case group than in the control group. The haplotypes, CTATAA, CCCCGG, and CTCCGG, were associated with risk of psoriasis, while the TCCCGG, TCATAA and CCATAA haplotypes were protective against psoriasis.
