The members of the monoacylglycerol acyltransferase (MOGAT) family are essential candidate genes that influence economic traits associated with triglyceride synthesis, dietary fat absorption, and storage in livestock. In addition, the MOGAT gene family may also play an essential function in human polygenic diseases, like type 2 diabetes and obesity. The present study was conducted on Holstein calves to find the association between MOGAT1, MOGAT3/g.A229G, and MOGAT3/g.G1627A and growth traits. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was performed for genotyping the MOGAT1, MOGAT3/g.A229G, and MOGAT3/g.G1627A genes' locus using the TaqI, MspI, and BsuRI restriction enzyme. The allele frequency of A and G of the MOGAT1 locus was 0.79 and 0.21, respectively, while the genotype frequency was 0.65, 0.28, and 0.07 for AA, AG, and GG, respectively. While the allele and genotype frequencies of the MOGAT3/g.A229G locus were 00.57( A 1 ), 0.43( G 1 ), 0.35( A 1 A 1 ), 0.45( A 1 G 1 ), and 0.20( G 1 G 1 ), the allele and genotype frequencies of the MOGAT3/g.G1627A locus were 0.49( A 2 ), 0.51( G 2 ), 0.25( A 2 A 2 ), 0.49( A 2 G 2 ), and 0.26( G 2 G 2 ). Chi-square analysis showed that MOGAT3/g.G1627A distribution was at the Hardy-Weinberg disequilibrium ( p 0.05). In total, two statistical methods (general linear model (GLM) and PROC MIXED) were used to identify an association between gene locus and growth traits. An association analysis showed a statistically significant difference between the MOGAT1 and body weight, body length, and chest circumference, MOGAT3/g.A229G with average daily gain (ADG) and withers height, and MOGAT3/g.G1627A with body weight and body length ( p less then 0.05). The results confirmed that the MOGAT1, MOGAT3/g.A229G, and MOGAT3/g.G1627A locus are strong candidate genes that could be considered molecular markers for growth traits in cattle breeding.To provide a cost-efficient parentage testing kit for red deer (Cervus elaphus), a 63 SNP set has been developed from a high-density Illumina BovineHD BeadChip containing 777 962 SNPs after filtering of genotypes of 50 stags. The successful genotyping rate was 38.6 % on the chip. The ratio of polymorphic loci among effectively genotyped loci was 6.5 %. The selected 63 SNPs have been applied to 960 animals to perform parentage control. Thirty SNPs out of the 63 had worked on the OpenArray platform. Their combined value of the probability of identity and exclusion probability was 4.9 × 10 - 11 and 0.99803, respectively. A search for loci linked with antler quality was also performed on the genotypes of the above-mentioned stags. Association studies revealed 14 SNPs associated with antler quality, where low-quality antlers with short and thin main beam antlers had values from 1 to 2, while high-quality antlers with long and strong main beams had values between 4 and 5. The chance for a stag to be correctly identified as having high-value antlers is expected to be over 88 %.To investigate the effects of miR-499-5p on muscle metabolism in broiler chickens, eight broiler chicks were assigned to the control group and eight to the treatment group, and then we monitored the effects using metabolomics. The results showed that miR-499-5p injection altered the concentrations of a variety of metabolites in the gastrocnemius muscle. Thereby, a total of 46 metabolites were identified at higher ( P less then 0.05 ) concentrations and 30 metabolites were identified at lower ( P less then 0.05 ) concentrations in the treatment group compared with the control group. Further metabolic pathway analysis revealed that fructose and mannose metabolism, galactose metabolism, inositol phosphate metabolism, and terpenoid backbone biosynthesis were the most critical pathway which may partially interpret the effects of miR-499-5p. To our knowledge, this research is the first report of metabolic signatures and related metabolic pathways in the skeletal muscle for miR-499-5p injection and provides new insight into the effect of miRNA on growth performance.The objective of this study was to determine the effect of the weaning method on lamb stress, body weight, sexual behavior, and semen quality of Saint Croix male lambs. The present study was carried out during the late spring and summer of 2018 in the northeast of Mexico. Sixty male lambs born as twins or triplets (3.2 ± 0.6 kg birth weight) and weaned at 60 d of age (19.21 ± 1.8 kg weaning weight) were divided into two weaning methods complete separation from the dams (CS; the lambs were moved to a pen that was at 500 m of distance from the dams) and separation with contact from the dams (SCD); the lambs were physically separated by a steel mesh that prevented the lambs from having the possibility of sucking milk from their mothers, but they maintained permanent visual and auditory contact. Cortisol levels were determined 3 d before and 7 d after weaning. Lambs were evaluated as 3-month-old lambs for sexual behavior and semen quality for 9 weeks. The effects of the weaning method (M), week (W), and the interaction M × W were significant on body weight and cortisol levels ( P less then 0.001 ). The SCD lambs had higher cortisol levels at 3, 5, and 7 d after weaning than CS lambs ( P less then 0.001 ). The CS lambs had higher body weight during the first 4 weeks after weaning than SCD lambs ( P less then 0.001 ). The weaning method had no effect on scrotal circumference, sexual behavior, and semen quality traits, except for progressive sperm motility, being better for the lambs that were completely separated ( P less then 0.05 ). The results from this study show that complete separation of lambs and ewes at weaning is an effective method to reduce lamb stress and improve lamb growth after weaning, but it did not have long-term effects on sexual behavior and semen quality of Saint Croix male lambs.Quantitative fluorescence spectroscopy requires corresponding reflectance measurements to correct for tissue absorption and scattering. However, it is unclear whether fluorescence adds value beyond the diffuse reflectance measurements necessary for correction. The goal of this study was to compare the accuracy of fluorescence and diffuse reflectance spectroscopy in recovering the concentration of a high-extinction fluorophore, methylene blue (MB), using a compact fiber-optic probe. Fluorescence and diffuse reflectance measurements of tissue simulating phantoms were made using a fiber-optic probe with source-detector separations of 288-1300 μm. Average error in recovered fluorophore concentration was 20.4% for fluorescence and 15.0% for reflectance, though this difference was not significant (p=0.77). Both methods returned concentrations that were similar to known MB concentrations (p≥0.79 in both cases). Fluorescence quantification of the concentration of a high extinction fluorophore did not significantly improve accuracy relative to diffuse reflectance. Investigators should consider whether fluorescence measurements are necessary for a given application.In the tumor microenvironment (TME), functional interactions among tumor, immune, and stromal cells and the extracellular matrix play key roles in tumor progression, invasion, immune modulation, and response to treatment. Intratumor heterogeneity is ubiquitous not only at the genetic and transcriptomic levels but also in the composition and characteristics of TME. However, quantitative inference on spatial heterogeneity in the TME is still limited. Here, we propose a framework to use network graph-based spatial statistical models on spatially annotated molecular data to gain insights into modularity and spatial heterogeneity in the TME. Applying the framework to spatial transcriptomics data from pancreatic ductal adenocarcinoma samples, we observed significant global and local spatially correlated patterns in the abundance score of tumor cells; in contrast, immune cell types showed dispersed patterns in the TME. Hypoxia, EMT, and inflammation signatures contributed to intra-tumor spatial variations. Spatial patterns in cell type abundance and pathway signatures in the TME potentially impact tumor growth dynamics and cancer hallmarks. Tumor biopsies are integral to the diagnosis and clinical management of cancer patients; our data suggest that owing to intra-tumor non-genetic spatial heterogeneity, individual biopsies may underappreciate the extent of clinically relevant, functional variations across geographic regions within tumors.The Government of Barbados welcomed cruise ships during the early COVID-19 period of 2020, allowing them to seek safe harbour at a time when many countries were turning them away. A total of 28 cruise ships were given unprecedented permission to anchor along the west and south coasts of the island during this period (1 March – 1 September 2020). This study examines the 132 anchoring events of these cruise ships, using automatic identification system (AIS) data to determine anchored locations and track vessel swing at anchor in relation to sensitive coral-rich habitat. These data, together with SCUBA surveys on several anchoring sites were used to describe the nature of the habitat damage and to assess the potential area of impacts on coastal marine habitats. The huge anchors and hundreds of metres of chain required to secure these mega-vessels, together with their typical wide swinging motion dragging the anchor chain over the bottom are estimated to have caused thousands of square metres of structural damage to the island’s valuable coral reefs. This study revealed weaknesses in coral conservation policy and practise to which the Government of Barbados has responded. A stated new Government policy prohibits cruise ships from anchoring on the coral-rich west coast and restricts anchoring to the designated anchorage on the south coast. Additional cruise ships that cannot be accommodated in the anchorage are now being asked to use their dynamic positioning system or to drift offshore.In this study, we use the sample data from Jan 22, 2020 to Jan 21, 2022 to investigate the impacts of added infection number on the volatility of BDI. Under this structure, the control variables (freight rate, Brent crude oil price, container idle rate, port congestion level, global port calls) are added to test whether the information contained in the added infection number is covered. In the GARCH-MIDAS model, we divide the volatility of BDI into the long-term and short-term components, then employ in the least squares regression to empirically test the influences of added infection number on the volatility. From the analysis, we find the added infection numbers effectively impact the BDI volatility. In addition, whether the freight rate, Brent crude oil price, container idle rate, port congestion level, global port calls and other variables are considered alone or at the same time, further the added infection number still significantly influences the volatility of BDI. By studying the ability of the confirmed number to explain the volatility of BDI, a new insight is provided for the trend prediction of BDI that the shipping industry can take the epidemic development of various countries as a reference to achieve the purpose of cost or risk control.
