Enzimler protein yapılı moleküller olduğundan proteinleri etkileyen sıcaklık değişimlerine duyarlıdırlar. Her enzimin en fazla aktivite gösterdiği bir sıcaklık derecesi vardır. Buna optimum sıcaklık denir. Bu değer pek çok canlıda 30 – 40 °C arasındadır (İnsanda 37°C). Sıcaklık derecesi optimum değerin altına düştüğünde veya üstüne çıktığında enzimatik reaksiyon hızı azalır. Yüksek sıcaklıkta (genellikle 55 – 60°C de) enzimlerin yapısı bozulur. Düşük sıcaklıkta enzimlerin etkinliği azalır. Bu durumda sıcaklığın belirlibir dereceye kadar artırılması, enzimin etkinliğini de artırır. Yani düşük sıcaklık enzimlerin yapısını bozmaz.
