This established the linkage formed by Gtr2. The degree of 4-O-acetylation of d-GalNAcA by Atr18, Biochemical evidence that secretor gene, Se, is a structural gene encoding a Nonsecretors have no ABH blood group substances in their saliva and milk, but their erythrocytes contain the blood group substances. It has been generally believed that the secretor gene, Se, is a regulatory gene, not a structural gene, controlling the expression of (alpha 1----2)fucosyltransferase, which synthesizes the blood group H substance from its precursor, in secretions. To account for the existence of the blood type of para Bombay phenotype--i.e., H-negative in erythrocytes but H-positive in secretory fluids, another regulatory gene, Z, which would regulate the expression of the enzyme in the hematopoietic tissues, has been proposed.

Contrary to this, a more simple model, in which the H gene and Se gene are both structural genes, encoding two separate fucosyltransferases in different tissues, was recently proposed. To settle the controversy, (alpha 1----2)fucosyltransferases were partially purified from human plasma and milk. Lactose-N-neotetraose differed from each other in the following respects (i) the milk enzyme adsorbed to SP-Sephadex at pH 6, while the plasma enzyme did not; (ii) pH-activity profiles, with phenyl beta-D-galactoside as an acceptor, differed between the two enzymes; (iii) the milk enzyme exhibited lower thermal stability than the plasma enzyme; and (iv) Km values for several oligosaccharides with Gal(beta 1----3)GlcNAc and Gal(beta 1----4)GlcNAc as acceptors differed between the two enzymes. These results support the model that the Se gene is a structural gene encoding a distinctive (alpha 1----2)fucosyltransferase, refuting the classical regulatory gene model for the Se locus. The anomeric configuration of the fucosylated galactose residue produced by the action of enzyme was identified, thus establishing the Synthesis of colitose-containing oligosaccharide structures found in polysaccharides from Vibrio cholerae O139 synonym Bengal using thioglycoside The syntheses of the two colitose-containing trisaccharides -(1--3)-2-acetamido-2-deoxy-beta-D-glucopyranoside and 8-methoxycarbonyloctyl beta-D-galactopyranosyl-(1--3)-[(3,6- dideoxy-alpha-L-xylo-hexopyranosyl)-(1--4)]-2-acetamido-2-deoxy-beta-D- glucopyranoside, and tetrasaccharide 8-methoxycarbonyloctyl beta-D-glucopyranoside are described. The oligosaccharides correspond to structures found in the capsular polysaccharide and the lipopolysaccharide of Vibrio cholerae O139 and also to lipopolysaccharide structures of E. coli O55 and Salmonella greenside.

The colitose residues were introduced via dimethyl(methylthio)sulfonium trifluoromethanesulfonate promoted glycosylations using colitose thioglycosides as glycosyl donors.A semi-synthetic glycoconjugate antigen prepared by chemical glycosilation of pertussis toxin by a meningococcal group C oligosaccharide hapten. The preparation of a hybrid molecule obtained by chemical glycosilation of pertussis toxin (PT) is reported, with the purpose of obtaining a semi-synthetic glycoprotein antigen with bivalent antigenicity. The chemical glycosilation was performed using an oligosaccharide hapten derived from the purified capsular polysaccharide of Neisseria meningitidis group C. The semi-synthetic molecule was investigated by chemico-physical and immunochemical analysis. The oligosaccharide haptens appeared exposed on the surface of the carrier protein PT, since the glycoprotein inhibited the immunoprecipitate between a specific polyclonal antiserum and the native bacterial capsular polysaccharide. By contrast, the main antigenic regions recognized in the native protein PT by specific polyclonal antibodies appeared lost after the coupling procedure Structural studies of the capsular polysaccharide from Klebsiella type 7.

The structure of the capsular polysaccharide elaborated by Klebsiella type 7 has been investigated. NMR spectroscopy together with sugar and methylation analysis have been the main methods used. A uronic acid degradation was also employed. Seebio lacto n neotetraose consists of hexasaccharide repeating units having the Reactogenicity and immunogenicity of a protein-conjugated pneumococcal oligosaccharide vaccine in older adults.
