We previously showed that extracellular ATP released under hypoxic conditions is an autocrine/paracrine, the angiogenic factor for pulmonary artery (PA) VV endothelial cells (VVECs), acting via P2Y purinergic receptors (P2YR) and the Phosphoinositide 3-kinase (PI3K)-Akt-Mammalian Target of Rapamycin (mTOR) signaling. To further elucidate the molecular mechanisms of ATP-mediated VV angiogenesis, we determined the profile of ATP-inducible transcription factors (TFs) in VVECs using a TranSignal protein/DNA array. C-Jun, c-Myc, and Foxo3 were found to be upregulated in most VVEC populations and formed nodes connecting several signaling networks. siRNA-mediated knockdown (KD) of these TFs revealed their critical role in ATP-induced VVEC angiogenic responses and the regulation of downstream targets involved in tissue remodeling, cell cycle control, expression of endothelial markers, cell adhesion, and junction proteins. Our results showed that c-Jun was required for the expression of ATP-stimulated angiogenic genes, c-Myc was repressive to anti-angiogenic genes, and Foxo3a predominantly controlled the expression of anti-apoptotic and junctional proteins. The findings from our study suggest that pharmacological targeting of the components of P2YR-PI3K-Akt-mTOR axis and specific TFs reduced ATP-mediated VVEC angiogenic response and may have a potential translational significance in attenuating pathological vascular remodeling.Rattlesnakes have venoms with a complex toxin mixture comprised of polypeptides and proteins. Previous studies have shown that some of these polypeptides are of high value for the development of new medical treatments. The aim of the present study is to evaluate, in vitro, the antibacterial and hemolytic activity of Crotalus triseriatus and Crotalus ravus venoms. A direct field search was conducted to obtain Crotalus triseriatus and Crotalus ravus venom samples. These were evaluated to determine their antibacterial activity against Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa through the techniques of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Hemolytic activity was also determined. Antibacterial activity was determined for treatments (Crotalus triseriatus 2) CT2 and (Crotalus ravus 3) CR3, obtaining a Minimum Inhibitory Concentration of 50 µg/mL and a Minimum Bactericidal Concentration of 100 µg/mL against Pseudomonas aeruginosa. CT1 (Crotalus triseriatus 1), CT2, and CR3 presented hemolytic activity; on the other hand, Crotalus ravus 4 (CR4) did not show hemolytic activity. The results of the present study indicate for the first time that Crotalus triseriatus and Crotalus ravus venoms contain some bioactive compounds with bactericidal activity against Pseudomonas aeruginosa which could be used as alternative treatment in diseases caused by this pathogenic bacterium.The aim of this work was the development of a thermoplastic/thermosetting combined system with a novel production technique. A poly(caprolactone) (PCL) structure has been designed and produced by fused filament fabrication, and impregnated with an epoxy matrix. The mechanical properties, fracture toughness, and thermal healing capacities of this blend (EP-PCL(3D)) were compared with those of a conventional melt mixed poly(caprolactone)/epoxy blend (EP-PCL). The fine dispersion of the PCL domains within the epoxy in the EP-PCL samples was responsible of a noticeable toughening effect, while in the EP-PCL(3D) structure the two phases showed an independent behavior, and fracture propagation in the epoxy was followed by the progressive yielding of the PCL domains. This peculiar behavior of EP-PCL(3D) system allowed the PCL phase to express its full potential as energy absorber under impact conditions. Optical microscope images on the fracture surfaces of the EP-PCL(3D) samples revealed that during fracture toughness tests the crack mainly propagated within the epoxy phase, while PCL contributed to energy absorption through plastic deformation. Due to the selected PCL concentration in the blends (35 vol %) and to the discrepancy between the mechanical properties of the constituents, the healing efficiency values of the two systems were rather limited.A simple color spot test for determining the presence of residual tetrabutylammonium (TBA) in various 18F-radiopharmaceuticals is described. The broad use of two vaccines, which are based on RVA strains from Europe and North America, significantly reduced rotavirus disease burden worldwide. However, a lower vaccine effectiveness is recorded in some regions of the world, such as sub-Saharan Africa, where diverse RVA strains are circulating. Here, a plasmid-based reverse genetics system was used to generate simian RVA reassortants with VP4 and VP7 proteins derived from African human RVA strains not previously adapted to cell culture. We were able to rescue 1/3 VP4 mono-reassortants, 3/3 VP7 mono-reassortants, but no VP4/VP7 double reassortant. Electron microscopy showed typical triple-layered virus particles for the rescued reassortants. All reassortants stably replicated in MA-104 cells; however, the VP4 reassortant showed significantly slower growth compared to the simian RVA or the VP7 reassortants. The results indicate that, at least in cell culture, human VP7 has a high reassortment potential, while reassortment of human VP4 from unadapted human RVA strains with simian RVA seems to be limited. The characterized reassortants may be useful for future studies investigating replication and reassortment requirements of rotaviruses as well as for the development of next generation rotavirus vaccines.In the tire industry, the combination of carbon black and silica is commonly utilized to improve the comprehensive performance of natural rubber so as to realize the best performance and cost-effectiveness. The corresponding mixing is divided into three processes (initial mixing, delivery, reactive mixing) by the serial modular continuous mixing method, thus achieving more accurate control of the mixing process, higher production efficiency and better performance. Moreover, the optimization of serial modular continuous mixing process parameters can not only improve the performance of composite materials, but help people understand the physical and chemical changes and the reinforcing mechanism of fillers in the mixing process. In this paper, the relationship among the parameters of eight processes and filler network structure, tensile strength, chemical reinforcing effect and tear resistance was explored through experiments. The deep causes of performance changes caused by parameters were analyzed. Consequently, the best process condition and the ranking of the influencing factors for a certain performance was obtained. Furthermore, the best preparation process of natural rubber (NR)/carbon black/silica composite was achieved through comprehensive analysis.The search for materials able to remineralize human hard tissues is a modern medical challenge. In this study, the protective effect on the enamel microhardness by a paste based on hydroxyapatite and sodium fluoride (Remin Pro) was evaluated after two different enamel bleaching procedures. Forty sound human incisors were randomly assigned to different treatments bleaching with an in-office agent (Perfect Bleach Office+); bleaching with an at-home agent (Perfect Bleach); bleaching with the in-office agent followed by the prophylaxis paste; bleaching with the athome agent followed by the prophylaxis paste; no treatment (control). Bleaching was performed at 0, 8, 24 and 32 h, followed by a 3min remineralizing treatment in the subgroups designed to receive it. Specimens underwent a microhardness tester and a mean Vickers Hardness number was considered for each specimen. ANOVA exhibited significant differences among groups. Posthoc Tukey testing showed significant microhardness decrease after the application of both the two bleaching agents. The treatment with prophylaxis paste significantly increased the microhardness values of bleached enamel.Platycosides, the saponins abundant in Platycodi radix (the root of Platycodon grandiflorum), have diverse pharmacological activities and have been used as food supplements. Since deglycosylated saponins exhibit higher biological activity than glycosylated saponins, efforts are on to enzymatically convert glycosylated platycosides to deglycosylated platycosides; however, the lack of diversity and specificities of these enzymes has limited the kinds of platycosides that can be deglycosylated. In the present study, we examined the enzymatic conversion of platycosides and showed that Cytolase PCL5 completely converted platycoside E and polygalacin D3 into deapiose-xylosylated platycodin D and deapiose-xylosylated polygalacin D, respectively, which were identified by LC-MS analysis. The platycoside substrates were hydrolyzed through the following novel hydrolytic pathways platycoside E → platycodin D3 → platycodin D → deapiosylated platycodin D → deapiose-xylosylated platycodin D; and polygalacin D3 → polygalacin D → deapiosylated polygalacin D → deapiose-xylosylated polygalacin D. Our results show that cytolast PCL5 may have a potential role in the development of biologically active platycosides that may be used for their diverse pharmacological activities.To characterise the dynamic of events during the early phases of fracture repair in humans, we investigated molecular events using gene expression profiling of bone fragments from the fracture site at different time points after trauma and immune/stromal cells recruitment at the fracture site using flow cytometry. Bone and inflammatory markers were expressed at low levels at homeostasis, while transcripts for bone constituent proteins were consistently detected at higher levels. Early after fracture (range 2-4 days), increased expression of CXCL12, suggested recruitment of immune cells associated with a change in the balance of degradation enzymes and their inhibitors. At intermediate time after fracture (4-8 days), we observed high expression of inflammatory cytokines (IL1-beta, IL6), CCL2, the T-cell activation marker CD69. Late after fracture (8-14 days), high expression of factors co-operating towards the regulation of bone turnover was detected. We identified potential soluble factors and explored circulating levels in patients for whom a union/non-union (U/NU) outcome was known.
