This is the typical skull structure of crested chickens. The associated regulatory genes are located on autosomes and are incompletely dominant. This trait is related to brain herniation, but the genetic mechanisms of its formation and development are unclear. In this study, RNA sequencing (RNA-Seq) analysis was conducted on 6 skull tissue samples from 3 Wumeng crested chickens with prominent skull protrusions and 3 without a prominent skull protrusion phenotype. A total of 46,376,934 to 43,729,046 clean reads were obtained, the percentage of uniquely mapped reads compared with the reference genome was between 89.73%-91.00%, and 39,795,458-41,836,502 unique reads were obtained. Among different genomic regions, the highest frequency of sequencing reads occurred in exon regions (85.44-88.28%). Additionally, a total of 423 new transcripts and 26,999 alternative splicings (AS) events were discovered in this sequencing analysis. This study identified 1,089 differentially expressed genes (DEGs), among which 485 were upregulated and 604 were downregulated. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses indicated that the DEGs were enriched in terms related to signal transduction, cell development, cell differentiation, the lysosome, serine, and threonine metabolism, and the interaction of cytokines with cytokine receptors. A total of 1,360 Cobb-500 slow-feathering (SF) pullets were randomly placed in 16-floor pens with 85 females per pen. Diets with corn, soybean meal, and wheat-midds were formulated without protein restriction maintaining minimum ratios between essential AA and Lys on a digestible (dig) ideal basis. Treatments consisted of 4 dietary AA levels with 80% (low-AA), 90% (moderate-AA), 100% (standard-AA), and 110% (high-AA) of the Cobb-Vantress recommendations guided by dig Lys using balanced protein. Up to 4 wk, all pullets were fed a common starter crumble diet. Grower and developer mash diets were fed to pullets from 5 to 15 wk and from 16 to 24 wk, respectively. Pullets fed standard-AA and high-AA diets were heavier (P less then 0.001) than those fed low-AA diets at 10, 15, and 20 wk of age. High-AA diets resulted in better (P = 0.040) flock uniformity at 10 wk. Pullets fed a high-AA diet had the highest (P = 0.041) relative breast weight at 20 wk of age and the lowest (P = 0.044) deposits of abdominal fat at 15 wk of age. Fleshing increased (P less then 0.05) as AA content rise in the diet, while the relative shank length (P less then 0.001) and the number of wing juvenile feathers (P = 0.004) decreased. Pullets fed the lowest dietary AA level had the longest (P = 0.007) small intestine relative to BW at 10 wk of age, but a smaller (P = 0.001) liver than those fed moderate and standard-AA diets at 20 wk of age. Cooking is considered the last defense of pathogen control for meals containing chicken livers. However, consumers' preference for pink color and a creamy texture as desired attributes in preparing liver pâté may lead to inadequate cooking, thereby increasing the risk of foodborne illness. This study aimed to investigate the effects of different cooking conditions (60-90°C, 0-65 min) on quality changes in frozen and fresh chicken livers and develop cooking recommendations to produce safe liver products with desired qualities. Frozen storage reduced the water holding capacity of raw chicken livers and led to more cooking loss (reduction in the weight of liver pieces during cooking) and area shrinkage after heating. The cooking loss and area shrinkage increased with increasing heating time and temperature, following the first-order fractional model. Compared with fresh livers, the shear resistance for cutting through the cooked livers increased after heating at 73.9°C to 90°C and decreased at 60°C, whereas the livers heated at 70°C had shear resistance (~4.5 N/g) similar to the fresh liver, regardless of the heating times used in this study. Heating resulted in color changes in livers, shifting from red hue (0°) toward yellow hue (90°), as characterized by the increased hue angles after heating. At 21 d of age, 150 male broilers (Ross 308) were randomly assigned to 3 treatment groups control (CON) group, in which broilers were provided standard finisher feed under thermoneutral condition (22 ± 1°C); HS group and HS + BT group, in which broilers were given the standard feed supplied with 0 and 1,000 mg/kg BT, respectively, under cyclic HS condition (33 ± 1°C for 8 h from 0800 to 1600 h and the thermoneutral temperature for the residual hours). Each treatment was replicated ten times with 5 broilers per replicate. The HS group showed an elevation (P less then 0.05) in serum corticosterone (CORT) concentration, D-lactate acid (D-LA) content, and diamine oxidase (DAO) activity, mucosal interleukin-1β (IL-1β) level, and expression of heat shock protein 70 (HSP70) gene, and a reduction (P less then 0.05) in mucosal interleukin-10 (IL-10) level and secretory immunoglobulin A (SIgA) content and relative abundance of mRNA for occludin (OCLN), zonula occludens-1 (ZO-1), claudin-1 (CLDN1), and claudin-4 (CLDN4). In contrast, broilers in the HS + BT group exhibited a raise (P less then 0.05) in mucosal IL-10 level and SIgA content and relative expression of OCLN and ZO-1 genes, and a decline (P less then 0.05) in serum CORT concentration and DAO activity, mucosal IL-1β level, and expression of HSP70 mRNA. The presence of multiple antibodies made the detection of a malignancy more likely and both patients were subsequently diagnosed with small cell lung carcinoma (SCLC). We highlight that the presence of multiple CNS autoantibodies increases the likelihood of detecting a malignancy but that the clinical presentation and response to treatment can vary despite similar antibody profiles. Clinicians should be alert to the need to search for occult malignancy in patients with multiple CNS autoantibodies.
