Full dynamics of biofluid biomarkers have been unknown in patients with Parkinson's disease (PD). Using data from 396 PD patients and 182 controls in the Parkinson's Progression Markers Initiative (PPMI) database, we estimated long-term temporal trajectories of CSF α-synuclein (α-syn), amyloid-β (Aβ), total tau (t-tau), phosphorylated tau (p-tau) and serum neurofilament light chain (NfL) by integrating function between the baseline levels and annual changes. At baseline, PD patients showed lower CSF α-syn, Aβ, t-tau and p-tau levels than those of the controls. In all PD patients, CSF α-syn and Aβ decreased in a negative exponential pattern before the onset of motor symptoms, whereas CSF t-tau and p-tau, and serum NfL increased. Patients with cognitive impairment exhibited faster decline of Aβ and α-syn and faster rise of t-tau, p-tau and NfL, when compared to those without. Similarly, low Aβ group showed earlier decline of α-syn, faster rise of t-tau, p-tau and NfL, and faster decline of cognitive performances, when compared to high Aβ group. Our results suggest that longitudinal changes in biomarkers can be influenced by cognitive impairment and Aβ burden at baseline. Bromodomains are epigenetic "readers" of histone acetylation. Recently, bromodomain inhibitors have exhibited promising therapeutic potential for cancer treatment. Using transcriptome sequencing, we identified BRPF1 (bromodomain and PHD finger containing 1) as the most significantly upregulated gene among the 43 bromodomain-containing genes in human HCC. BRPF1 upregulation was significantly associated with poor patient survival. Gene ablation or pharmacological inactivation of BRPF1 significantly attenuated HCC cell growth in vitro and in vivo. BRPF1 was involved in cell cycle progression, senescence and cancer stemness. Transcriptome sequencing revealed that BRPF1 is a master regulator controlling the expression of multiple key oncogenes, including E2F2 and EZH2. We demonstrated that BRPF1 activated E2F2 and EZH2 expression by facilitating promoter H3K14 acetylation through MOZ/MORF complex. In conclusion, BRPF1 is frequently upregulated in human HCCs. Targeting BRPF1 may be an approach for HCC treatment.Optical coherence tomography angiography (OCTA) provides several data regarding the status of retinal capillaries. This information can be further enlarged by employing quantitative metrics, such as vessel density (VD). A mandatory preliminary step of OCTA quantification is image binarization, a procedure used to highlight retinal capillaries on empty background. Although several binarization thresholds exist, no consensus is reached about the thresholding technique to be used. In this study, we tested thirteen binarization thresholds on a dataset made by thirty volunteers. The aim was to assess the impact of binarization techniques on (I) detection of retinal capillaries, assessed by the calculation of overlapping percentages between binarized and original images; (II) quantitative OCTA metrics, including VD, vessel tortuosity (VT) and vessel dispersion (Vdisp); (III) foveal avascular zone (FAZ) detection. Our findings showed Huang, Li, Mean and Percentile as highly reliable binarization thresholds (p  less then  0.05), whereas the worst binarization thresholds were Intermodes, MaxEntropy, RenylEntropy and Yen (p  less then  0.05). All the thresholds variably underestimated VD metric and FAZ detection, with respect to the original OCTA images, whereas VT and Vdisp turned out to be more stable. The usage of a Fixed threshold resulted extremely useful to reduce VD and FAZ underestimations, although bound to operators' experience.Recently, it was proposed that a processing principle called adaptive stochastic resonance plays a major role in the auditory system, and serves to maintain optimal sensitivity even to highly variable sound pressure levels. As a side effect, in case of reduced auditory input, such as permanent hearing loss or frequency specific deprivation, this mechanism may eventually lead to the perception of phantom sounds like tinnitus or the Zwicker tone illusion. Using computational modeling, the biological plausibility of this processing principle was already demonstrated. Here, we provide experimental results that further support the stochastic resonance model of auditory perception. In particular, Mongolian gerbils were exposed to moderate intensity, non-damaging long-term notched noise, which mimics hearing loss for frequencies within the notch. Remarkably, the animals developed significantly increased sensitivity, i.e. improved hearing thresholds, for the frequency centered within the notch, but not for frequencies outside the notch. In addition, most animals treated with the new paradigm showed identical behavioral signs of phantom sound perception (tinnitus) as animals with acoustic trauma induced tinnitus. In contrast, animals treated with broadband noise as a control condition did not show any significant threshold change, nor behavioral signs of phantom sound perception.Force enhancement is one kind of myogenic spontaneous fasciculation in lengthening preload striated muscles. In cardiac muscle, the role of this biomechanical event is not well established. The physiological passive property is an essential part for maintaining normal diastole in the heart. In excessive preload heart, force enhancement relative erratic passive properties may cause muscle decompensating, implicate in the development of diastolic dysfunction. In this study, the force enhancement occurrence in mouse cardiac papillary muscle was evaluated by a microstepping stretch method. The intracellular Ca2+ redistribution during occurrence of force enhancement was monitored in real-time by a Flou-3 (2 mM) indicator. The force enhancement amplitude, the enhancement of the prolongation time, and the tension-time integral were analyzed by myography. The results indicated that the force enhancement occurred immediately after active stretching and was rapidly enhanced during sustained static stretch. The presence of the force and the increase in the amplitude synchronized with the acquisition and immediate transfer of Ca2+ to adjacent fibres. In highly preloaded fibres, the enhancement exceeded the maximum passive tension (from 4.49 ± 0.43 N/mm2 to 6.20 ± 0.51 N/mm2). The occurrence of force enhancement were unstable in each static stretch. The increased enhancement amplitude combined with the reduced prolongation time to induce a reduction in the tension-time integral. We concluded that intracellular Ca2+-synchronized force enhancement is one kind of interruption event in excessive preload cardiac muscle. During the cardiac muscle in its passive relaxation period, the occurrence of this interruption affected the rhythmic stability of the cardiac relaxation cycle.While the precise diagnosis of early stage epiretinal membrane (ERM) at the time of cataract surgery and evaluation of risk factors for development or progression of ERM after cataract surgery is increasingly important, there is only limited information. In the present study, we evaluated the risk factors for onset or progression of ERM on spectral domain optical coherence tomography (SD-OCT) after cataract surgery. The univariate analysis showed that eyes with partial posterior vitreous detachment (PVD; p  less then  0.001), hyper-reflective foci (HF) on the inner retinal surface (p  less then  0.001), vitreoschisis (p = 0.014), and discrete margin of different retinal reflectivity (DMDRR; p = 0.007) on ultra-widefield fundus photography (UWF-FP) had significant risk for the onset or progression of ERM after cataract surgery. The multivariate analysis showed that partial PVD (HR, 3.743; 95% confidence interval [CI], 1.956-7.162; p  less then  0.001), HF (HR, 2.330; 95% CI, 1.281-4.239; p = 0.006), and DMDRR on UWF-FP (HR, 3.392; 95% CI, 1.522-7.558; p = 0.003) were the independent risk factors for the onset or progression of ERM after cataract surgery after adjustment for other confounding factors. Our study shows that the onset or progression of ERM after cataract surgery depends on an abnormal vitreoretinal interface (VRI) represented by partial PVD, HF on SD-OCT, and DMDRR on UWF-FP, not on age, axial length, or presence of ERM at the time of surgery. A meticulous funduscopic evaluation of the VRI would help to predict the ERM risk before cataract surgery.Tuberculosis has the most considerable death rate among diseases caused by a single micro-organism type. The disease is a significant issue for most third-world countries due to poor diagnosis and treatment potentials. Early diagnosis of tuberculosis is the most effective way of managing the disease in patients to reduce the mortality rate of the infection. Despite several methods that exist in diagnosing tuberculosis, the limitations ranging from the cost in carrying out the test to the time taken to obtain the results have hindered early diagnosis of the disease. This work aims to develop a predictive model that would help in the diagnosis of TB using an extended weighted voting ensemble method. The method used to carry out this research involved analyzing tuberculosis gene expression data obtained from GEO (Transcript Expression Omnibus) database and developing a classification model to aid tuberculosis diagnosis. A classifier combination of Naïve Bayes (NB), and Support Vector Machine (SVM) was used to develop the classification model. The weighted voting ensemble technique was used to improve the classification model's performance by combining the classification results of the single classifier and selecting the group with the highest vote based on the weights given to the single classifiers. Experimental analysis indicates a performance accuracy of the enhanced ensemble classifier as 0.95, which showed a better performance than the single classifiers, which had 0.92, and 0.87 obtained from SVM and NB, respectively. The developed model can also assist health practitioners in the timely diagnosis of tuberculosis, which would reduce the mortality rate caused by the disease, especially in developing countries.
