The genus Ceratocystis contains several significant plant pathogens, causing wilt and canker disease on a wide range of plants species. Currently, there are over 40 known species of Ceratocystis, some of which are becoming increasingly important in agricultural or natural ecosystems. The diagnostics for most Ceratocystis species currently relies on time consuming and labour-intensive culturing approaches. To provide more time efficient and sensitive molecular diagnostic tools for Ceratocystis, a generic Taq-Man real-time PCR assay was developed using the ITS gene. This novel two-probe Taq-man assay amplified DNA from all tested Ceratocystis species. Some non-specific amplification of a few species from closely related genera was observed under certain conditions; however, these false positive detections could be ruled out using the additional PCR primers developed for further sequence based identification of the detected species. The assay was highly sensitive as it detected 0.2 pg/µl of Ceratocystis DNA in water as well as in host DNA matrix. Further validation with artificially inoculated fig stem tissue demonstrated that the assay was also able to effectively detect the pathogen in infected asymptomatic stem tissue. This newly developed real-time PCR assay has practical applications in biosecurity, conservation, and agriculture, enabling to detect Ceratocystis species directly from plant material, to facilitate more sensitive screening of imported plant germplasm, and allow rapid tracking of pathogens in case of disease outbreaks.Peanut (Arachis hypogaea L.) is cultivated in tropical and subtropical regions of the world as an important source of oil and protein. Until now, bacterial wilt, caused by Ralstonia solanacearum, was the only known bacterial disease of peanut. In 2020, widespread incidence of poor stand establishment were observed in multiple production fields planted to the Spanish-type peanut varieties in the Texas Panhandle. The observed symptoms included seed rot, pre- and post-emergence damping-off, poor seedling vigor and death, and poorly developed root systems with little or no nodule formation. Subsequent diagnosis of symptomatic seedlings recovered two bacterial species identified by BLAST using 676 bp and 661 bp 16S rRNA fragments as a R. species and a Pantoea sp., respectively. To investigate a possible causative role of these bacteria in the observed peanut disease, the pathogenicity of the two isolates was evaluated under greenhouse conditions relying on Koch's postulates. Cell suspensions of the two bacteria, scus sequence analysis (MLSA) involving six genes (dnaK, fumC, gyrB, murG, trpB, and tuf) showed that these new strains are most closely related to R. pickettii and P. dispersa, but also phylogenetically distinct. The two bacteria were designated Ralstonia sp. strain B265 and Pantoea sp. strain B270. Losses from the disease in affected fields in 2020 averaged fifty percent ($1.12 million) from a total of nine production fields. Findings from this study provide evidence for two new bacterial pathogens of peanuts capable of infecting Spanish and Valencia peanut varieties.Powdery mildew (PM), caused by two fungal species, Podosphaera xanthii and Erysiphe vignae, is a yield limiting foliar disease commonly found in mungbean (Vigna radiata) cropping areas of eastern region of Australia. Effective control of the disease relies largely on fungicide applications, mainly of the triazole group. Uncertainty in the current fungicide spray schedule recommendations, which advise commencing with a spray at the first signs of PM, prompted this study to evaluate PM severity and crop yield data obtained from fungicide trials which also tested spray schedules starting before (early) or after (late) first signs, applied singly or combined with a follow-up spray. A meta-analytic approach was employed to obtain mean differences of the PM severity and crop yield between plots sprayed with specific triazole-based spray schedules and nontreated plots. From 26 trials, 14 and 15 met the criteria for inclusion in the respective PM severity and yield analyses. The schedule with the first spray starting at first sign, with a follow-up spray 14 days later, resulted in significantly lower disease severity compared to all other schedules. However, the yield protected was only numerically higher and not statistically different compared to single-spray at first sign, single-spray late or two-spray starting late. PM severity and yield in the early sprayed plots did not differ from the nontreated plots. These findings support the current recommendations and provide additional evidence that yields are still protected when delaying the first spray up to a week after disease onset. Fanconi anemia (FA) and ataxia-telangiectasia (AT) are rare inherited syndromes characterized by abnormal DNA damage response and caused by pathogenic variants in key DNA repair proteins that are also relevant in the pathogenesis of breast cancer and other cancer types. The risk of cancer in children with these diseases is poorly understood and has never been assessed in a population-based cohort before.

We identified 421 patients with FA and 160 patients with AT diagnosed between 1973 and 2020 through German DNA repair disorder reference laboratories. We linked patients' laboratory data with childhood cancer data from the German Childhood Cancer Registry.

Among 421 patients with FA, we observed 33 cases of childhood cancer (15 cases of myelodysplastic syndrome; seven cases of acute myeloid leukemia; two cases of lymphoma, carcinoma, medulloblastoma, and nephroblastoma, respectively; and one case of rhabdomyosarcoma, acute lymphoblastic leukemia, and glioma, respectively) versus 0.74 expected (on the basis of population-based incidence rates in Germany). This corresponds to a 39-fold increased risk (standardized incidence ratio [SIR] = 39; 95% CI, 26 to 56). For all FA subgroups combined, the cancer-specific SIR for myeloid neoplasms was 445 (95% CI, 272 to 687). Among the 160 patients with AT, we observed 19 cases of childhood cancer (15 cases of lymphoma, three cases of leukemia, and one case of medulloblastoma) versus 0.32 expected. This corresponds to a 56-fold increased risk (SIR = 56; 95% CI, 33 to 88). The cancer-specific SIR for Hodgkin lymphoma was 215 (95% CI, 58 to 549) and for non-Hodgkin lymphoma 470 (95% CI, 225 to 865).

The activity of androgen-deprivation therapy (ADT) in androgen receptor-positive (AR+) salivary gland carcinomas (SGCs) has been established in the past few years. Second-line treatment in castration-resistant patients is still unknown. We investigated the activity of abiraterone acetate as second-line treatment in ADT-resistant, AR+ patients with SGC.

This was a single-institution phase II trial. A two-stage Simon's design was applied. The primary end point was confirmed objective response rate. Secondary end points were disease control rate, safety, progression-free survival, and overall survival. Patients were eligible when the following criteria were met histologic diagnosis of AR-overexpressing SGC, measurable disease according to RECIST 1.1, clinical and/or radiologic progression on ADT, suppressed serum testosterone, and no limits for the number of previous chemotherapy lines. Because of the reconstructed band structure, we find that this device acts as a robust valley filter for electrons in the graphene sheet. We show that, by appropriate alignment, the Weyl semimetal draws away current in one of the two graphene valleys, while allowing current in the other to pass unimpeded. We use recurrent neural networks to describe the large deviations of the dynamical activity of model glasses, kinetically constrained models in two dimensions. We present the first finite size-scaling analysis of the large-deviation functions of the two-dimensional Fredrickson-Andersen model, and explore the spatial structure of the high-activity sector of the South-or-East model. These results provide a new route to the study of dynamical large-deviation functions, and highlight the broad applicability of the neural-network state ansatz across domains in physics.
