Here, two newly emerged PPR virus (PPRV) isolates from India and from the Middle East were tested in an animal trial to analyse their pathogenesis, and to evaluate serological and molecular detection methods. Animals infected with the two different PPRV isolates showed marked differences in clinical manifestation and scoring. The PPRV isolate from India was less virulent than the virus from the Middle East. Commercially available rapid detection methods for PPRV antigen (two Lateral Flow Devices (LFDs) and one antigen ELISA) were evaluated in comparison with a nucleic acid detection method. For this purpose, ocular and nasal swabs were used. Due to the easy non-invasive sampling, faecal samples were also analysed. For all rapid antigen detection methods, a high specificity of 100% was observed independent of the sample matrix and dilution buffers used. Both antigen ELISA and LFD tests showed highest sensitivities for nasal swabs. Here, the detection rate of the antigen ELISA, the LFD-PESTE-TEST and the LFD-ID Rapid-Test was 78%, 75% and 78%, respectively. Ocular swabs were less suitable for antigen detection of PPRV. These results reflect the increased viral load in nasal swabs of PPRV infected goats compared to ocular swabs. The faecal samples were the least suitable for antigen detection. In conclusion, nasal swab samples are the first choice for the antigen and genome detection of PPRV. Nevertheless, based on the excellent diagnostic specificity of the rapid tests, positive results generated with other sample matrices are solid. In contrast, negative test results can be caused on the reduced analytical sensitivity of the rapid antigen tests and must be treated with caution.Alpine ski racing is known as a high-risk sport; however, little is known on the health problems occurring at youth level. The purpose of this study was to investigate the prevalence of health problems in youth competitive alpine skiers with respect to sex, age, and season differences, to describe their severity and location and to assess the influence of biological maturation. Over a 12-month period, 155 youth competitive alpine skiers around the growth spurt were prospectively monitored for health problems using the Oslo Sports Trauma Research Centre (OSTRC) questionnaire. These data were verified by supplemental interviews at the end. Biological maturation was estimated by a non-invasive, anthropometric-based assessment method. During competition season, 42.2% of the skiers suffered from health problems and 19.8% reported their problem being substantial. Females had significantly higher rates of acute and overuse injuries compared to males, while there was no sex difference in duration, severity, and time loss. Skiers of the category U15 showed higher prevalence of acute and overuse injuries than U14 skiers. During preparation season, health problem prevalence was substantially lower. The knee was found to be the most affected body part for both acute and overuse injuries. There was a direct association between the offset to the age at peak height velocity (APHV) and the occurrence and severity of acute injuries. In conclusion, youth competitive alpine skiers suffer from a large number of health problems. Since sex, age, and biological maturation are important, effective injury prevention should already start before reaching the APHV.Background Gastric neoplasms exhibiting oxyntic gland differentiation (GAOG) is a gastric neoplasm subtype that shows low-atypia, thus being similar to non-neoplastic gastric oxyntic glands. Therefore, its diagnosis in biopsy specimens is difficult. GAOG was first described in 2007 and introduced in the latest WHO classification book as gastric adenocarcinoma fundic gland type (GA-FG) and oxyntic gland adenoma. Previously we assessed α1,4-linked N-acetylglucosamine residues (αGlcNAc) attached to the MUC6 scaffold in human gastric neoplasms and observed decreased αGlcNAc glycosylation in both differentiated type gastric cancer and in high-grade pyloric gland adenoma (PGA), a gastric cancer precursor. Recently it was reported that GA-FG and PGA often exhibit same mutations. However, αGlcNAc status in GAOG remained unknown. Methods and results We assessed expression of αGlcNAc, the mucin markers MUC6, MUC5AC and MUC2, the gastric gland cell markers MIST1, pepsinogen 1 (PG1), H/K-ATPase and chromogranin-A (CGA), and the proliferation marker Ki-67 in 13 GAOG lesions. All 13 (100%) were MUC6 positive, while 10 (76.2%) were αGlcNAc negative. Moreover, all 13 (100%) were MIST1 positive, 11 (84.6%) were PG1 positive, 3 (23.1%) were MUC5AC positive, 4 (30.8%) were H/K-ATPase positive, and 1 (7.7%) was CGA positive. Conclusions GAOGs frequently lost αGlcNAc on MUC6 but expressed the gastric gland progenitor marker MIST1 and aberrantly express various types of gastric gland cell lineage markers, suggestive of immature differentiation to gastric gland cells. Thus, diffuse MIST1 positivity and decreased αGlcNAc glycosylation on MUC6 positive cells could serve as important biomarkers for histopathological diagnosis of GAOG.Cassava (Manihot esculenta Crantz) is one of the important staple foods in Sub-Saharan Africa. It produces starchy storage roots that provide food and income for several hundred million people, mainly in tropical agriculture zones. Increasing cassava storage root and starch yield is one of the major breeding targets with respect to securing the future food supply for the growing population of Sub-Saharan Africa. The Cassava Source-Sink (CASS) project aims to increase cassava storage root and starch yield by strategically integrating approaches from different disciplines. We present our perspective and progress on cassava as an applied research organism and provide insight into the CASS strategy, which can serve as a blueprint for the improvement of other root and tuber crops. Extensive profiling of different field-grown cassava genotypes generates information for leaf, phloem, and root metabolic and physiological processes that are relevant for biotechnological improvements. A multi-national pipeline for genetic engineering of cassava plants covers all steps from gene discovery, cloning, transformation, molecular and biochemical characterization, confined field trials, and phenotyping of the seasonal dynamics of shoot traits under field conditions. Together, the CASS project generates comprehensive data to facilitate conventional breeding strategies for high-yielding cassava genotypes. It also builds the foundation for genome-scale metabolic modelling aiming to predict targets and bottlenecks in metabolic pathways. The relatively large difference in transmetalation selectivity between the neighboring Tm3+ and Yb3+ species is noteworthy because the reactivities of heavy lanthanides are generally considerably similar. The structural strain of the polymeric framework is likely responsible for this unusual trend. Powder X-ray diffraction analysis indicated that, in the cases of only Yb3+ and Lu3+ , large differences in their ionic sizes compared to that of Ce3+ in the parent framework may induce a structural strain after solid solution formation, while cleavage of the relatively weak Ce-O bond allows the formation of new Yb-O and Lu-O bonds with the incoming Yb3+ and Lu3+ , respectively.
