Semen samples collected from buffalo bulls were assessed for sperm functional attributes (n = 11) and proteome profiling (n = 6). Sperm proteins were extracted and profiled by employing LC-MS/MS. Overall, the buffalo sperm contained 1365 proteins, of which 458 were common between the groups. The unique proteins were 477 and 430 in good and poor quality semen, respectively. In the whole proteome of buffalo sperm, sexual reproduction with phosphatidylethanolamine-binding protein1 (PEBP1), fetuin-B (FETUB) and acrosin (ACR) was the most enriched (p = 8.44E-19) biological process, also with thermogenesis (p = 0.003), oocyte meiosis (p = 0.007) and vascular smooth muscle contraction (p = 0.009) apart from metabolic pathways. In good quality semen, mesenchyme migration (p = 1.24E-07) and morphogenesis (p = 0.001) were abundant biological processes. In good quality semen, the fluid shear stress (p = 0.01) and, in poor quality semen, valine, leucine and isoleucine degradation (p = 3.8E-05) pathways were enriched. In good quality semen, 7 proteins were significantly (p less then 0.05) upregulated and 33 proteins were significantly (p less then 0.05) downregulated. On validating the abundantly expressed sperm proteins, serine protease inhibitor Kazal-type 2-like (SPINK2; 2.17-fold) and neddylin (NEDD8; 1.13-fold) were upregulated and YBX2 was downregulated (0.41-fold) in good quality semen as compared with poor quality semen (1-fold). The present findings revealed the importance of sperm proteins in oocyte maturation, fertilization process and early embryonic development. The variations in the proteomic composition can be used as potential markers for the selection of breeding bulls.Fibroblast growth factor receptor 4 (FGFR4) has been indicated as a potential "oncogene" in various types of cancer. However, the effects and underlying mechanisms of FGFR4 on uterine leiomyosarcoma (ULMS) progression remain unclear. In this study, we firstly discovered that FGFR4 was upregulated in ULMS specimens and cell lines and closely associated with poor prognosis of ULMS patients. Cell viability and apoptosis assays showed that FGFR4 deletion inhibited cell proliferation and promoted cell apoptosis. Moreover, FGFR4 silence increased cytoplasmic GABP (GA binding protein) expression, while it decreased the nuclear GABP level to inhibit nuclear localization of GABP. Mechanistically, the inhibition ability of FGFR4 silence on nuclear localization of GABP was mediated via mammalian Ste20-like kinases 1 (MST1) activation, which could promote phosphorylation of large tumor suppressor 1 (LATS1) to reduce nuclear localization of GABP. Gain- and loss-of-functional assays indicated that FGFR4 promoted nuclear localization of GABP to inhibit cell apoptosis in ULMS. In conclusion, our findings indicated that FGFR4 inhibited cell apoptosis in ULMS via the promotion of MST1/LATS1-mediated GABP nuclear localization, shedding light on the underlying mechanism of FGFR4-induced ULMS progression.One class of molecules that are now coming to be recognized as essential for our understanding of the nervous system are the lysophospholipids. Optical coherence tomography showed a great ability to identify adverse features during percutaneous coronary intervention with drug-eluting stents and resulted in better clinical outcomes. The study aimed to assess the impact of optical coherence tomography on intraoperative decision-making during implantation of Absorb bioresorbable scaffolds versus everolimus drug-eluting stents.

We performed an observational study that included 223 consecutive patients post optical coherence tomography-guided implantation of either Absorb bioresorbable scaffolds (162 patients) or everolimus drug-eluting stents (61 patients). We studied the influence of optical coherence tomography on intraoperative decision-making during implantation of bioresorbable scaffolds versus drug-eluting stents by analyzing the total rate of optical coherence tomography-dependent modifications in each device. After satisfactory angiographic results, the total rate of required intervention for optical coherence tomography detected complications on the reasons of scaffolds failure in current PCI practice; it offers a new insight for the enhancement of BRS safety and presents and adds to the growing literature for successful BRS utilization.
In this study, we aimed to maximize glutathione (GSH) production by a metabolically engineered Yarrowia lipolytica strain using a small-scale optimization approach.

A three levels four factorial Box-Behnken Design was used to assess the effect of pH, inulin extract, yeast extract and ammonium sulfate concentrations on cell growth and to generate a mathematical model which predict optimal conditions to maximize biomass production and thus GSH titer. The obtained results revealed that only yeast and inulin extract concentrations significantly affect biomass production. Based on the generated model, a medium composed of 10g/L of yeast extract and 10g/L of inulin extract from Jerusalem artichoke was used to conduct batch cultures in 2 L bioreactor. After 48h of culture, the biomass and the glutathione titer increased by 55% (5.8 g
/L) and 61% (1011.4mg/L), respectively, as compared to non-optimized conditions.

The present study examined the effects that different ratings made during initial exposure have on the illusory truth effect with news headlines. In two experiments, participants (total N = 575) rated a set of news headlines in one of two conditions. Some participants rated how interesting they were, and others rated how truthful they were. Participants later rated the perceived accuracy of a larger set of headlines that included previously rated and new headlines. In both experiments, prior exposure increased perceived accuracy for participants who made initial interest ratings, but not for participants who made initial truthfulness ratings. The increase in perceived accuracy that accompanies repeated exposure was attenuated when participants considered the accuracy of the headlines at initial exposure. Experiment 2 also found evidence for a political bias participants rated politically concordant headlines as more accurate than politically discordant headlines. The magnitude of this bias was related to performance on a cognitive reflection test; more analytic participants demonstrated greater political bias. The association of BCL2 polymorphism with breast cancer was investigated in the Isfahan province of Iran. PCRamplification of the CA-repeat was followed by polyacrylamide gel electrophoresis and direct sequencing for 120 breast cancer women and an equal number of corresponding healthy control individuals. Seven different alleles, ranging from 11 to 17 CA-repeats were observed. Short alleles with 11 to 14 repeats were protective (OR 0.363, P = 0.001), but large alleles with 15 to 17 repeats were threatening against breast cancer development (OR 2.780, P = 0.001). Accordingly, genotypes with large alleles showed a higher risk of breast cancer development (OR 3.400, P = 0.004). ERS1ERBB2 positive breast cancer patients, but not PGRpositive ones, showed protection against breast cancer (OR 0.405, OR 0.346 respectively). In conclusion, women with at least one large allele of BCL2 were 3.4 times at higher risk of breast cancer development in the Isfahan province of Iran.Birds have been historically affected by metals and are considered powerful sentinels of environmental pollution. Some toxic elements (Pb, Cd, Hg, As) have been broadly studied in avian species and are well known for their accumulation capacity and deleterious effects, including alterations in vitamin levels. Vitamins A, E and D3 are fat-soluble nutrients involved in multiple physiological functions (e.g., immune function, vision, reproduction, growth and development). Publications reporting metal-induced effects on vitamins in birds are growing and, in some cases, results seem contradictory, making them difficult to interpret. Therefore, a clear view of the overall picture is needed. This mini-review article aims to compile relevant data and describe current knowledge on the effects of the most toxic elements (i.e., Pb, Cd, Hg, As) on vitamins A, E and D3 in birds. Although vitamins are diet dependent, they are strongly regulated and transformed in the organism, and metal-related disruption in their homeostasis may provoke alterations in different directions. Moreover, vitamin status and form in vivo is the result of complex interacting processes in the organism and metal exposure may produce cascade effects. Different factors that may contribute to the variable response of vitamins to metals in birds are discussed. Some final remarks and recommendations are provided for future studies. This mini-review shows an overview of the current knowledge in metal-induced alterations in vitamins of special concern for avian ecotoxicology, a research discipline facing important challenges in the coming years.
