The binding affinity of EVO and γ-secretase complex was analyzed by molecular docking. Cellular thermal shift assay (CETSA) was performed to study the drug-target interactions in NSCLC cells. Protein levels were determined by Western blotting. RESULTS EVO dramatically inhibited cell viability, induced G2/M cell cycle arrest, suppressed cell migration, and reduced stemness in NSCLC cells. Mechanistic studies indicated that EVO prevented the γ-secretase cleavage of Notch3 at the cell surface and hence inhibited Notch3 activation. Moreover, EVO notably reduced tumor growth in the mouse model and inhibited Notch3 activity in the tumors. CONCLUSION This study provides new insights into the anti-NSCLC action of EVO, and suggests that suppressing Notch3 signaling by inhibiting γ-secretase is a mechanism of action underlying the anti-NSCLC effect of EVO. BACKGROUND The dried heartwood of Caesalpinia sappan L. is traditionally prescribed in the formula of traditional Chinese medicine (TCM) for the treatment of acute myeloid leukemia (AML), while nothing is yet known of the active fractions and the underlying mechanisms. PURPOSE This study aims to investigate the effect of the ethyl acetate extract of the dried heartwood of Caesalpinia sappan L. (C-A-E) on induction of apoptosis and promotion of differentiation in vitro and anti-AML activity in vivo. STUDY DESIGN/METHODS The aqueous extract was sequentially separated with solvents of increasing polarity and the active fraction was determined through the inhibition potency. The inhibition of the active fraction on cell viability, proliferation and colony formation was performed in different AML cells. Induction of apoptosis and the promotion of differentiation were further determined. Then, the level of the reactive oxygen species (ROS) and its potential role were assessed. Finally, anti-AML activity was evaluatapoptosis and differentiation of HL-60 cells was significantly mitigated by NAC. Additionally, C-A-E also exhibited an obvious anti-AML effect in NOD/SCID mice with the injection of HL-60 cells. CONCLUSIONS C-A-E exhibited an inhibitory effect on AML cells by inducing mitochondrial apoptosis and promoting differentiation, both of which were highly correlated to the activation of ROS. The objective of this study was to evaluate consumer behaviors, including barriers, surrounding e-waste recycling at a large Midwestern university in the United States. A survey was administered to faculty, graduate students, undergraduate students, and staff to determine their personal recycling habits, knowledge, and beliefs. The results indicate that free access to disposal, lack of consumer knowledge about products and disposal sites, and access to a recycling facility within a reasonable distance are all important factors in consumer decisions. Policy-makers and waste management professionals should focus on promotion of e-waste recycling behaviors through increased access to free or low-cost recycling as well as through the creation of recycling incentives. A system that can detect the intention to move and decode the planned movement could help all those subjects that can plan motion but are unable to implement it. In this paper, motor planning activity is investigated by using electroencephalographic (EEG) signals with the aim to decode motor preparation phases. A publicly available database of 61-channels EEG signals recorded from 15 healthy subjects during the execution of different movements (elbow flexion/extension, forearm pronation/supination, hand open/close) of the right upper limb was employed to generate a dataset of EEG epochs preceding resting and movement's onset. A novel system is introduced for the classification of premovement vs resting and of premovement vs premovement epochs. For every epoch, the proposed system generates a time-frequency (TF) map of every source signal in the motor cortex, through beamforming and Continuous Wavelet Transform (CWT), then all the maps are embedded in a volume and used as input to a deep CNN. The proposed system succeeded in discriminating premovement from resting with an average accuracy of 90.3% (min 74.6%, max 100%), outperforming comparable methods in the literature, and in discriminating premovement vs premovement with an average accuracy of 62.47%. The achieved results encourage to investigate motor planning at source level in the time-frequency domain through deep learning approaches. We present a case of a healthy 29 year-old female with an uneventful vaginal delivery who had transient, sudden onset of rigors and fever 36 hours postpartum. She was found to have Fusobacterium gonidiaformans bacteremia due to retained placental tissue. We report this organism as it is not well-described and rarely reported. It does bear some similarities to other Fusobacterium species that have been reported to cause septicemia in young otherwise healthy patients. Haemonchus contortus transthyretin domain-containing protein (HcTTR) with 136 amino acids belongs to a transthyretin-like (TTL) family member. In our previous study, it was reported that HcTTR was a novel antagonist of the goat cytokine Interleukin 4 (IL-4), and was involved in the regulation of host immune responses, implying that it might be applied for therapeutic strategies or vaccine development against the infection of H. contortus. Thus, the protective capacities of HcTTR against H. contortus infections via active and passive immunization trials were examined. For the passive protection trials, H. contortus-infected goats were intravenously immunized twice with 5 mg of total IgG containing anti-rHcTTR goat polyclonal antibodies. The results showed that the significant rates of reduction in egg shedding and worm burden was 58.12% and 64.61%, respectively, as compared with the positive control group. For the active protection trials, local goats were vaccinated twice with 500 μg of recombinant HcTTR to generate antigen-specific circulating antibodies, resulting in 63.7% reduction in eggs shedding and 66.4% reduction in worm burdens after H. contortus challenge. In both passive and active trials, the immunized goats displayed higher mucosal IgA levels and less anaemic compared to the challenged positive controls. Pen trials indicated that HcTTR generated partial immune protective effects against H. contortus challenge and it could be a promising vaccine candidate for development of effective strategy to control H. contortus. E-cigarette use has surged in recent years. Many of these new users are cigarette smokers. It is unclear whether e-cigarette adoption by smokers will lead to improved public health due to uncertainty about whether e-cigarettes help smokers quit using cigarettes and about whether ongoing dual use reduces exposure to toxins. A third source of uncertainty is whether providing cigarette smokers with sample e-cigarettes increases e-cigarette adoption. To provide insight into this final issue, we follow up with cigarette smokers who left an experimental auction with an e-cigarette, contacting them after two weeks, six weeks, and six months to determine which demographic and smoking-related characteristics predict continued e-cigarette use. We find that smokers who have made a serious quit attempt, have been advised to quit smoking, or have previously tried e-cigarettes are significantly more likely to report continued e-cigarette use. Women and smokers from racial and ethnic minority groups are significantly less likely to use e-cigarettes at follow up, as are those who said they would rather quit than switch to e-cigarettes. We also use experimental auction bids to estimate the price elasticity of demand for e-cigarettes, finding that a 10% increase in price results in a 5.6% reduction in quantity demanded. This suggests e-cigarette demand is less price sensitive than some earlier studies have found. While a tax on e-cigarettes can still be an effective tool for reducing e-cigarette demand, the reduction in demand may be smaller than some earlier studies would suggest. The effectiveness of tube and sonotrode reactors for the sonication of sewage sludge under identical conditions was compared for the first time. Despite the considerable structural differences, sonication with each ultrasonic reactor led to an accelerated degradation rate and an increased methane production within the first five days for the majority of the sewage sludge samples tested. On closer examination, however, it becomes clear that the investigated sonication systems are not equally suitable for the substrates considered. While the use of a sonotrode proved to be particularly advantageous for the treatment of waste activated sludge (+25% methane yield at 300 kJ/kgTS), the use of a 2-inch tube reactor achieved the highest enhancement for low-intensity sonication in digested sludge (+22% methane yield at 300 kJ/kgTS). With increasing energy input, more chemical oxygen demand was solubilized, but this did not result in an increase in methane yield for all samples. Sonication of waste activated sludge led to a significant reduction in viscosity of up to 50%, and a reduction of up to 60% was observed after sonication of digested sludge with low energy inputs. The study, therefore, demonstrates that the choice of the most suitable sonication system essentially depends on the properties of the sludge to be sonicated. We report a new soft template method for the synthesis of unique polyaniline (PANI) films with microporous structures. In this process, ultrasonication is used to foam an electrolyte solution containing a surfactant, which is subsequently employed as a soft template for PANI growth via the electrochemical polymerization of aniline. Analysis by scanning electron microscopy demonstrates that the resulting PANI films contain numerous micropores. These microporous PANI films exhibit faster charging (doping) and discharging (dedoping) current responses compared to ordinary flat films of the same material. In paper production industry, microbial contaminations of process waters are common and can cause damage to paper products and equipment as well as the occurrence of pathogens in the end products. Chlorine omission has led to the usage of costly reagents and products of lower mechanical quality. In this study, we have tested a rotation generator equipped with two sets of rotor and stator assemblies to generate developed cavitation (unsteady cloud shedding with pressure pulsations) or supercavitation (a steady cavity in chocked cavitation conditions) for the destruction of a persistent bacteria Bacillus subtilis. Our results showed that only supercavitation was effective and was further employed for the treatment of waters isolated from an enclosed water recycle system in a paper producing plant. The water quality was monitored and assessed according to the chemical (COD, redox potential and dissolved oxygen), physical (settleable solids, insolubles and colour intensity) and biological methods (yeasts, aerobic and anaerobic bacteria, bacterial spores and moulds).
