Background Constipated patients remain dissatisfied with current treatments suggesting a need for alternative therapies. Aim Evaluate the mechanistic effects of oral vibrating capsule in chronic idiopathic constipation (CIC) by examining the temporal relationships between the onset of vibrations, complete spontaneous bowel movements (CSBM), and circadian rhythm. Methods In post hoc analyses of two double-blind studies, CIC patients (Rome III) were randomized to receive 5 active or sham capsules/week for 8 weeks. The capsules were programmed for single vibration (study 1) or two vibration sessions with two modes, 8 hours apart (study 2). Daily electronic diaries assessed stool habit and percentage of CSBMs associated with vibrations. Responders were patients with ≥ 1 CSBM per week over baseline. Results 250 patients were enrolled (active = 133, sham = 117). During and within 3 hours of vibration, there were significantly more % CSBMs in the active vs. sham group (50% vs. 42%; P = .0018). In study 2, there were two CSBM peaks associated with vibration sessions. Significantly more % CSBMs occurred in active mode 1 (21.5%) vs. sham (11.5%); (P = .0357). Responder rates did not differ in study 1 (active vs. sham 26.9% vs. 35.9%, P = .19) or study 2 (mode 1 vs. sham 50% vs. 31.8%, P = .24; mode 2 vs. sham 38.1% vs. 31.8%, P = .75). Device was well-tolerated barring mild vibration sensation. Conclusions Vibrating capsule may increase CSBMs possibly by enhancing the physiologic effects of waking and meals, and augmenting circadian rhythm, although responder rate was not different from sham. However, the currently effective long wavelength operating photosensitizers have to rely on heavy atoms to enhance ISC. Unfortunately, the heavy atoms, besides their toxicity, can also cause significant reduction in the triplet lifetime of the photosensitizers, which is fundamentally detrimental to their applications. To overcome this challenge, herein we report the co-existence of efficient ISC and long triplet excited lifetime in a heavy atom-free Bodipy helicene molecule. Via advanced theoretical computation and time-resolved electron paramagnetic resonance spectroscopy, we confirmed the ISC of the Bodipy is resulted from its twisted molecular structure and reduced symmetry. The twisted Bodipy shows exceptionally intense long wavelength absorption (ε = 1.76 × 10 5 M -1 cm -1 at 630 nm), satisfactory triplet quantum yield (Φ T = 52%) and long-lived triplet state (τ T = 492 μs), leading to unprecedented performance as a triplet photosensitizer for PDT. Moreover, nanoparticles constructed with such helical Bodipy show surprisingly efficient PDT mediated antitumor immunity amplification with ultra-low dose (0.25 μg/kg body weight), which is several hundred times lower than that of the existing PDT reagents. This study evaluated the efficacy of 50 mg/kg purified anacardic acids (AAs), isolated from cashew nut shell liquid, on multiple steps of oxidative stress and inflammation induced by rotenone in the substantia nigra (SN) and striatum. Adult mice were divided into four groups Control, rotenone, AAs + rotenone, and AAs alone. Lipoperoxidation, nitric oxide (NO) levels, and reduced glutathione (GSH)/oxidized gluthatione (GSSG) ratio were evaluated. NF-kB-p65, pro-IL-1β, cleaved IL-1β, metalloproteinase-9, Tissue Inhibitory Factor-1 (TIMP-1), tyrosine hydroxylase (TH), and glial fibrillary acidic protein (GFAP) levels were assessed by Western blot. In silico studies were also made using the SwissADME web tool. Rotenone increased lipoperoxidation and NO production and reduced TH levels and GSH/GSSG ratio in both SN and striatum. It also enhanced NF-kB-p65, pro, and cleaved IL-1β, MMP-9, GFAP levels compared to control and AAs groups. The AAs alone reduced pro-IL-1β in the striatum while they augmented TIMP1 and reduced MMP-9 amounts in both regions. AAs reversed rotenone-induced effects on lipoperoxidation, NO production, and GSH/GSSG ratio, as well as increased TH and attenuated pro-IL-1β and MMP-9 levels in both regions, NF-kB-p65 in the SN and GFAP in the striatum. Altogether, the in vivo and in silico analysis reinforced multiple and defined molecular targets of AAs, identifying that they are promising neuroprotective drug candidates for PD, acting against oxidative and inflammatory conditions induced by rotenone.Patient-dentist discourse is a core nonoperational competency in dental education. The skills of querying patients and responding to questions are noncognitive attributes, and their evaluation by the standardized patient objective structured clinical examination (OSCE) is particularly necessary. However, it is not clear whether students' test anxiety affects these attributes. This study aims to examine the relationship between dental students' state-trait anxiety, noncognitive performance, and examination results during their first OSCE. A single dental school cohort (n = 226) of 5 year students attending their first clinical examination from 2014 to 2017 was studied. Participants completed the Chinese Mandarin Version State-Trait Anxiety Inventory Y form before taking the OSCE. The difference between state and trait anxiety levels was compared by paired t test. Gender differences and the effect of age group in these anxiety levels were analyzed using multivariate analysis of variance. Moreover, gender, age group, state anxiety, and trait anxiety scores were compared with the OSCE items of noncognitive performance using a chi-square test. Students showed significantly higher state anxiety than trait anxiety levels; moreover, women showed significantly higher state anxiety than men. Furthermore, gender, age group, state anxiety, and trait anxiety had no association with the noncognitive performance examination results. Most participants showed moderate state and trait anxiety levels during their first OSCE. Further, the state-trait anxiety had no significant effect on their noncognitive performance. However, 26.5% of participants did not pass the examination; therefore, dental educators should increase communication skill training courses during clerkship training to improve students' noncognitive attributes.Pathogenic germline variants in the gap junction protein alpha 1 (GJA1) gene have been identified in several congenital disorders affecting cutaneous, skeletal, and cardiac tissues. Risks in different spatial and temporal scales must be considered to ensure preconditions for sustainable multifunctional management of forests for different ecosystem services. For this purpose, the present review article summarizes the most recent findings on major abiotic and biotic risks to boreal forests in Finland under the current and changing climate, with the focus on windstorms, heavy snow loading, drought and forest fires and major insect pests and pathogens of trees. In general, the forest growth is projected to increase mainly in northern Finland. In the south, the growing conditions may become suboptimal, particularly for Norway spruce. Although the wind climate does not change remarkably, wind damage risk will increase especially in the south, because of the shortening of the soil frost period. The risk of snow damage is anticipated to increase in the north and decrease in the south. Increasing drought in summer will boost the risk of large-scale forest fires.
