The results were better than those in the control group in the total motility recovery rate (77.8 ± 11.2% vs 56.6 ± 11.9%, P  less then  0.01), progressive motility recovery rate (77.6 ± 13.2% vs 47.7 ± 14.6%, P  less then  0.01), 24 h survival index (60.9 ± 13.4% vs 42.1 ± 14.1%, P  less then  0.01) and the sperm DNA fragment index (4.2 ± 3.7% vs 5.8 ± 3.7%, P = 0.126). This method was applied to the oligozoospermic specimens. Motile spermatozoa could be found in 12 of 15 cases in the ultra-rapid freezing group, while only in 7 cases in control group. The results indicated that this freezing method was simple, convenient and bio-safe for cryopreservation of severe oligozoospermic specimens. OBJECTIVE The present study aimed to compare the force of aesthetic wires and their non-aesthetic equivalents from two commercial brands at different displacement points, during the unloading phase at 36°C. MATERIALS AND METHODS Forty samples (n=10) were assessed, which were presented in two groups of aesthetic wires and two groups of non-aesthetic wires from the brands GAC® (Dentsply GAC® International, NY, USA) and Aditek®(Cravinhos, SP, Brazil), with 0.016-inch gauge. The orthodontic wires were maintained at a temperature of 36°C throughout the experiment and they were subjected to the three-point bending test at a speed of 1mm/min. The unloading values were recorded as 0.5, 1.5, and 2.5mm of displacement. The data were subjected to Student's t-test and unequal variance t test, at 5% significance level. Published by Elsevier Masson SAS.OBJECTIVE The main aim of this randomized clinical trial was to determine 3 dimensional skeletal and dental changes six months after the use of bone-anchored versus tooth-anchored rapid maxillary expanders in adolescents. The secondary aim was to determine the symmetrical or asymmetrical expansion pattern between both appliances. MATERIALS AND METHODS Fifty adolescents with skeletally constricted maxilla (mean age 13-14 years) were randomly assigned into Dresden B-RME, Hyrax T-RME, or untreated control groups. CBCT scans were taken at initial and expander removal (6 months). Three-dimensional references and treatment landmarks were identified. Orthogonal distances were calculated from those landmarks. The main outcome was to compare skeletal and dental changes in each group and the secondary outcome was to verify if these changes were symmetric or not. Data was analysed using descriptive statistics and repeated measure MANCOVA and MANOVA. RESULTS Both treatment groups showed significant skeletal and dental exponent of dental expansion. Due to the Dresden B-RME configuration, asymmetrical expansion was noted. Investigation of the microbial community in the female reproductive tract with the use of sequencing techniques has revealed that endometrial samples obtained through a transvaginal catheter are dominated by Lactobacillus species. Dysbiotic changes in the endometrial microbiota may be associated with implantation failure or early spontaneous abortion in patients who undergo assisted reproductive technology treatment. Whether or not there is an endometrial microbiota in early pregnancy is unknown. Herein we describe, the human endometrial microbiota in a patient who subsequently had an 8th week spontaneous clinical miscarriage with euploid embryos in the next cycle and, for the first time, during a successful pregnancy in which the endometrial fluid was sampled at 4 weeks of gestation. The microbial profile found on the endometrial sample before the spontaneous abortion had higher bacterial diversity and lower Lactobacillus abundance than the endometrial fluid from the healthy pregnancy. Functional metagenomics detected different Lactobacillus species between the 2 samples. Lactobacillus crispatus was present in the endometrium before the spontaneous abortion, as were other bacteria involved in dysbiosis, which had an unstable functional pattern characterized by transposases and insertion elements. Lactobacillus iners was the most prevalent microbe found in the endometrium during early pregnancy; its presence was associated with defense mechanisms and basal functions. These novel observations prompt future investigations to understand the potential implications of microbiology on healthy and pathologic human pregnancy. OBJECTIVE Resveratrol has shown benefit for pulmonary hypertension improvement. Our previous reports showed NR4A3/cyclin D1 pathway promoted pulmonary arterial smooth muscle cells (PASMCs) proliferation. This study tried to explore the mechanism underlying this process, focusing on the role of resveratrol in regulation of miRNA and NR4A3. METHODS Rats were injected with monocrotaline (MCT) to establish pulmonary hypertension (PH) models. Resveratrol was used to prevent pulmonary vascular remodeling. Primary rat PASMCs were cultured in vitro and stimulated by platelet-derived growth factor (PDGF) with or without resveratrol. Cells proliferation and expression of miR-638 as well as NR4A3 were evaluated. RESULTS MCT resulted in significant pulmonary vascular remodeling and down-regulation of miR-638, which could be suppressed by resveratrol. Moreover, PDGF-induced PASMC proliferation and miR-638 down-regulation were both significantly prevented by resveratrol treatment in vitro. MiR-638 mimics markedly inhibited PASMC proliferation and percentage of PCNA-positive cells in vitro. But anti-miR-638 could markedly promote cells proliferation and percentage of PCNA-positive cells. The luciferase reporter assay showed that NR4A3 was a direct target of miR-638. The loss-of-function and gain-of-function experiments indicated that NR4A3 promoted proliferation via cyclin D1 pathway. CONCLUSION Our data indicated that resveratrol prevented MCT-induced pulmonary vascular remodeling via miR-638 regulating NR4A3/cyclin D1 pathway. Here, a simple continuous assay for agmatinase (AGM) activity was established by following the degradation of agmatine to urea and putrescine using isothermal titration calorimetry (ITC). ITC is particularly useful for kinetic assays when substrates of interest do not possess suitable chromophores that facilitate the continuous spectrophotometric detection of substrate depletion and/or product formation. In order to assess the accuracy of the ITC-based assay, catalytic parameters were also determined using a discontinuous, colorimetric assay. Both methods resulted in comparable kinetic parameters. From the colorimetric assay the kcat and KM values are 131 s-1 and 0.25 mM, respectively, and from the ITC assay the corresponding parameters are 30 s-1 and 0.45 mM, respectively. The continuous ITC-based assay will facilitate functional studies for an enzyme that is an emerging target for the development of addiction treatments. We report on a new sensitive method, for bovine serum albumin quantification, which is based on the use of methylene blue as a labelling agent combined with photothermal lens detection. In the presence of sodium dodecyl sulfate, methylene blue forms a dimer which can react with bovine serum albumin producing dedimerization. We found that, the photothermal signal decreases proportional to the concentration of bovine serum albumin added to the system composed by dodecyl sulfate and methylene blue. Therefore, the change of the signal intensity is linearly proportional to the concentration of bovine serum albumin. Under the optimized analytical conditions, used in this work, the photothermal signal is linearly dependent on the concentration of bovine serum albumin in the range of 0.5 × 10-6-7.5 × 10-5 gmL-1 with a regression coefficient R2 = 0.9954. The relative standard deviation for BSA determination at 3.5 × 10-5 gmL-1 (n = 5) is 2.3% and the achieved detection limit is 3.5 × 10-7 gmL-1. In this note we describe a four-channel paper microfluidic device (μPAD) that utilizes gold nanoparticles (AuNPs) as colorimetric sensors and specifically tailored aptamers to capture a set of targeted drugs, including cocaine, codeine and methamphetamine. The design utilizes salt-induced aggregation of AuNPs to produce a color change that indicates the presence of target analytes. In the absence of the targets, the aptamers bind to AuNPs, preventing aggregation. This μPAD was developed by optimizing the concentrations of aggregating agents, AuNPs and aptamers. The resultant device is sensitive and specific, producing a positive result upon detection of the target analyte. BACKGROUND AND AIMS The importance of having quality assessment, assurance, and improvement tools in healthcare is increasingly recognized. However, the additional administration burden, which is still often associated therewith, progressively interferes with the structural implementation and adoption of such tools, especially when it concerns high-volume procedures such as colonoscopies. The development of the Dutch Gastrointestinal Endoscopy Audit (DGEA), a registry with automated extraction of colonoscopy quality data, and its first results, are described. METHODS In close cooperation with commercial endoscopy reporting systems and a national histopathology database, healthcare professionals performing colonoscopies initiated a quality registry that extracts data from its core hospital resource or histology database without manual interference of the healthcare providers. Data extracted consisted of patients' age, gender, indication of the colonoscopy, ASA-score, Boston Bowel Preparation Score, Cecal intubation, and for the colonoscopy after a positive FIT in the colorectal cancer screening program also the polyp detection rate, which was available for all 48 hospitals or endoscopy centers, and adenoma detection rate, which was available for 26 hospitals or endoscopy centers. RESULTS Between 1 January 2016 and 31 March 2019, 48 hospitals or endoscopy centers voluntarily participated in the DGEA and 275,017 unique patients with 313,511 colonoscopies were registered. Overall missing values were limited to less then 1%. CONCLUSIONS The results of this study demonstrate that it is feasible to deploy a quality registry collecting uniform data without additional administration burden for the healthcare professionals. AIMS We aim i) to describe self-reported oral health among patients with diabetes in Spain; ii) to recognize which variables (socio-demographic, lifestyle and health-related), were related to poor self-reported oral health in patients with diabetes. METHODS We used data from subjects of the National Health Interview Survey performed in 2017. Diabetes status was self-reported. One non-diabetes patient was matched by gender and age for each diabetes case. Poor self-reported oral health was defined using the answers "I have dental caries" to the following question "What is the state of your teeth and molars?". Socieconomic, demographic, comorbidities, health status, health care-related were independent variables. RESULTS We found that the prevalence of dental caries was higher among subjects with diabetes than their matched controls (23.8% vs. 16.5%; P less then 0.001). The OR of dental caries for people with diabetes was 1.45 (95% CI 1.12-1.30). Older age, having lower monthly income, obesity, periodontal disease, very poor/poor/ fair self-rated health and having public dental health insurance increased the probability of suffering caries.
