Look at bactericidal outcomes of ultraviolet mild D irradiation upon cariogenic microorganisms: A good within vitro review.
Nanostructure adsorbents namely nanofibers have been demonstrated to have a high adsorption rate and are efficient to treat wastewater. Herein, chitosan/poly(vinyl alcohol) (PVA) blend nanofiber membranes prepared by electrospinning method were crosslinked using glutaraldehyde and functionalized with 1-allyl-3-methylimidazolium chloride to be used as a potential bio-sorbent for heavy metal ions removal. The chitosan was first hydrolyzed before electrospinning with PVA, followed by crosslinking and further functionalized by ionic liquid to overcome the limitation of chitosan which has low adsorption capacity and unsuitable physical properties for the adsorption process. The morphology and the chemical bond formed were investigated by using field emission scanning electron microscopy with energy dispersive x-ray spectroscopy (FESEM-EDX) and Fourier transform infrared (FTIR) showing that the hydrolyzed chitosan/PVA nanofiber membranes were successfully crosslinked and functionalized. The synthesized adsorbent was evaluated in pure heavy metal ions solutions namely Pb(II), Mn(II), and Cu(II) and shown best performance for Pb(II) ions. The highest adsorption capacity recorded for Pb(II) ions was 166.34 mg/g and are well fitted to the Freundlich isotherm model and pseudo-second-order kinetic model to describe the adsorption equilibrium and kinetic rate of the Pb(II) uptake, respectively. The synthesized adsorbent clearly shows a great capability to remove Pb(II) ions. The study aimed to develop pre-gelatinized starch-based orally disintegrating films (ODFs) containing catechin/β-cyclodextrin (CAT/β-CD) complex and to evaluate the influence of the complex on the physicochemical properties of the ODFs. SEM images showed that a compacter and more homogeneous ODFs were formed due to interactions between starch matrix and CAT/β-CD. FTIR spectra demonstrated that the interactions between starches or starch and CAT/β-CD were enhanced by hydrogen bonds. Thermal stability of ODFs was improved by incorporating CAT/β-CD, its peak decomposition temperature was enhanced from 310.74 to 321.83 °C. Tensile strength was increased from 11.597 ± 0.153 to 22.172 ± 0.752 MPa, while elongation at break decreased by from 11.233% ± 1.079% to 3.633% ± 0.058%. The prepared ODFs have an acceptable in vitro disintegration time, which were between 9.03 ± 0.79 s and 42.23 ± 1.76 s. Antimicrobial test showed that ODFs incorporating CAT/β-CD inhibited the growth of S. aureus and S. mutans successfully. The limited release of CAT molecules from the ODFs was also found. In addition, the ODFs have excellent antioxidant capacity. Its antioxidant activity remained at around 70% after 28 days of storage. EUK 134 molecular weight The study indicated that the combination of ODFs and β-CD complex have a high potential for the delivery of natural active ingredients.High-density lipoproteins (HDLs) are heterogeneous lipoproteins that modify their composition and functionality depending on physiological or pathological conditions. The main roles of HDL are cholesterol efflux, and anti-inflammatory and antioxidant functions. These functions can be compromised under pathological conditions. HDLs play a role in the immune system as anti-inflammatory molecules but when inflammation occurs, HDLs change their composition and carry pro-inflammatory cargo. Hence, many molecular intermediates that influence inflammatory microenvironments and cell signaling pathways can modulate HDLs structural modification and function. This review provides a comprehensive assessment of the importance of HDL composition and anti-inflammatory function in the onset and progression of atherosclerotic cardiovascular diseases. On the other hand, immune cell activation during progression of atheroma plaque formation can be influenced by HDLs through HDL-derived cholesterol depletion from lipid rafts and through HDL interaction with HDL receptors expressed on T and B lymphocytes. Cholesterol efflux is mediated by HDL receptors located in lipid rafts in peripheral cells, which undergo membrane structural modifications, and interferes with subsequent molecules interactions or intracellular signaling cascades. Regarding antigen-presentation cells such as macrophages or dendritic cells, HDL function may then modulate lymphocytes activation in immune response. Our review also contributes to the understanding of the effects exerted by HDLs in signal transduction associated to our immune cell population during chronic diseases progression.Although the increasing studies have corroborated the biological activities and great market utilization value of polysaccharide fractions derived from Polygonati rhizome, a well-known edible and medicinal plant, Polygonati rhizome polysaccharides (PRPs) still lack sufficient attention. Herein, we make attempt to systematically summarize recent advances in the extraction, purification, structural characteristics, biological activities, and commercial products of PRPs. Based on the detailed extraction and structural characteristics, the biological activities of PRPs including immune-regulation, anti-osteoporosis, anti-Alzheimer's disease, anti-diabetes and anti-atherosclerotic, are emphatically summarized, as well as the possible related mechanisms. Most importantly, about 365 kinds of commercial functional foods and over 500 patents related to PRPs as the main raw material were analyzed to explore the status quo and bottleneck for the development and utilization of PRPs. In conclusion, this review will benefit to bridge the gap between basic knowledge and market innovations, and facilitate the in-depth utilization of PRPs.
Lattice stereotactic body radiation therapy (SBRT) is a form of spatially fractionated radiation therapy (SFRT) using SBRT methods. This study reports clinical dosimetric endpoints achieved for Lattice SBRT plans delivering 20Gy in 5 fractions to the periphery of a tumor with a simultaneous integrated boost (SIB) of 66.7Gy, as part of a prospective Phase I clinical trial (NCT04133415). Additionally, it updates previously reported planning and delivery techniques based on extended experience with a broader patient population.

Patients were enrolled on a single-arm phase I trial conducted between November 2019 and August 2020. Eligibility was restricted to tumors >4.5cm in the largest dimension. Characteristic SFRT dose gradients were achieved using a lattice of 1.5cm diameter spheres spaced within the GTV in a regular pattern, with peak-to-valley dose varying from 66.7Gy to 20Gy within 1.5cm. Organ-at-risk (OAR) sparing followed AAPM TG101 recommendations for 5-fraction SBRT.

Twenty patients (22 plans) were enrolled on study, with one additional plan treated off study. All OAR and target coverage planning objectives were achieved, with the exception of a single small bronchus. Conformity of the 20Gy isodose line significantly improved over the course of the study. The majority (85.2%) of treatment fractions were delivered in a 30 minutes timeslot, with 4 (3.5%) exceeding a total treatment time of 40 minutes.

Lattice SBRT planning techniques produce consistent and efficient treatment plans. Refined techniques described here further improve the quality of the planning technique.

This study presents an improved protocol for the enhanced amplification and enrichment of begomovirus DNA for use in next generation sequencing of the viral genomes. An enhanced rolling circle amplification (RCA) method using EquiPhi29 polymerase was combined with size selection to generate a cost-effective, short-read sequencing method. This improved short-read sequencing produced at least 50 % of the reads mapping to the target viral reference genomes, African cassava mosaic virus and East African cassava mosaic virus. This study provided other insights into common misconceptions about RCA and lessons that could be learned from the sequencing of single-stranded DNA virus genomes. This protocol can be used to examine the viral DNA as it moves from host to vector, thus producing valuable information for viral DNA population studies, and would likely work well with other circular Rep-encoding ssDNA viruses (CRESS) DNA viruses.Dengue and chikungunya viruses are arthropod borne virus spread through common vector instigating infection in human. There has been an increased recognition that more attention needs to be paid to similar sympotoms caused by both of the virus as they spread in the same region at same time. It warrants need of cost effective, user friendly and rapid multiplex diagnostic technique which could simultaneously diagnose and identify between two virus diseases in resource poor setting. A magnetic multiplex loop mediated isothermal amplification (MM-LAMP) technique was developed by coupling multiplex LAMP with magnetic particle-based naked eye visualization to overcome the shortcoming of simultaneous detection of both diseases. In recent years this technology has emerged as a particularly attractive candidate as amplification reaction process completes within 45 min. The first step involves multiplexing biotin and digoxigenin coated dengue and chikungunya primers respectively in LAMP reaction followed by precipitation of the amplified DNA with polyethylene glycol (PEG) buffer and finally clumping with streptavidin and anti-digoxigenin coated magnetic particle for virus discrimination and naked eye visualization. The DNA detection limit of MM LAMP visualization was 51.65 ng/μl which is comparable to the electrophoresis base UV light visualization. The results showed potential superiority over standard methods polymerase chain reaction (PCR). This current advancement empowers multiplex LAMP utility in resource limited setting without using any of the florescent dyes, turbidimeter, or the sophisticated quantitative PCR machine etc which restrict multiplex LAMP technique to laboratorial use only. We have proposed a novel method without such limitations. This technique has potential as a point of care technique for simultaneous detection of two diseases.COVID-19 has posed a worldwide public health challenge affecting millions of people in different countries. Rapid and efficient detection of SARS-CoV-2 is essential for pandemic control. Reverse Transcription quantitative PCR (RT-qPCR) of nasopharyngeal swabs is the gold standard method for the virus detection, but the high demand for tests has substantially increased the costs and reduced the availability of reagents, including genetic material purification kits. Thus, the present study aimed to compare two bead-based RNA extraction methods (an in-house and a commercial kit) from nasopharyngeal swabs and RT-qPCR detection of SARS-CoV-2. Twenty-five positive and five negative nasopharyngeal swab samples were subjected to extraction of nucleic acids using both methods in an automated platform. Both protocols revealed a high correlation between Cycle Quantifications (Cqs) (r = 0.99, p less then 0.0001). In addition, the in-house kit was 89.5 % cheaper when compared to the mean cost of commercial RNA extraction kits.
