Traditionally, target-flanker interactions have been considered as local, mostly deleterious, low-level, and feature specific, occurring when information is pooled along the visual processing hierarchy. Recently, a vast literature of high-level effects in crowding (grouping effects and face-holistic crowding in particular) led to a different understanding of crowding, as a global, complex, and multilevel phenomenon that cannot be captured or explained by simple pooling models. It was recently argued that these high-level effects may still be captured by more sophisticated pooling models, such as the Texture Tiling model (TTM). Unlike simple pooling models, the high-dimensional pooling stage of the TTM preserves rich information about a crowded stimulus and, in principle, this information may be sufficient to drive high-level and global aspects of crowding. In addition, it was proposed that grouping effects in crowding may be explained by post-perceptual target cueing. Here, we extensively tested the predictions of the TTM on the results of six different studies that highlighted high-level effects in crowding. Our results show that the TTM cannot explain any of these high-level effects, and that the behavior of the model is equivalent to a simple pooling model. In addition, we show that grouping effects in crowding cannot be predicted by post-perceptual factors, such as target cueing. Taken together, these results reinforce once more the idea that complex target-flanker interactions determine crowding and that crowding occurs at multiple levels of the visual hierarchy.Color difference sensitivity as represented by the size of discrimination ellipsoids is known to depend on where the colors reside within color space. In the past, various color spaces and color difference formulas have been developed as parametric fits to the experimental data with the goal of establishing a color coordinate system in which equally discriminable colors are equal distances apart. These empirical models, however, provide no explanation as to why color discrimination varies in the way it does. This article considers the hypothesis that the variation in color discrimination tolerances reflects the uncertainty created by the degree of metamer mismatching for a given color. Specifically, the greater the degree of metamer mismatching for a color, the wider the range of spectral reflectances that could have led to it and, hence, the more finely a color needs to be discriminated in order to reliably identify materials and objects. To test this hypothesis, the available color discrimination data sets for surface colors are gathered and analyzed. A strong correlation between color discrimination and the degree of metamer mismatching is found. This correlation provides evidence that metamer mismatching provides an explanation as to why color discrimination varies throughout color space as it does.Conjoint measurement was used to investigate the joint influence of facial gender and facial age on perceived gender (Experiment 1) and perceived age (Experiment 2). A set of 25 faces was created, covarying independently five levels of gender (from feminine to masculine) and five levels of age (from young to old). Two independent groups of observers were presented with all possible pairs of faces from this set and compared which member of the pair appeared as more masculine (Experiment 1) or older (Experiment 2). Three nested models of the contribution of gender and age to judgment (i.e., independent, additive, and saturated) were fit to the data using maximum likelihood. The results showed that both gender and age contributed to the perceived gender and age of the faces according to a saturated observer model. In judgments of gender (Experiment 1), female faces were perceived as more masculine as they became older. In judgments of age (Experiment 2), young faces (age 20 and 30) were perceived as older as they became more masculine. Taken together, the results entail that (a) observers integrate facial gender and age information when judging either of the dimensions, and that (b) cues for femininity and cues for aging are negatively correlated. This correlation exerts stronger influence on female faces, and can explain the success of cosmetics in concealing signs of aging and exaggerating sexually dimorphic features.It takes less effort to walk from here to the Tiki Hut on the brick walkway than on the sandy beach. If perceived distance depends on the energetic cost or anticipated effort of walking (Proffitt, 2006), then the distance of a target viewed over sand should appear much greater than one viewed over brick. If perceived distance is specified by optical information (e.g., declination angle from the horizon; Ooi et al., 2001), then the distances should appear similar. Participants (N = 13) viewed a target at a distance of 5, 7, 9, or 11 m over sand or brick and then blind-walked an equivalent distance on the same or different terrain. First, we observed no main effect of walked terrain; walked distances on sand and brick were the same (p = 0.46), indicating that locomotion was calibrated to each substrate. Second, responses were actually greater after viewing over brick than over sand (p less then 0.001), opposite to the prediction of the energetic hypothesis. This unexpected overshooting can be explained by the slight incline of the brick walkway, which partially raises the visually perceived eye level (VPEL) and increases the target distance specified by the declination angle. The result is thus consistent with the information hypothesis. We conclude that visually perceived egocentric distance depends on optical information and not on the anticipated energetic cost of walking.Functional amyloids are abundant in living organisms from prokaryotes to eukaryotes playing diverse biological roles. In contrast to the irreversible aggregation of most known pathological amyloids, we postulate that naturally-occurring functional amyloids are reversible under evolutionary pressure to be able to modulate the fibrillization process, reuse the composite peptides, or perform their biological functions. β-Endorphin, an endogenous opioid peptide hormone, forms such kinds of reversible amyloid fibrils in secretory granules for efficient storage and returns to the functional state of monomers upon release into the blood. The environmental change between low pH in secretory granules and neutral pH in extracellular spaces is believed to drive the reversible fibrillization of β-endorphin. Here, we investigate the critical role of a buried glutamate, Glu8, in the pH-responsive disassembly of β-endorphin fibrils using all-atom molecular dynamics simulations along with structure-based pKa prediction. The fibril was stable at pH 5.5 or lower with all the buried Glu8 residues protonated and neutrally charged. After switching to neutral pH, the Glu8 residues of peptides at the outer layers of the ordered fibrils became deprotonated due to partial solvent exposure, causing sheet-to-coil conformational changes and subsequent exposure of adjacent Glu8 residues in the inner chains. Via iterative deprotonation of Glu8 and induced structural disruption, all Glu8 residues would be progressively deprotonated. Electrostatic repulsion between deprotonated Glu8 residues along with their high solvation tendency disrupted the hydrogen bonding between the β1 strands and increased the solvent exposure of those otherwise buried residues in the cross-β core. Overall, our computational study reveals that the strategic positioning of ionizable residues into the cross-β core is a potential approach for designing reversible amyloid fibrils as pH-responsive smart bio-nanomaterials.Phosphoramide, as an important framework of many biologically active molecules, has attracted widespread attention in recent decades. However, little is known about the anti-aging and health-promoting effects of ginkgo seed products. Here, we showed that ginkgo seed powder extract (GSP-E) is abundant in alkaloids and flavonoids, and can extend the lifespan of Caenorhabditis elegans. GSP-E improved most physiological indicators related to aging of C. elegans, including locomotor activity, reproductive capacity, and resistance to oxidation and heat. Moreover, GSP-E reduced the accumulation of lipofuscin and reactive oxygen species (ROS) in C. elegans. Further studies demonstrated that GSP-E improved longevity and stress resistance by mediating lipid metabolism and autophagy, as well as by regulating gene expression (e.g., FASN-1, POD-2, GPX-7, FAT-5). GSP-E has an anti-amyloid effect and delayed amyloid-induced paralysis of C. elegans.
