Acute myeloid leukemia (AML) is a heterogeneous group of leukemias with an overall poor prognosis. Circular RNAs (circRNAs) have been verified to play important regulatory roles in AML progression. We aimed to investigate the usage trends in a nationwide asthma cohort in Denmark from 1999 to 2018. Using national registers, we identified young adults (18-45 years) with two or more asthma drug collections within 12 months since the age of 15 years as indicative of active asthma. OCS exposure level was stratified as high use (≥5 mg prednisolone/day/year) and low use ( less then 5 mg/day/year). Lorenz curves were computed to illustrate potential skewness of consumption among the OCS users. We identified 318 950 individuals with a median age of 29 years (IQR 20-38 years) whereof 57% were women. The 1-year prevalence of OCS users was stable at 4.8% (median, IQR 4.7%-4.8%), but with nearly 40% decrease in high-users from 0.54% in 1999 to 0.33% in 2018. The median annual exposure decreased from 500 mg/year (1999) to 250 mg/year (2018). We found a substantial skewness in the distribution of OCS usage with 10% of users accounting for almost 50% of all OCS use. The prevalence of OCS users among young adults with active asthma has been relatively stable from 1999 to 2018, but with a decreasing prevalence of high-users and annual consumption.
It is unknown how β-adrenergic stimulation affects calcium dynamics in individual RyR2 clusters and leads to the induction of spontaneous calcium waves. To address this, we analysed spontaneous calcium release events in green fluorescent protein (GFP)-tagged RyR2 clusters.

Cardiomyocytes from mice with GFP-tagged RyR2 or human right atrial tissue were subjected to immunofluorescent labelling or confocal calcium imaging.

Spontaneous calcium release from single RyR2 clusters induced 91.4%±2.0% of all calcium sparks while 8.0%±1.6% were caused by release from two neighbouring clusters. Sparks with two RyR2 clusters had 40% bigger amplitude, were 26% wider, and lasted 35% longer at half maximum. Consequently, the spark mass was larger in two- than one-cluster sparks with a median and interquartile range for the cumulative distribution of 15.7±20.1 vs 7.6±5.7 a.u. (P<.01). β2-adrenergic stimulation increased RyR2 phosphorylation at s2809 and s2815, tripled the fraction of two- and three-cluster sparks, and significantly increased the spark mass. Interestingly, the amplitude and mass of the calcium released from a RyR2 cluster were proportional to the SR calcium load, but the firing rate was not. The spark mass was also higher in 33 patients with atrial fibrillation than in 36 without (22.9±23.4 a.u. vs 10.7±10.9; P=.015).

The differential diagnosis of anemia is an important issue for hematology laboratories. We aimed at investigating the performance of a powerful computer-based model to aid diagnosis.

Our work presents a new feature selection-based automated disease diagnosis model. To create a testbed, a new corpus is collected retrospectively. Our data sets contain beta thalassemia trait, iron deficiency anemia, and healthy groups. Our presented automated ailment classification model consists iterative chi2 (IChi2) feature selection and classification phases. The used data set includes 25 features, and IChi2 selects the 20 most valuable of them. These are forwarded to 24 traditional classifiers.

In this work, two data sets have been used to test our proposal. In the classification phase of this model, 24 shallow classifiers have been used and the best accurate classifiers are Medium Gaussian Support Vector Machine (MGSVM) and Coarse Tree (CT) for the first and second data sets, respectively. These classifiers have been attained 97.48% and 99.73% classification accuracies using the first and second data sets, consecutively. These results are calculated using 10-fold cross-validation. Moreover, hold-out validation has been used in this work, and the results are given in the experiments.

To assess the validity of a Written Clinical Reasoning Prompt (WCRP) to help nursing students' clinical reasoning (CR).

This is a methodological study of translation and cross-cultural adaptation into Brazilian Portuguese of instruments that aim to promote CR improvement, composed of (1) WCRP; (2) two case studies; (3) a questionnaire about students' perceptions during decision-making in case studies; (4) a scoring rubric for correcting case studies. For translation and cross-cultural adaptation, stages 1-8 of the Research Diagnostic Criteria for Temporomandibular Disorders (RDC/TMD) Consortium Network were adopted. Agreement values among experts >80% and content validity coefficient (CVC) > 0.8 were considered satisfactory. For the pretest, a randomized clinical trial was carried out with 24 nursing students (intervention group, n = 14, using the WCRP to solve case studies; control group, n = 10, without using the WCRP).

New studies with larger samples, a sample power of at least 80%, and a level of significance of 5% are needed.We sought to apply natural language processing to the task of automatic risk of bias assessment in preclinical literature, which could speed the process of systematic review, provide information to guide research improvement activity, and support translation from preclinical to clinical research. We use 7840 full-text publications describing animal experiments with yes/no annotations for five risk of bias items. We implement a series of models including baselines (support vector machine, logistic regression, random forest), neural models (convolutional neural network, recurrent neural network with attention, hierarchical neural network) and models using BERT with two strategies (document chunk pooling and sentence extraction). We tune hyperparameters to obtain the highest F1 scores for each risk of bias item on the validation set and compare evaluation results on the test set to our previous regular expression approach. The F1 scores of best models on test set are 82.0% for random allocation, 81.6% for blinded assessment of outcome, 82.6% for conflict of interests, 91.4% for compliance with animal welfare regulations and 46.6% for reporting animals excluded from analysis. Our models significantly outperform regular expressions for four risk of bias items. For random allocation, blinded assessment of outcome, conflict of interests and animal exclusions, neural models achieve good performance; for animal welfare regulations, BERT model with a sentence extraction strategy works better. Convolutional neural networks are the overall best models. The tool is publicly available which may contribute to the future monitoring of risk of bias reporting for research improvement activities.
To examine the prevalence of Australian Defence Force veterans among people sleeping rough and explore their health and social needs relative to non-veteran rough sleepers.

Analysis of responses to the Vulnerability Index - Service Prioritisation Decision Assistance Tool (VI-SPDAT) collected from 8,027 rough sleepers across five Australian States from 2010-2017.

Veterans were found to comprise 5.6% of people sleeping rough in Australia, with veterans reporting having spent an average of 6.3 years on the street or in emergency accommodation (compared with an average of five years for their non-veterans counterparts). Veterans had a higher prevalence of self-reported physical health, mental health and social issues compared with non-veteran rough sleepers.

This is the first study of its kind to elucidate the presence of Australian veterans among people sleeping rough. That they are likely to have spent more years on the street, and have a higher prevalence of health and social issues, highlights the impnsequently the myriad of health, psychosocial and adjustment issues faced by homeless veterans has also been hidden. With heightened attention on veteran suicide and self-harm, earlier intervention to prevent veterans becoming homeless constitutes sound public health prevention and mental health policy.The scalable production of hydrogen through water electrolysis is recognized as a green and sustainable pathway for a future energy supply. Benefiting from similar electronic properties as Pt-group metals, molybdenum (Mo)-based carbides and nitrides have been considered as promising catalysts for the hydrogen evolution reaction (HER). Currently, one of the substantial challenges using Mo-based HER electrocatalysts lies in establishing well-defined precursors which can be transformed into Mo-based carbides/nitrides with controllable structure and porosity. Here we report the synthesis of a series of superstructures consisting of organic-polyoxometalate co-crystals (O-POCs) as a new type of metal-organic precursor to synthesize Mo-based carbides/nitrides in a highly controlled fashion and to use them for efficient catalytic hydrogen production. This protocol enables to create electrocatalysts composed of abundant nanocrystallites and heterojunctions with tunable micro- and nanostructure and mesoporosity. The best performing electrocatalyst shows high HER activity and stability with a low overpotential of 162 mV at 100 mA/cm 2 (in comparison to Pt/C with 263 mV), which makes it to one of the best non-noble metal HER catalysts in alkaline media and seawater.
