LACS2-overexpressors (LACS2-OEs) showed improved tolerance to submergence, with higher accumulation of cuticular wax and cutin in their rosettes. In contrast, knockout of LACS2 in the lacs2-3 mutant resulted in hypersensitivity to submergence with reduced wax crystals and thinner cutin layer. By analyses of plant surface permeability, we observed that the hypoxic sensitivities in the LACS2-OEs and lacs2-3 mutant were physiologically correlated with chlorophyll leaching, water loss rates, ionic leakage, and gas exchange. Thus, our findings suggest the role of LACS2 in plant response to submergence by modulating cuticle permeability in plant cells.The emergence of castration-resistance is one of the major challenges in the management of patients with advanced prostate cancer. Although the spectrum of systemic therapies that are available for use alongside androgen deprivation for treatment of castration-resistant prostate cancer (CRPC) is expanding, none of these regimens are curative. Therefore, it is imperative to apply systems approaches to identify and understand the mechanisms that contribute to the development of CRPC. Using comprehensive proteomic approaches, we show that a glycosylation-related enzyme, alpha (1,6) fucosyltransferase (FUT8), which is upregulated in CRPC, might be responsible for resistance to androgen deprivation. Mechanistically, we demonstrated that overexpression of FUT8 resulted in upregulation of the cell surface epidermal growth factor receptor (EGFR) and corresponding downstream signaling, leading to increased cell survival in androgen-depleted conditions. We studied the coregulatory mechanisms of EGFR and FUT8 expression in CRPC xenograft models and found that castration induced FUT8 overexpression associated with increased expression of EGFR. Taken together, our findings suggest a crucial role played by FUT8 as a mediator in switching prostate cancer cells from nuclear receptor signaling (androgen receptor) to the cell surface receptor (EGFR) mechanisms in escaping castration-induced cell death. These findings have clinical implication in understanding the role of FUT8 as a master regulator of cell surface receptors in cancer-resistant phenotypes.This paper investigates the root cause of a failure in gas-turbine blades, made of Nimonic-105 nickel-based superalloy. The failure was reported in two blades in the second stage of a turbine-compressor of a gas turbine in the hot section. Two failed blades were broken from the root and from the airfoil. The failure took place after 20 k h of service exposure in the temperature range 700-850 °C, with the rotating speed being in the range 15,000-16,000 rpm. The microstructures of the failed blades were studied using optical/electron microscopes. Energy dispersive X-ray spectroscopy (EDS) was employed for phase identification. Results showed that failure first initiated from the root. The dominant failure mechanism in the root was concluded to be corrosion-fatigue. The failure scenario was suggested based on the results obtained.Radiation-induced normal tissue toxicity often limits the curative treatment of cancer. We propose a strategy based on transcriptome analysis of patient-derived human intestinal organoids (HIO) to determine molecular surrogates for radioresponse of gastrointestinal (GI) organs at risk in a personalized manner. HIO were generated from induced pluripotent stem cells (iPSC), which were derived from skin biopsies of three patients, including two patients with FANCA deficiency as a paradigm for enhanced radiosensitivity. For the two Fanconi anemia (FA) patients (HIO-104 and 106, previously published as FA-A#1 IND-iPS1 and FA-A#2 IND-iPS3), FANCA expression was reconstituted as a prerequisite for generation of HIO via lentiviral expression of a doxycycline inducible construct. For radiosensitivity analysis, FANCA deficient and FANCA rescued as well as wtHIO were sham treated or irradiated wirbons from proton and photon irradiation. Genes associated with dual incision steps of TC-NER were differentially regulated in photon vs. proton and carbon irradiation. Consequently, SUMO3, ALC1, POLE4, PCBP4, MUTYH expression correlated with the higher RBE of carbon ions. An interaction between the two studied parameters FA and RQ was identified (p less then 0.01, 2-way ANOVA n = 476). A comparison of genes regulated as a function of FA, RQ and RBE suggest a role for p53 interacting genes BRD7, EWSR1, FBXO11, FBXW8, HMGB1, MAGED2, PCBP4, and RPS27 as modulators of FA in response to radiation. Aging and cardiac pathologies are also associated with increased senescence in the heart. However, the cause and consequences of cardiac senescence during aging or in cardiac pathologies are mostly unrecognized. Further, despite recent advancement in anti-senescence therapy, the targeted cell type and the effect on cardiac structure and function have been largely overlooked. The unique cellular composition of the heart, and especially the functional properties of cardiomyocytes, need to be considered when designing therapeutics to target cardiac aging. In recent years, light detection and ranging (LiDAR) sensors have been widely used for gathering information about plant architecture to extract biophysical parameters for decision-making programs. The study reconstructed vineyard crops using light detection and ranging (LiDAR) technology. Its accuracy and performance were assessed for vineyard crop characterization using distance measurements, aiming to obtain a 3D reconstruction. A LiDAR sensor was installed on-board a mobile platform equipped with an RTK-GNSS receiver for crop 2D scanning. The LiDAR system consisted of a 2D time-of-flight sensor, a gimbal connecting the device to the structure, and an RTK-GPS to record the sensor data position. The LiDAR sensor was facing downwards installed on-board an electric platform. It scans in planes perpendicular to the travel direction. Measurements of distance between the LiDAR and the vineyards had a high spatial resolution, providing higd, pruning the remaining weight. The number of scans by LiDAR influenced the relationship with the actual biomass measurements and had a significant effect on the treatments. A positive linear fit was obtained for the comparison between actual dry biomass and LiDAR volume. The influence of individual treatments was of low significance. The results showed strong correlations with actual values of biomass and volume with R2 = 0.75, and when comparing LiDAR scans with weight, the R2 rose up to 0.85. The obtained values show that this LiDAR technique is also valid for branch reconstruction with great advantages over other types of non-contact ranging sensors, regarding a high sampling resolution and high sampling rates. Even narrow branches were properly detected, which demonstrates the accuracy of the system working on difficult scenarios such as defoliated crops.
