We determined Myc is implicated in numerous aspects of gene expression during early-phase muscle fiber growth. Specifically, brief induction of Myc protein in muscle represses Reverbα, Reverbβ, and Myh2 while increasing Rpl3, recapitulating gene expression in myonuclei during acute overload. Experimental, comparative, and in silico analyses place Myc at the center of a stable and actively transcribed, loading-responsive, muscle fiber-localized regulatory hub. Collectively, our experiments are a roadmap for understanding global and Myc-mediated transcriptional networks that regulate rapid remodeling in postmitotic cells. We provide open webtools for exploring the five RNA-seq datasets as a resource to the field.The human cytomegalovirus (HCMV) UL97 protein is a conserved herpesvirus protein kinase (CHPK) and a viral cyclin-dependent kinase (v-CDK). However, mechanisms regulating its activity in the context of infection are unknown. Here, we identified several cellular regulatory 14-3-3 proteins as UL97-interacting partners that promote UL97 stability. Humans are known to encode seven isoforms of 14-3-3 proteins (β, ε, η, γ, σ, θ, and ζ) that bind phosphoserines or phosphothreonines to impact protein structure, stability, activity, and localization. Our proteomic analysis of UL97 identified 49 interacting partners, including 14-3-3 isoforms β, η, and γ. Furthermore, coimmunoprecipitation with Western blotting assays demonstrated that UL97 interaction with 14-3-3 isoforms β, ε, η, γ, and θ occurs in a kinase activity-dependent manner. Using mutational analysis, we determined the serine residue at amino acid 13 of UL97 is crucial for 14-3-3 interaction. We demonstrate UL97 S13A (serine to alanine substitution at residue 13) retains kinase activity but the mutant protein accumulated at lower levels than WT UL97. Finally, we show both laboratory (AD169) and clinical (TB40/E) strains of HCMV encoding UL97 S13A replicated with WT kinetics in fibroblasts but showed decreased UL97 accumulation. Taken together, we conclude that 14-3-3 proteins interact with and stabilize UL97 during HCMV infection.The heme enzyme myeloperoxidase (MPO) is one of the key players in the neutrophil-mediated killing of invading pathogens as part of the innate immune system. MPO generates antimicrobial oxidants, which indiscriminately and effectively kill phagocytosed pathogens. Staphylococcus aureus, however, is able to escape this fate, in part by secreting a small protein called SPIN (Staphylococcal Peroxidase Inhibitor), which specifically targets and inhibits MPO in a structurally complex manner. Here, we present the first crystal structures of the complex of SPIN-aureus and a truncated version (SPIN-truncated) with mature dimeric leukocyte MPO. We unravel the contributions of the two domains to the kinetics and thermodynamics of SPIN-aureus binding to MPO by using a broad array of complementary biochemical and biophysical methods. The C-terminal "recognition" domain is shown to mediate specific binding to MPO, while interaction of the N-terminal "inhibitory" domain is guided mainly by hydrophobic effects and thus is less sequence dependent. We found that inhibition of MPO is achieved by reducing substrate migration, but SPIN-aureus cannot completely block MPO activity. Its' effectiveness is inversely related to substrate size, with no discernible dependence on other factors. Thus, SPIN-aureus is an extremely high-affinity inhibitor and highly efficient for substrates larger than halogens. As aberrant MPO activity is implicated in a number of chronic inflammatory diseases, SPIN-aureus is the first promising protein inhibitor for specific inhibition of human MPO.In France, recent advances in cervical cancer screening include an organized cervical cancer-screening program and the introduction of HPV testing as a first-line test for women aged 30-65 years. Herein, reported the utilization of the microemulsion technique for the preparation of nanoporous fibrous silica-molybdenum oxide (FSMo) towards simultaneous photocatalytic abatement of hexavalent chromium (Cr(VI)) and tetracycline (TC). The FESEM analysis showed the spherical morphology of the FSMo catalyst with dendrimeric silica fiber. The synthesized FSMo catalyst exhibited narrowed bandgap, high crystallinity, and well Mo element dispersion for enhanced photo-redox of Cr(VI) and TC. Remarkably, simultaneous remediation of the Cr(VI) and TC over FSMo demonstrated superior photocatalytic efficiency, 69% and 75%, respectively, than in the individual system, possibly due to the effective separation of photoinduced charges. The introduction of the Mo element to the silica framework via microemulsion technique demonstrated better dispersion of Mo compared to the incipient wetness impregnation method and thus, yielded higher photocatalytic activity towards simultaneous removal of TC and Cr(VI). Besides, quenching experiments revealed the electrons and holes as the active species that play a dominant role in the simultaneous photo-redox of Cr(VI) and TC. The current work examines the synthesis of a nanocomposite combined with Moringa olifera aqueous leaf extract as a reducing and stabilizing agent for the green synthesis of nanocomposites. Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), Thermogravimetric analysis (TGA), Scanning Electron Microscope (SEM), Transmission Electron Microscope (TEM) were used to investigate metal based functional nanocomposites. The absorption band centered at a wavelength of 243 nm, which corresponds to the surface plasmon resonances of the produced nanocomposite, is confirmed in UV-vis spectra. The distinctive band at this particular wavelength is attributed to a particular group of nanocomposites based on the result from the Fourier transform infrared spectroscopy spectra. The spherical with irregularly shaped aggregates was confirmed by transmission electron microscopy, and the average size of nanoparticles was found to be 1 nm. For the elimination of pharmaceutical contaminants such as paracetamol from aqueous solutions, the adsorptive characteristics of nanocomposites were examined. Temperature, pH, adsorbent dosage, and agitation speed were investigated as adsorption parameters using Box-Behnken Design (BBD). The best removal outcomes were found under the following circumstances temperature at 303.15 K, pH = 7.5, 0.05 g of nanocomposites at 200 rpm. Based on the adsorption study, the kinetics was found to be pseudo first order (R2 > 0.9481) which was validated and fitted by Langmuir isotherm (R2 > 0.9973). This study was undertaken to analyze PRs in the water, sediments, shrimps, and feed at different growth stages to evaluate the ecotoxicological and human health risks. PRs in the seawater and sediments ranged from not detected (ND) to 0.027 μg/L and 0.006-12.39 μg/kg, and the concentrations were within the maximum residual limits (MRLs) and sediment quality guidelines prescribed by the World Health Organization and Canadian Environment Guidelines, respectively. PRs in shrimps at three growth stages viz. Postlarvae, juvenile, and adults, ranged from ND to 0.522 μg/kg, below the MRLs set by Codex Alimentarius Commission and European Commission. Most of the PRs in water, sediments, and shrimps did not vary significantly (p > 0.05) from days of culture (DOC-01) to DOC-90. The hazard quotient (HQ) and hazard ratio (HR) were found to be less then 1, indicating that consumption of shrimps has no noncarcinogenic and carcinogenic risks. PRs in shrimp feed ranged from ND to 0.777 μg/kg and were found to be below the MRLs set by EC, which confirms that the feed fed is safe for aquaculture practices and does not biomagnify in animals. The risk quotient (RQ) and toxic unit (TU) ranged from insignificant level (ISL) to 0.509 and ISL to 0.022, indicating that PRs do not pose acute and chronic ecotoxicity to aquatic organisms. The study suggested no health risk due to PRs in shrimps cultured in India and exported to the USA, China, and Japan. However, regular monitoring of PRs is recommended to maintain a sustainable ecosystem.
