We have therefore developed a method for slice selection in low-temperature, DNP-enhanced MRI that limits the FOV to about 50 μm in one or more dimensions. This method uses small-amplitude phase modulation of LG irradiation in the presence of a strong magnetic field gradient to invert spin-locked 1H magnetization in the selected slice. Experimental results are reported, including effects of radio-frequency field inhomogeneity, variations in the amplitude of phase modulation, and shaped phase modulation. Administration of exogenous creatine (Cr) or phosphocreatine (PCr) has been used for improving exercise performance and protecting the heart during surgery including during valve replacements, coronary artery bypass grafting and repair of congenital heart defects. In this work we investigate whether it is possible to use chemical exchange saturation transfer (CEST) MRI to monitor uptake and clearance of exogenous creatine and phosphocreatine following supplementation. We were furthermore interested in determining the limiting conditions for distinguishing between creatine (1.9 ppm) and phosphocreatine (2.6 ppm) signals at ultra-high fields (21 T) and determine their concentrations could be reliably obtained using Bloch equation fits of the experimental CEST spectra. We have tested these items by performing CEST MRI of hind limb muscle and kidneys at 11.7 T and 21.1 T both before and after intravenous administration of PCr. We observed up to 4% increase in contrast in the kidneys at 2.6 ppm which peaked ~30 min after administration and a relative ratio of 1.3 in PCrCr signal. Overall, these results demonstrate the feasibility of independent monitoring of PCr and Cr concentration changes using CEST MRI. Sixty-five years after the discovery of rapid eye movement (REM) sleep, the reasons why we sleep and why we need two states of sleep are still largely unclear. Moreover, the functional relationship between the two types of sleep remains the matter of much conjecture. Several questions come to mind. How does sleep regulation in monophasic and polyphasic animals compare? What are the circadian and homeostatic influences on both states? Are non-rapid eye movement (NREM) and REM states dependent on each other, or are they regulated independently? What about long-term and short-term regulation? In addition, what determines the number and duration of cycles per night? What roles are played by temperature and energy allocation? The evidence collected over the years regarding these questions is summarized here, trying to address each issue. BACKGROUND/OBJECTIVES The relationship between physical activity and sleep quality has been reported in specific conditions or in selected individuals. To better understand this association at the population level, we aimed to assess the association between physical activity and sleep quality in community-dwellers residing in three neighboring rural villages of Coastal Ecuador. METHODS A total of 1438 individuals aged ≥40 years were included. Data collection focused on the levels of physical activity and sleep quality. Multivariate models were fitted to assess independent associations between adequate physical activity and good sleep quality, after adjusting for relevant covariates. RESULTS Physical activity was categorized as ideal in 676 (47%) individuals, intermediate in 644 (45%), and poor in 118 (8%). According to the Pittsburgh Sleep Quality Index (PSQI), 880 (61%) individuals had a good sleep quality. In univariate analysis, the percentage of individuals with a good sleep quality was lower among individuals with poor and intermediate physical activity when compared to those with ideal physical activity. Using individuals with poor physical activity as the referent category, a logistic regression model showed that a good sleep quality was significantly associated with intermediate (odds ratio, OR 1.54; 95% confidence interval, CI 1.03-2.30; p = 0.036) and ideal (OR 1.78; 95% C.I. 1.18-2.69; p = 0.006) physical activity, after adjusting for relevant covariates. CONCLUSIONS This population-based study provides robust epidemiological evidence favoring an association between adequate levels of physical activity and good sleep quality in middle-aged and older adults living in rural settings. IMPORTANCE Although several strategies using transcranial direct current stimulation (tDCS) have been investigated to treat major depressive disorder (MDD), the efficacy of this treatment for patients with MDD who also have insomnia is unclear. OBJECTIVE To observe the effects of tDCS on sleep quality and depressive symptoms in patients with MDD who have insomnia. METHODS We conducted a randomized, double-blinded study involving adults with major depression and insomnia. We randomly assigned patients to either add tDCS or to sham tDCS to their regular treatment. After randomization, we treated a total of 90 patients at the Kangning Hospital, Ningbo, China. We allocated 47 patients to the tDCS group and 43 to the sham tDCS group. The tDCS treatment procedure included 20 sessions of 2-mA stimulation of the dorsolateral prefrontal cortex (DLPFC) for 30 min, which was followed by four weekly treatments. The anode and cathode electrodes were placed on the left and right DLPFC, respectively. We recorded the Self-rating Depression Scale (SDS), Self-rating Anxiety Scale (SAS), Pittsburgh Sleep Quality Inventory (PSQI), and Polysomnography (PSG) at Day 1 and Day 28. RESULTS Compared with the sham tDCS group, the active tDCS group showed improved total scores of SAS and SDS. PSQI total score and all PSQI sub-divisions, except for "sleep duration and sleep efficiency," significantly improved after treatment. We also observed that tDCS affected sleep architecture, by increasing total sleep time and improving sleep efficiency through PSG. CONCLUSIONS Our study demonstrated the effect of tDCS on sleep quality and depressive symptoms in patients with MDD and insomnia. These results suggested that tDCS stimulation not only improved symptoms of depression and anxiety but also had a positive effect on sleep quality in patients with MDD. For patients with depression and insomnia, tDCS stimulation could be a good supplement to drugs. BACKGROUND Iron overload can result in a disorder in glucose metabolism. However, the underlining mechanism through which iron overload induces beta cell death remains unknown. METHODS According to the concentration of ferric ammonium citrate (FAC) and N-acetylcysteine, INS-1 cells were randomly divided into four groups normal control (FAC 0 μM) group, FAC 80 μM group, FAC 160 μM group, FAC 160μM + NAC group. Cell proliferation was assessed by Cell Counting Kit-8. Reactive oxygen species (ROS) level was further evaluated using flow cytometer with a fluorescent probe. The mitochondrial membrane potential was detected by JC-1 kit, and transmission electron microscopy was used to observe the mitochondrial changes. The related protein expressions were detected by western bolt to evaluate mitophagy status. RESULTS It was shown that FAC treatment decreased INS-1 cell viability in vitro, resulted in a decline in mitochondrial membrane potential, increased oxidative stress level and suppressed mitophagy. Furthermore, these effects could be alleviated by the ROS scavenger. CONCLUSIONS We proved that increased iron overload primarily increased oxidative stress and further suppressed mitophagy via PTEN-induced putative kinase 1/Parkin pathway, resulting in cytotoxicity in INS-1 cells.
