Interferon regulatory factor 4 (IRF4) is a crucial member of IRF family, which acts as an imperative transcription factor in the development and maturation of multiple lineages of blood cells and also plays a pivotal role in host defense against microbial infections. In the present study, we aimed to investigate the detailed structural and functional aspects of a redlip mullet IRF4 homolog (LhIRF4). The LhIRF4 open reading frame consists of 1347 base pairs encoding 449 amino acids, with the DNA-binding domain sharing significant homology with that of other vertebrate IRF4 homologs. The highest transcription levels of LhIRF4 were observed in the mullet intestine and spleen under normal physiological conditions. Furthermore, a time-dependent upregulation of LhIRF4 transcription was observed in the spleen and head kidney tissues upon pathogenic challenges. When overexpressed in mullet cells, LhIRF4 was localized to the nucleus and significantly stimulated the transcription of several host antiviral genes. Moreover, the overexpression of LhIRF4 strongly inhibited the replication of viral hemorrhagic septicemia virus (VHSV) in vitro. The function of LhIRF4 in regulation of macrophage M2 polarization has also been evidently demonstrated in RAW 264.7 cells. Taken together, our findings indicate the profound role of LhIRF4 in modulating immune responses against microbial infections in redlip mullet.Lytic polysaccharide monooxygenases (LPMOs) are copper-dependent enzymes, which can catalyze the oxidative cleavage of polysaccharide β-1,4 glycosidic bonds to improve the hydrolysis efficiency of the substrate by other glycoside hydrolases. To improve the conversion efficiency of cellulose and chitin, a strain was screened from the soil of Yuelu Mountain in Hunan province, China. The gene sequence of a novel AA10 LPMO (BaLPMO10) was successfully cloned from the genome of the strain and heterologously expressed in E. coli BL21 (DE3). The optimal enzyme activity of BaLPMO10 was observed at pH 6.0 and 70 °C using 2,6-dimethoxyphenol as substrate, and its maximum specific activity was 91.4 U/g. When BaLPMO10 synergized with glycoside hydrolase to degrade microcrystalline cellulose and colloidal chitin, the reducing sugar content increased by 7% and 23%, respectively, compared to glycoside hydrolase alone. Moreover, the results of molecular docking and molecular dynamics simulation showed that the distance between BaLPMO10 and cellohexaose were further than that of BaLPMO10 and chitohexaose, and the number of hydrogen bonds between BaLPMO10 and cellohexaose were lower than that of BaLPMO10 and chitohexaose. Finally, the hot spot residues of BaLPMO10 interacting with chitohexaose/cellohexaose were identified.Carboxymethyl cellulose/ graphene composite aerogel beads (CMC/GAs) were prepared by the easily scaling-up method, i.e., wet spinning- environmental pressure drying method. The influences of the type and concentration of coagulating bath on the formation of aerogel beads were discussed, and the forming mechanism was analyzed. The CMC/GAs was characterized through SEM, XRD, FI-IR, Raman, XPS, electronic universal testing machine and other methods. The CMC/GAs-30 has an average particle size and a mean pore diameter of 3.83 mm and 82 μm, respectively. The analysis results indicated that the adsorption mechanisms of CMC/GAs on methylene blue (MB) are mainly through the electrostatic interaction. The adsorption process conforms to the Langmuir model (R2 = 0.9964) and pseudo-second-order kinetic model (R2 is higher than 0.99). When the particle size of CMC/GAs-30 decreases, the equilibrium adsorption capacity for MB increases. Under the experimental conditions explored, the Langmuir maximum adsorption capacity of CMC/GAs-30 for MB is 222.72 mg.g-1. The CMC/GAs-30 show good recycle performance in MB adsorption. The removal rate of MB from water by CMC/GAs-30 remained at about 90% after 30-times adsorption- regeneration cycles.Virus-like particles (VLPs) are nano-scale particles that are morphologically similar to a live virus but which lack a genetic component. CoVs contain four structural proteins, though the minimum requirement for VLP formation differs among virus species. CoV VLPs are commonly produced in mammalian and insect cell systems, sometimes in the form of chimeric VLPs that enable surface display of CoV epitopes. VLPs are an ideal model for virological research and have been applied as vaccines and diagnostic reagents to aid in clinical disease control. This review summarizes and updates the research progress on the characteristics of VLPs from different known CoVs, mainly focusing on assembly, in vitro expression systems for VLP generation, VLP chimerism, protein-based nanoparticles and their applications in basic research and clinical settings, which may aid in development of novel VLP vaccines against emerging coronavirus diseases such as SARS-CoV-2.Aging is a process involving physiological changes that lead to the decline of biological functions of various tissues and organs of the body. Although T helper type 2 cytokines downregulate the expression of epidermal barrier proteins, the signaling mechanism underlying these effects remains unclear. IL-33, a chromatin-associated cytokine, is highly expressed in the nuclei of epidermal keratinocytes in AD skin; however, it is unclear whether this expression promotes the development of AD. Immunology inhibitor TSLP, an epithelial cells-derived pro‒T helper type 2 cytokine, is elevated in the epidermis of patients with AD. TSLP affects the pathogenesis of AD by activating T helper type 2 responses and impairing epidermal barrier integrity. In this study, we stimulated postconfluent human keratinocytes and living skin equivalent with TSLP to investigate the role of nuclear IL-33 in TSLP-induced epidermal barrier defects. We observed that TSLP reduced the levels of FLG, hBD2, S100A7, and claudin-1, which required nuclear IL-33 expression. Similar to the T helper type 2 cytokines IL-4, IL-13, and IL-31, TSLP was shown to upregulate IL-33 expression and triggered the formation of nuclear IL-33/phosphorylated signal transducer and activator of transcription 3 complex, which bound to the FLG promoter, thereby inhibiting transcription. Moreover, nuclear IL-33 acted as a cofactor of signal transducer and activator of transcription 3 in the TSLP-induced transcriptional repression of hBD2, S100A7, and claudin-1. Therefore, epidermal nuclear IL-33 may be a key regulator of TSLP-mediated epidermal barrier dysfunction.Much of the research from Canada's oil sands region (OSR) shows contaminants of concern (CoCs) throughout the ambient environment surrounding the industrial facilities. While there are some well-established sources of the CoCs, there is also spatial and temporal variability suggesting activity intensity, changes in technology, types and amounts of fuels combusted at the facilities, and climate may affect the results of deposition studies. This study re-analysed published data on the deposition of elements and polycyclic aromatic compounds (PACs) in snow and the sediments of some lakes by incorporating production data from facilities and climate. Using the Elastic Net (EN) regularized regression, variables describing potential associations between facility-specific activity and climate on the deposition of CoCs were identified. Among the selected variables, the combustion of delayed petroleum coke at the Suncor Basemine was associated with the deposition of CoCs, including elements in snow and in some lakes. Srelease.Decomposition of plant organic matter plays a key role in the terrestrial biogeochemical cycles. Sunlight has recently been identified as an important contributor to carbon [C] turnover through photodegradation, accelerating decomposition even in forest ecosystems where understorey solar irradiance remains relatively low. However, it is uncertain how C and nutrients dynamics respond to fluctuations in solar spectral irradiance caused by canopy structure (understorey vs. gaps) and season (open vs. closed canopy phenology). Spectral-attenuation treatments were used to compare litter decomposition over eight months, covering canopy phenology, in a temperate deciduous forest and an adjacent gap. Exposure to the full spectrum of sunlight increased the loss of litter C and lignin by 75% and 64% in the forest gap, and blue light was responsible for respectively 27% and 42% of that loss. Whereas in the understorey, C and lignin loss were similar among spectral-attenuation treatments over the experimental period, except prior to and during spring canopy flush when exposure to the full spectrum of sunlight promoted C loss by 15% overall, 80% of which was attributable to ultraviolet-B (UV-B) radiation. Nitrogen [N] was immobilized in the understorey during canopy flush before the canopy completely closed but N was swiftly released during canopy leaf-fall. Our study suggests that blue-driven photodegradation plays an important role in lignin decomposition and N dynamics in canopy gaps, whereas seasonal canopy phenology affecting sunlight reaching the forest floor drastically changes patterns of C and N in litter during decomposition. Hence, including sunlight dynamics driven by canopy structure and phenology would improve estimates of biogeochemical cycling in forests responding to changes in climate and land-use.The organic flame retardants (OFRs) have attracted global concerns due to their potential toxicity and ubiquitous presence in the aquatic environment. Hormesis refers to a biphasic dose response, characterized by low-dose stimulation and high-dose inhibition. The present study provided substantial evidence for the widespread occurrence of OFRs-induced hormesis in aquatic animals, including 202 hormetic dose response relationships. The maximum stimulatory response (MAX) was commonly lower than 160% of the control response, with a combined value of 134%. Furthermore, the magnitude of MAX varied significantly among multiple factors and their interactions, such as chemical types and taxonomic groups. Moreover, the distance from the dose of MAX to the no-observed-adverse-effect-level (NOAEL) (NOAEL MAX) was typically below 10-fold (median = 6-fold), while the width of the hormetic zone (from the lowest dose inducing hormesis to the NOAEL) was approximately 20-fold. Collectively, the quantitative features of OFRs-induced hormesis in aquatic animals were in accordance with the broader hormetic literature.
