This paper described a high-performance molecular test for the detection of Mycobacterium bovis (M. bovis) based on digital loop-mediated isothermal amplification (dLAMP). M. bovis is a persistent pathogen that causes zoonotic tuberculosis and can infect both animals and human beings. The detection of M. bovis in milk samples is critical for effective control and prevention of zoonotic diseases but there lacks effective and sensitive methods. Here, we developed a convenient and low-cost system for M. bovis detection in milk, which incorporated automated DNA extraction and dLAMP by interfacial emulsification technique. Versus real-time PCR, dLAMP provides higher accuracy and sensitivity for direct M. bovis detection in milk, offering a limit of detection of 14 CFU/mL within 2 h. The dLAMP system can become a powerful platform for the detection of pathogens in complex samples and provide more reliable guidance for food safety testing, epidemiological research and clinical diagnosis.Presence of bisphenol A (BPA) and its analogues in various environmental matrices have been well studied by now, but there is still an insufficient data on the occurrence of BPA analogues in animal-derived foodstuffs. Hence, this work describes a quick, sensitive and reliable analytical method for the simultaneous determination of eight BPA analogues in eggs, applying a solid-phase extraction procedure with PRiME HLB cartridges that combines isolation of the analytes from matrix and the sample clean-up on a single column. The limits of detection (LODs) and quantification (LOQs) for BPA analogues were in the ranges of 0.02-0.05 μg kg-1 and 0.1-0.2 μg kg-1, respectively. Average recoveries were ranged between 82.7% and 105.2%. Quantitative analysis was performed on 24 real samples, the results indicate that low levels of BPA, BPB and BPS were detected, and interestingly, over ten folds higher BPP contamination levels compared to BPA were found in two egg samples. Objective To investigate resource use and time investments of healthcare professionals, patients and their family and to compare healthcare and societal costs of one single hospital-based and one single home-based subcutaneous administration of trastuzumab in The Netherlands. Method We conducted a bottom-up micro-costing study. Patients diagnosed with HER2+ early or metastatic breast cancer were recruited in four Dutch hospitals. For healthcare costs, data were collected on drug use, consumables, use of healthcare facilities, time of healthcare professionals, and travelling distance of the nurse. For societal costs, data were collected on patient and family costs (including travelling expenses and time of informal caregivers) and productivity losses of paid and unpaid work. Results Societal costs of one single administration of SC trastuzumab were €1753 within the home-based and €1724 within the hospital-based setting. Tofacitinib mouse Drug costs of trastuzumab were identical in both settings (€1651). Healthcare costs were higher for home-based administration (€91 versus €47) mainly because of more time of healthcare professionals (110 versus 38 minutes). Costs for patient and family were, however, lower for home-based administration due to travelling expenses (€7 versus €0) and time of informal caregivers (€14 versus €4). Costs for productivity losses were similar for both settings. Conclusions Home-based subcutaneous administration of trastuzumab is more time consuming for healthcare professionals and therefore more costly than hospital-based administration. The total budget impact can be large considering that a large number of patients receive a large number of cycles of oncology treatments. If home-based administration is the way forward, novel approaches are crucial for ensuring efficiency of home-based care. The aim of this study was to optimize the laccase immobilization on a silica/chitosan composite support. The innovative method proposed is based on the use of Ca(II) as a linker of the support precursors. In addition for the laccase immobilization the glycosidic part of enzyme is oxidized by periodate in order to form aldehydes reactive towards the NH2 groups of chitosan. The immobilized laccase presents a good storage stability (7 months 40% residual activity) and a good affinity towards ABTS substrate (Kmapp 0.008 mM, Vmaxapp = 0.0034 μmol/min). Furthermore, the biocatalyst showed optimum pH 3 and optimum incubation temperature of 50 °C.Introduction Citicoline can have beneficial effects both in degenerative and in vascular cognitive decline; it works through an increase in acetylcholine intrasynaptic levels and promoting phospholipid synthesis, (chiefly phosphatidylcholine), cellular function, and neuronal repair. Memantine is an N-methyl-D-aspartate (NMDA) receptor antagonist used for the treatment of mild to moderate Alzheimer's disease (AD). When co-administered they could have a synergistic action in patients affected with AD and mixed dementia (MD) too. Scope The aim of the present study was to show the effectiveness of oral citicoline plus memantine in patients affected with AD and MD. Patients and methods This was a retrospective study between 2015 and 2017 on 126 patients aged 65 years old or older affected with AD or MD (mean age 80.7 ± 5.2 years old). The study involved four different centers for dementia all over Italy. Diagnosis of AD was made according to clinical symptoms, neuropsychological tests and brain imaging. Diagnosis ptoms. Results and conclusions Patients treated with citicoline plus memantine showed an increase in MMSE between T0 and T1 (16.6 ± 2.9 vs 17.4 ± 2.7) and between T1 and T2 (17.4 ± 2.7 vs 17.7 ± 2.8). The difference in MMSE score was significant when comparing the two groups, both at T1 (p = 0.003) and T2 (p = 0.000). Since it is important to maximize the pharmacological means in AD and MD, the present study encourages the role of combined administration of memantine plus citicoline in disease management and in slowing down the progression of disease.Purpose This study aimed to compare the physical fitness levels of nursing home residents and community-dwelling older adults. Materials and methods The 118 older adults aged between 65-85 living in a nursing home or community participated in the study. The Senior Fitness Test assessed the physical fitness levels of older adults. The older adults performed the chair stand test, arm curl test, 2-min step test, chair sit and reach test, back-scratch test, 8-foot up, and go test. Results The age, cognitive status, gender, body weight, height, smoking, and education status were similar between the groups (p > 0.05). Aerobic endurance, balance, and agility were higher in nursing home residents than in the community-dwelling older adults (p 0.05). Conclusion Aerobic endurance, balance, and agility may vary depending on the living environment of older adults. The determination of differences in physical fitness between community-dwelling older adults and nursing home residents could provide objective information to develop a physical activity program for older adults.Triclosan (TCS), as an antimicrobial agent, is considered as a representative emerging contaminant and was frequently detected in excess sludge. This study investigated the effect of TCS on activate wastewater sludge (WAS) digestion through laboratory methane production experiment. It was concluded that TCS had a tendency to restrain methane production from sludge with its exposure level increasing. The results displayed that the yields of final maximum cumulative methane production were similar about 108.4 mL/g VSS at TCS level lower 200 mg TCS/kg TSS, while the values were approximately 95.2 mL/g VSS with TCS level over 550 mg TCS/kg TSS. Although TCS could be degraded, its intermediates in this study had no effect on sludge digestion. In addition, TCS at higher levels had seriously negative effect on the solubilization, hydrolysis, acidification, and methanogenesis processes. Microbial community was further analyzed to understand the TCS's effect on digestion system from a micro perspective.In-situ detection on primary volatiles and stable radicals is of great importance for better understanding of lignin pyrolysis mechanisms and utilization. In this study, a novel in-situ pyrolysis time-of-flight mass spectrometry with double ionization sources was taken to in-situ detect primary volatiles and gas products, and the evolution of stable radicals in lignin pyrolysis residues was explored by EPR spectroscopy. The results show that the cleavage of β-O-4 linkage is mainly responsible for lignin depolymerization at 100-300 °C, releasing the G-type compounds. And these G-type compounds can further undergo O-CH3, Car-OCH3 and Car-OH bonds cleavage to form biphenolic hydroxyl compounds, phenols and aromatic hydrocarbons. According to the EPR analysis, the radical concentration increased from 1017 to 1019 spins/g with the temperature, and stable free-radical species are mainly composed of the o-methoxy and hydroxyl substituted phenoxy radicals and carbon-centered aromatic radicals, which can well interpret the demethylation, demethoxylation and dehydroxylation mechanisms. This study investigates the integration of hydrothermal carbonisation (HTC) with anaerobic digestion (AD) as a valorisation route for two macroalgae species; S. latissima (SL) and F. serratus (FS). HTC reactions were conducted at temperatures of 150 °C, 200 °C and 250 °C, with resulting hydrochars, process waters and hydrothermal slurries assessed for biomethane potential yields. Un-treated SL generated similar biomethane levels compared to all SL slurries. Whereas all FS slurries improved biomethane yields compared to un-treated FS. Hydrochars represent a greater energy carrier if used as a solid fuel, rather than a feedstock for anaerobic digestion. Integrating HTC and AD, through hydrochar combustion and process water digestion has a greater energetic output than anaerobic digestion of the un-treated macroalgae. Treatment at 150 °C, with separate utilisation of products, can improve the energetic output of S. latissima and F. serratus by 47% and 172% respectively, compared to digestion of the un-treated macroalgae.Acute thromboembolic events appear to be frequent in patients with SARS-CoV-2 infection. We report a case of an intubated patient, who developed a threatening lower limb ischemia. Intra-arterial fibrinolysis and intravenous heparin infusion did not lead to complete recanalization of the tibial arteries, which were successfully treated by surgical embolectomy.Introduction Parasitic infections of the eye are a major cause of ocular-surface diseases globally. While most infections are treatable, parasites can cause varying levels of damage mostly due to late diagnosis or misdiagnosis as a result of doctors' unfamiliarity with their characteristics of latency and crypsis, as well as lack of awareness by the patients. Case reports In this study, we present three cases of phthiriasis palpebrarum, thelaziasis, and ophthalmomyiasis, respectively. Two of the cases were treated at the clinic and did not recur. One patient refused treatment and was lost to follow-up. Discussion By evaluating the natural histories, morphology, symptoms, clinical findings, and treatment of these parasitic diseases, we systematically analyzed several distinct and unique parasite characteristics, especially latency and crypsis. Furthermore, we have proposed specific examination techniques and methods as well as prevention and treatment strategies from these specific perspectives, aiming to prompt timely diagnoses and early interventions for these diseases by health care workers and improve the public's awareness of parasitic infections.
