Chromosome region maintenance 1 (CRM1) is the sole nuclear exporter of several tumor suppressor, a growth regulatory protein as an attractive cancer drug target. In the present work, a novel CRM1 degrader was discovered from newly synthesized α, β-unsaturated-δ-lactone based on a natural product Goniothalamin. It induces apoptosis of both MGC803 and HGC27 cell lines via degrading CRM1. Selective inhibition was observed for the proliferation of gastric cancer cell lines MGC803, HGC27 comparing to Human Gastric Mucosal Epithelial Cell Line (GES1). For the first time, CRM1 inhibitor or degrader inducing apoptosis in gastric carcinoma was investigated.This review fully describes the coronavirus 3CLpro peptidomimetic inhibitors and nonpeptidic small molecule inhibitors developed from 2010 to 2020. Specifically, the structural characteristics, binding modes and SARs of these 3CLpro inhibitors are expounded in detail by division into two categories peptidomimetic inhibitors mainly utilize electrophilic warhead groups to covalently bind the 3CLpro Cys145 residue and thereby achieve irreversible inhibition effects, whereas nonpeptidic small molecule inhibitors mainly interact with residues in the S1', S1, S2 and S4 pockets via hydrogen bonds, hydrophobic bonds and van der Waals forces. Based on the emerging PROTAC technology and the existing 3CLpro inhibitors, 3CLpro PROTAC degraders are hypothesised to be next-generation anti-coronavirus drugs.Chagas disease is one of the most prevalent tropical neglected diseases and causes high mortality and morbidity in endemic countries. Current treatments for this disease, nifurtimox and benznidazole, are ineffective in the chronic phase of the disease and produce severe adverse effects. Therefore, novel therapies are urgently required. The trace element selenium has an important role in human health, due to its antioxidant, antiinflammatory and pro-immune properties. Actually, its deficiency has been related to several diseases and supplementation with this element has been proven to be beneficial for multiple pathologies. Furthermore, the usefulness of organic-selenium compounds has been studied in many disorders, showing promising results. The aim of this review is to analyse the available literature regarding the role of selenium in Chagas disease in order to determine whether its use could be beneficial for the management of this pathology.The perceived value of our possessions extends well beyond their monetary worth or utility. FoundationOne® next-generation sequencing identified single-nucleotide variants (SNVs), copy number alterations, high TMB (TMB-H) and high-grade microsatellite instability (MSI-H) in patients treated in the FIRE-3 trial. Data were correlated with objective response rate (ORR), progression-free survival (PFS) and overall survival (OS).
Three hundred seventy-three (49.6%) of 752 patients provided material for this analysis. Frequent SNVs included TP53, APC, KRAS, PIK3CA, BRAF, SMAD4 and FBXW7. KRAS, BRAF V600E and SMAD4 mutations were confirmed as prognostic biomarkers by logistic penalised regression for ORR. OS was significantly longer in patients with SMAD4 wild-type (WT) tumours than in those with SMAD4-mutated tumours (hazard ratio=0.59 [95% confidence interval CI = 0.34-1.01], p=0.05), with a higher probability of ORR [odds ratio, SMAD4 SNV versus WT=0.32 [95% CI = 0.10-0.98], p=0.05] when treated with cetuximab. MSI-H (30.0%, p=0.03) and TMB-H (17.3%, p=0.003) tumours were enriched by FBXW7 mutations. Numerically lower ORR, OS and PFS were observed in MSI-H tumours.

To avoid their accumulation, raw water in the Netherlands and many other countries is purified by sand filtration. These drinking water filtration systems select for microbial communities that mediate the biodegradation of organic and inorganic compounds. In this study, the top layers and wall biofilm of a Dutch drinking water treatment plant (DWTP) were sampled from the filtration units of the plant over three years. We used high-throughput sequencing in combination with differential coverage and sequence composition-based binning to recover 56 near-complete metagenome-assembled genomes (MAGs) with an estimated completion of ≥70% and with ≤10% redundancy. These MAGs were used to characterize the microbial communities involved in the conversion of ammonia and methane. The methanotrophic microbial communities colonizing the wall biofilm (WB) and the granular material of the primary rapid sand filter (P-RSF) were dominated by members of the Methylococcaceae and Methylophilaceae. The abundance of these bacteria drastically decreased in the secondary rapid sand filter (S-RSF) samples. In all samples, complete ammonia-oxidizing (comammox) Nitrospira were the most abundant nitrifying guild. Clade A comammox Nitrospira dominated the P-RSF, while clade B was most abundant in WB and S-RSF, where ammonium concentrations were much lower. In conclusion, the knowledge obtained in this study contributes to understanding the role of microorganisms in the removal of carbon and nitrogen compounds during drinking water production. We furthermore found that drinking water treatment plants represent valuable model systems to study microbial community function and interaction.Although a combination of aggregate characteristics dictate particle settling, it is commonly assumed that large particles have higher terminal velocities. This simplifying assumption often leads to overprediction of large aggregate settling velocities which in turn negatively impacts on estimates of sedimentation clarification efficiency. Despite its importance, little attention has been given to large aggregates with slow-settling velocities. This paper addresses this gap by investigating slow-settling velocities of large, heterodisperse and multi-shape Al-kaolin aggregates using non-intrusive methods. A particle image velocimetry technique (PIV) was applied to track aggregate velocity and a non-intrusive image technique was used to determine aggregate characteristics, including size (df), three-dimensional fractal dimension (Df), density (ρf), aggregate velocity (Vexp) and Reynolds number (Re). Results showed no strict dependence of settling velocity on large aggregate size, shape and density, as Al-kaolin aggregates with the same size exhibited different settling velocities. A comparison of the results with the well-known Stokes' law for velocity modified by a shape factor showed that the settling velocities measured here can vary from 2 to 14 fold lower than the predicted values for perfect sphere-shape aggregates with the same density and size. Furthermore, results have also shown large Al-kaolin aggregate's drag coefficient (Cd) to be around 56/Re, for average fractal aggregate sphericity of around 0.58.Algogenic organic matter (AOM) produced by Microcystis cells inhibits coagulation harvesting; however, the harvesting inhibitory mechanisms at the functional groups level remain to be determined. This study fractionated extracellular organic matter (EOM) and intercellular organic matter (IOM) from Microcystis flos-aquae into five different hydrophilic and hydrophobic fractions and investigated their inhibition of chitosan coagulation harvesting. The proton-active functional groups in the inhibitory fractions were further analysed by potentiometric titration, and the interaction between these functional groups and chitosan was elucidated. The results showed that the harvesting inhibition of M. flos-aquae cells was dominated by HPI in AOM due to its high charge density, which resulted in greater consumption of coagulant. Potentiometric titration results suggested that the proton-active functional groups of both HPIEOM and HPIIOM consist mainly of phosphodiester, carboxylic, phosphoryl and amine/hydroxyl functional groups, and the harvesting inhibition of HPI on M. flos-aquae cells at pH 6.5 was mainly due to the deprotonation of phosphodiester and carboxylic functional groups. Moreover, carboxylic functional groups with stronger polarity could enhance the intermolecular interaction between HPI and chitosan more effectively than phosphodiester at pH 6.5. Preventing the deprotonation of carboxylic functional groups by adjusting the pH to 4.3 could effectively alleviate the harvesting inhibition caused by HPI. These findings revealed the inhibition mechanism of AOM on the coagulation harvesting of M. flos-aquae cells from the perspective of deprotonation of proton-active functional groups, which may provide important insights for assessing the role of AOM in the coagulation harvesting of Microcystis cells.
