Further patients with high β2 antibodies had significantly more frequently activated HLA-DR+ T cells and more frequently thyreoperoxidase and anti-nuclear antibodies. In patients receiving rituximab maintenance treatment achieving prolonged B-cell depletion, elevated β2 and M4 receptor autoantibodies significantly declined in clinical responder, but not in non-responder. We provide evidence that 29.5% of patients with CFS had elevated antibodies against one or more M acetylcholine and β adrenergic receptors which are potential biomarkers for response to B-cell depleting therapy. The association of autoantibodies with immune markers suggests that they activate B and T cells expressing β adrenergic and M acetylcholine receptors. Dysregulation of acetylcholine and adrenergic signalling could also explain various clinical symptoms of CFS.Polyozellus multiplex (PM), a Korean edible mushroom, has biological activities such as chemoprevention of stomach cancer, inhibition of lipid peroxidation, and reduction of prolyl endopeptidase activity. However, there are little reports on the protective effects of PM or its constituents against glutamate-induced mouse hippocampal neuronal cell (HT22) death. In this study, polyozellin (PZ), a key constituent of PM, was applied to glutamate-treated HT22 cells to evaluate its neuroprotective mechanisms. PZ (25 μM) dramatically increased the HT22 cell viability when the cell death was induced by 5 mM glutamate for 12 h, which was mediated by inhibition of Ca(2+) influx, intracellular reactive oxygen species (ROS) production, and lipid peroxidation. PZ also regulated expression of Bid, Bcl-2, and apoptosis-inducing factor (AIF), as well as phosphorylation of mitogen-activated protein kinases (MAPKs). These data suggest that PM and its constituent PZ might be useful for prevention and treatment of neurodegenerative disorders.Weyl and Dirac semimetals recently stimulate intense research activities due to their novel properties. Combining first-principles calculations and effective model analysis, we predict that nonmagnetic compounds BaYBi (Y = Au, Ag and Cu) are Dirac semimetals. As for the magnetic compound EuYBi, although the time reversal symmetry is broken, their long-range magnetic ordering cannot split the Dirac point into pairs of Weyl points. However, we propose that partially substitute Eu ions by Ba ions will realize the Weyl semimetal.Longitudinal tracking is a powerful approach to understand the biology of single cells. In cancer therapy, outcome is determined at the molecular and cellular scale, yet relationships between cellular response and cell fate are often unknown. The selective inhibitor of nuclear export, selinexor, is in development for the treatment of various cancers. Selinexor covalently binds exportin-1, causing nuclear sequestration of cargo proteins, including key regulators of the cell cycle and apoptosis. The cell cycle effects of selinexor and the relationships between cell cycle effects and cell fates, has not been described for individual cells. Using fluorescent cell cycle indicators we report the majority of cell death after selinexor treatment occurs from a protracted G1-phase and early S-phase. G1- or early S-phase treated cells show the strongest response and either die or arrest, while those treated in late S- or G2-phase progress to mitosis and divide. Importantly, the progeny of cell divisions also die or arrest, mostly in the next G1-phase. Cells that survive selinexor are negative for multiple proliferation biomarkers, indicating a penetrant, arrested state. Selinexor acts quickly, shows strong cell cycle selectivity, and is highly effective at arresting cell growth and inducing death in cancer-derived cells.To find cheap, efficient and durable hydrogen evolution reaction catalysts is one of the major challenges when developing an alkaline water electrolysis system. In this paper we describe an electrochemically reduced graphene oxide (RGO)-modified Ni electrode, which could be used as a pre-eminent candidate for such a system. The experimentally determined characteristics of this electrode showing superior electrocatalytic activity were complemented by density functional theory calculations. Thermodynamic considerations led to the conclusion that H atoms, formed upon H2O discharge on Ni, spill onto the RGO, which serves as an H adatom acceptor, enabling continuous cleaning of Ni-active sites and an alternative pathway for H2 production. This mode of action is rendered by the unique reactivity of RGO, which arises due to the presence of O surface groups within the graphene structure. The significant electrocatalytic activity and life time (>35 days) of the RGO towards the HER under conditions of alkaline water electrolysis are demonstrated.Ion-sensitive field-effect transistors (ISFETs), although they have attracted considerable attention as effective immunosensors, have still not been adopted for practical applications owing to several problems (1) the poor sensitivity caused by the short Debye screening length in media with high ion concentration, (2) time-consuming preconditioning processes for achieving the highly-diluted media, and (3) the low durability caused by undesirable ions such as sodium chloride in the media. Here, we propose a highly sensitive immunosensor based on a self-amplified transistor under dual gate operation (immuno-DG ISFET) for the detection of hepatitis B surface antigen. To address the challenges in current ISFET-based immunosensors, we have enhanced the sensitivity of an immunosensor by precisely tailoring the nanostructure of the transistor. In the pH sensing test, the immuno-DG ISFET showed superior sensitivity (2085.53 mV per pH) to both standard ISFET under single gate operation (58.88 mV per pH) and DG ISFET with a non-tailored transistor (381.14 mV per pH). Moreover, concerning the detection of hepatitis B surface antigens (HBsAg) using the immuno-DG ISFET, we have successfully detected trace amounts of HBsAg (22.5 fg mL(-1)) in a non-diluted 1× PBS medium with a high sensitivity of 690 mV. Our results demonstrate that the proposed immuno-DG ISFET can be a biosensor platform for practical use in the diagnosis of various diseases.It is important to detect hydrogen peroxide (H2O2) near mitochondrial DNA (mtDNA) because mtDNA is more prone to oxidative attack than nuclear DNA (nDNA). In this study, a mitochondria-targeted fluorescence probe, pep3-NP1, has been designed and synthesized. The probe contains a DNA-binding peptide, a H2O2 fluorescence reporter, and a positively charged red emissive styryl dye to facilitate accumulation in mitochondria. Due to groove binding of the peptide with DNA, the styryl dye of pep3-NP1 intercalated into the bases of DNA, leading to an increase in red fluorescence intensity (centered at 646 nm) and quantum yield. In this case, pep3-NP1 was a turn-on probe for labeling DNA. Subcellular locations of pep3-NP1 and MitoTracker suggested that pep3-NP1 mostly accumulated in the mitochondria of live cells. Namely, as an intracellular DNA marker, pep3-NP1 bound to mtDNA. In the presence of H2O2, pep3-NP1 emitted green fluorescence (centered at 555 nm). Thus, the ratio of green with red fluorescence of pep3-NP1 was suitable to reflect the change of the H2O2 level near mtDNA in living cells. The detecting limit for H2O2 was estimated at 2.9 and 5.0 μM in vitro and in cultured cells, respectively. The development of pep3-NP1 could help in studies to protect mtDNA from oxidative stress.Dental malocclusions (open bite and lateral cross-bite) or functional disturbances (persistent infantile swallowing) in young children do not occur rarely. They are often induced or reinforced by oral habits, such as thumb sucking or bottle feeding. The aim of this case series is to show the effect of a new type of pacifier on the development of open bite, cross-bite or tongue dysfunction. Three cases selected from the case series are reported to describe the effect of early treatment through the pacifier on the above-mentioned malocclusions in the 2nd and 5th years of age. The longitudinal observation after 3, 9 and 24 months of use has shown that the open bite and the posterior cross-bite can be reduced. A shift from an infantile swallowing pattern to a mature one has also been observed. The results are promising the method is simple and possibly cost-reducing. However, the limited number of cases presented does not support the reproducibility of the results. For this reason the reported observations have to be verified by a longitudinal study in a larger population.
Statistical genetics shows that the success of both genetic association studies and genomic prediction methods is positively associated with the heritability of the trait used in the analysis. Identifying highly heritable components of a complex disease can thus enhance genetic studies of the disease. Existing heritable component analysis methods use data from related individuals to compute linearly-combined traits to maximize heritability. Recent advances in acquiring genome-wide markers have enhanced heritability estimation using genotypic data from apparently unrelated individuals, which is referred to as the chip heritability. E6446 Novel statistical models are thus needed to identify disease components (subtypes) with high chip heritability.

The hydrothermal process results in not only the fabrication of TiO2/g-C3N4 heterojunctions, but also the coordination between Ti species and g-C3N4, which are verified by X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). The electrical resistance test confirms that the coordination can improve the electrical conductivity of composites and can make the charge transfer easier. The photoluminescence (PL) and photocurrent measurements exhibit that the hybridization enhances the separation efficiency of photo-induced electrons and holes. As a result, the Ti species modified g-C3N4 photocatalysts exhibit much higher photocatalytic H2 evolution than the simple heterojunction of TiO2/g-C3N4 obtained via a microwave method and the mechanical mixture of TiO2 and g-C3N4 under visible-light irradiation. The coordination mechanism and synthesis route of TiO2/g-C3N4 heterojunctions are proposed.One-electron oxidation of the 1,2,4-diazaphospholide anion [3,5-R2dp](-) by BiCl3 generated several remarkable paddlewheel dibismuthanes [L2(Bi-Bi)L2] (L = η(1),η(1)-3,5-R2dp, R = tBu, iPr, or Ph) with very short Bi-Bi single bond lengths (2.7964(4)-2.8873(3) Å).
