The present study was conducted to provide new insights into the mechanisms that may be responsible for cadmium (Cd)-induced toxicity in zebrafish larvae as well as the role of the trace element zinc (Zn) in reversing Cd harmful effects. For this purpose, zebrafish eggs were exposed to Cd or/and Zn for 96 h. The effects on morphological aspect; mortality rate; Cd, Zn, and metallothionein (MT) levels; oxidative stress biomarkers; as well as molecular expression of some genes involved in Zn metabolism (Zn-MT, ZIP10, and ZnT1) and in antioxidant defense system (Cu/Zn-SOD, CAT and GPx) were examined. Our results showed that Cd toxicity was exerted, initially, by an interference with Zn metabolism. Thus, Cd was able to modify the expression of the corresponding genes so as to ensure its intracellular accumulation at the expense of Zn, causing its depletion. An oxidative stress was then generated, representing the second mode of Cd action which resulted in developmental anomalies and subsequently mortality. Interestingly, significant corrections have been noted following Zn supplementation based, essentially, on its ability to interact with the toxic metal. The increases of Zn bioavailability, the improvement of the oxidative status, as well as changes in Zn transporter expression profile are part of the protection mechanisms. The decrease of Cd-induced MTs after Zn supplement, both at the protein and the mRNA level, suggests that the protection provided by Zn is ensured through mechanisms not involving MT expression but which rather depend on the oxidative status.Concrete is one of the most important materials that are used in the construction industry all around the world. A larger part of the capacity in concrete is generally employed by the coarse aggregate. Due to the tremendous use of coarse aggregate in the construction industry, the material is getting degraded. In order to preserve the natural material, we are in search of an alternate material that can be used in concrete instead of the original one. So in this research work, it has been attempted to study the mechanical behaviour of lightweight concrete when we use waste coconut shell as coarse aggregate inside concrete. To improve the strength of the concrete, we also use the sisal fibres in various proportions ranging between 1 and 5% in accordance to the binder weight. After the mechanical property tests such as the compression test, spilt tensile strength, flexural test, modulus of elasticity test, and impact resistant test were conducted, finally it was concluded that there was increment in the compression strength up to 5%, and tensile strength was increased to 17% and elastic modulus to 7% when the fibre content used was 3%. Thus, with the use of these waste materials, it was found that the concrete's strength gets increased and it leads to the formation of sustainable concrete thus reducing the pollution in the environment.
Preserved ejection fraction heart failure (HFpEF) can be treated by installing a shunt in the interatrial septum, which relieves excess pressure in the left atrium by allowing blood to flow from left to right. This technique has proven effective in clinical trials, but the details of the flow through the shunted heart are not well understood. The current study aims to collect quantitative data on the relationship between pressure and flow rate in such shunts.
An in vitro, shunted double atrium flow phantom was fabricated and used to investigate the relationship between pressure drop and flow across an interatrial shunt. Flow rate was controlled and the resulting pressure drop across the shunt was measured for a variety of flow cases, including steady and pulsatile flow, flow rate waveforms typical of healthy and failing hearts, and low and high heart rates.
The results show a positive relationship between shunt flow rate and pressure drop which is more pronounced in steady flow than in pulsatile flow. Is. Among other things, the results suggest that steady flow approximations to the heart's pulsatile flow should be used with caution and simplified theoretical models do not approximate the flow rate/pressure drop relationship accurately.
The significantly higher incidence of aneurysms in patients with arteriovenous malformations (AVMs) suggests a strong hemodynamic relationship between these lesions. The presence of an AVM alters hemodynamics in proximal vessels by drastically changing the distal resistance, thus affecting intra-aneurysmal flow. This study discusses the challenges associated with patient-specific modeling of aneurysms in the presence of AVMs.
We explore how the presence of a generic distal AVM affects upstream aneurysms by examining the relationship between distal resistance and aneurysmal wall shear stress using physiologically realistic estimates for the influence of the AVM on hemodynamics. Using image-based computational models of aneurysms and surrounding vasculature, aneurysmal wall-shear stress is calculated for a range of distal resistances corresponding to the presence of AVMs of various sizes and compared with a control case representing the absence of an AVM.

Multidrug chemotherapy is recommended for treating pulmonary Mycobacterium avium and Mycobacterium intracellulare disease. Although ethambutol has been demonstrated to inhibit macrolide resistance, the ethambutol dosage is sometimes decreased due to concerns about optic neuropathy. We aimed to assess whether lower ethambutol doses impact treatment outcomes.
Patients treated over 12months between 2016 and 2020 were collected retrospectively. Clinical outcomes, including negative culture conversion, microbiological cure, adverse events, resistance to macrolides, and recurrence, were compared according to daily ethambutol dosage.
Among 146 patients, 42 were treated with ethambutol dosages over 12.5mg/kg/day, and 104 were treated with lower dosages. Negative culture conversion was achieved for 125 patients, and 90 patients achieved microbiological cure. Recurrence was identified in 16 patients who achieved microbiological cure. No macrolide resistance was observed, and no significant difference was observed in the percentage of negative culture conversion (P = 1.00) or microbiological cure (P = 0.67) between the high- and low-dosage ethambutol groups. Sputum smear positivity was associated with a lower adjusted odds ratio (aOR) of negative culture conversion (aOR 0.48, 95% CI 0.29-0.80). A lower aOR of microbiological cure was independently associated with sputum smear positivity (aOR 0.52, 95% CI 0.37-0.74) and with the use of an intermittent regimen (aOR 0.60, 95% CI 0.41-0.87). Daily ethambutol dosage was not identified as a prognostic factor for any of the outcomes. Optic neuropathy was observed in 7.1% of the high-dose ethambutol group and 1.0% of the low-dosage ethambutol group (P = 0.07).

Cisplatin has been widely accepted as an effective chemotherapy drug with various side effects, including nephrotoxicity. The mechanisms of cisplatin-induced acute kidney injury (AKI) are complex, and there are limited renoprotective approaches. Leonurine is the main active compound of a Chinese herb and has recently been reported to have a protective effect on the kidneys. This study aimed to verify the renoprotective effect of leonurine in attenuating cisplatin-induced AKI and explore the potential associated mechanisms.
C57BL/6 mice were divided into four groups (Sham, Cisplatin, Leonurine, and Cisplatin + Leonurine). Mice in the leonurine-treated groups were pretreated with a daily intraperitoneal injection of 25mg/kg leonurine. AKI was induced by injecting cisplatin once intraperitoneally at 20mg/kg body weight. Mice were killed on day 5. Kidney injury was assessed using a serum biochemical and histological assay. Apoptosis was evaluated using a terminal deoxyribonucleotide transferase-mediated dUTP nick-end labeling (TUNEL) staining assay and Western blot.
