The most intriguing finding of all was that TNF-α inhibited osteoblastic differentiation and the Wnt/β-catenin signaling pathway by down-regulating autophagy, and autophagy positively regulated the Wnt/β-catenin pathway and thus influenced osteoblastic differentiation. Our study provides a theoretical basis for autophagy-inducer therapy for the alveolar bone loss caused by periodontitis.The carapace from the Chinese soft-shelled turtle (Pelodiscus sinensis) is used as a traditional Chinese medicine. Acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC) from turtle carapace were isolated and characterized to screen novel collagen material in this study. Yields of 1.0% and 2.8% were obtained for ASC and PSC which contained glycine as the major amino acid and had high imino acid content. Both collagens had maximum ultraviolet absorption peaks of 220 nm. SDS-PAGE revealed that the structure of both collagens was similar, belonging to type I collagen. Relative viscosities of collagens were decreased as the temperature increased. Collagens showed minimum solubility at pH 8 and maximum solubility at a salt concentration of 3%. The denaturation temperature (Td) of PSC was higher whereas the melting temperature was lower than that of ASC. Both ASC and PSC appeared to be spongy like microstructure with fibrillar pores shown by scanning electron microscopy. The results suggest that collagens isolated from turtle carapace has high thermal stability with potential uses as new substitute for mammalian collagen in medicinal, food or biomaterial fields. However, their biological or pharmacological activities are needed to be further studied.
We try to examine the role of transmembrane protein 92 (TMEM92) in the progression of breast carcinoma (BC) and assess its prognostic value. Moreover, the effects of TMEM92 on BC cell phenotypes was explored.
The levels of TMEM92 in BC tissues were evaluated using bioinformatics analysis according to the Oncomine and The Cancer Genome Atlas databases. mRNA levels of TMEM92 in BC cells were measured by qRT-PCR. Kaplan-Meier methods together with log-rank tests were used to conduct survival analysis, and chi-square tests were employed to assess the relationship between TMEM92 levels and clinicopathological parameters. Cox regression analysis was carried out to identify the independent prognosticators. Small interference RNA targeted to TMEM92 and plasmid vectors pcDNA3.1-TMEM92 were respectively used to silence and over-express TMEM92. Protein levels of molecules in this study were tested by western blot. Cell viability, invasiveness and motility of BC cells were determined by cell counting kit 8, clone for to facilitate tumor cells growth, invasiveness and motility by modulating the EMT relative proteins.
TMEM92 had an independent prognostic value for BC patients, and might act as an oncogene to facilitate tumor cells growth, invasiveness and motility by modulating the EMT relative proteins.
We investigated uterine histopathological and ultrastructural changes in female dogs with pyometra induced by Escherichia coli (E. coli) inoculation using progesterone and/or estradiol.
Dogs were ovariectomized and classified into six groups Groups 1-6 corresponding to estradiol treatment followed by progesterone supplementation, progesterone supplementation only, estradiol supplementation only, simultaneous treatment using estradiol and progesterone, similar to Group 1 but with a double dose, and control group, respectively.
Pyometra was successfully induced in Groups 1, 2, 4, and 5, but not in Group 3. An uneven endometrial surface was observed, along with a purulent discharge, bleeding, inflammatory lesions, cystic endometrial hyperplasia (CEH) or cystic endometrial atrophy. This study was conducted to investigate the impact of WJMSCs-derived secretome (WJMSCs-Se) in apoptosis and the growth of HT-29 cells. HT-29 cells treated with 25 or 50 μg/mL WJMSCs-Se for 24 h. Colony formation and MTT test was used to assess the proliferation and cytotoxicity of the HT-29 cells. Annexin V/PI staining was done for the assessment of apoptosis. The mRNA expression of important apoptosis-related genes was also examined. In the WJMSCs-Se-treated HT-29 cells, colony numbers and viability percentages were significantly reduced in a concentration-dependent manner. Apoptotic and necrotic indexes of WJMSCs-Se-treated HT-29 cells considerably enhanced in comparison to the control. The Caspase-9 and -3 activities were significantly increased in the WJMSCs-Se-exposed HT-29 cells. The mRNA expression of Caspase-9, Caspase-3, and Bax/ Bcl-2 ratio was considerably elevated in the WJMSCs-Se-treated HT-29 cells. Caspase-8 activity and expression of the p53 gene were not affected by the WJMSCs-Se. Taken together, we concluded that WJSCs-Se significantly prevented cell growth and enhanced colon cancer cell death in a concentration-dependent manner mainly through the intrinsic apoptotic pathway.The Congjiang Xiang pig is a rare Chinese miniature breed whose epididymal histologic features through the postnatal development are poorly understood. To clarify the histomorphological differences between each region of epididymis during postnatal development, 24 male Congjiang Xiang pigs aged from neonatal (15 d), peri-puberty (30 d), puberty (60 d) to adult (180 d) stages, were examined. Postnatal differentiation of the different regions (I-V) of the epididymis started from birth and continued until maturity that showed regional variations. Developmental progression was disto-proximal. At the neonatal stage, Wolffian duct differentiation starts in the distal region, then ascends to the middle region which forms regions V, IV and III, respectively. A simple lined cuboidal in the epidydimal epithelial, which gradually differentiated into a pseudostratified columnar with stereocilia from neonatal to post-pubertal. After puberty cell rearrangement occurred in the epithelium, differentiation accelerated, and spermatozoon seen in the lumen, especially the lumen of region II. In region III, both halo and apical cells were frequently observed. At the post-pubertal stage, clear cells were frequently observed in Region IV-V, and the epididymal duct was markedly increased in size and fully packed with spermatozoa. The information presented in this study will be helpful for future evaluations of Congjiang Xiang pig fertility. Artemia franciscana is an animal extremophile that uniquely expresses LEA proteins concurrently from Groups 1, 3, and 6. In this study we examine the subcellular localization of AfrLEA6, a Group 6 LEA protein from embryos of A. franciscana. Immunohistochemistry reveals that AfrLEA6 is located in the cytoplasm of diapause embryos and does not co-localize with nuclei or mitochondria. Due to a trace contaminant arising from chitin-based affinity chromatography during AfrLEA6 purification, the primary antiserum displayed affinities for both AfrLEA6 as well as Artemia chitin-binding proteins. This contaminant (fusion protein of intein plus chitin binding domain) co-migrates with AfrLEA6 during SDS-PAGE. Pre-adsorption of the antiserum with dechorionated embryos was required to remove the non-specific fluorescence in the embryonic cuticular membrane. Results of this study are consistent with the apparent importance of distributing multiple types of LEA proteins across many subcellular locations in anhydrobiotic organisms.The ultrastructural characteristics of the mature spermatozoon of Holorchis pycnoporus (Digenea, Lepocreadioidea, Aephnidiogenidae) are described by means of transmission electron microscopy (TEM). Live worms were collected from the digestive tract of the Striped seabream Lithognathus mormyrus (Teleostei, Sparidae), off the Gulf of Gabès at La Chebba (Tunisia). The ultrastructural study reveals that the male gamete of H. pycnoporus is a filiform cell tapered at both extremities and exhibiting the type III of the digenean spermatozoon proposed by Bakhoum et al. (2017a), characterized by the presence of (1) two axonemes with the 9 + '1' pattern of the Trepaxonemata, (2) external ornamentation of the plasma membrane located in a posterior part of the anterior region of the spermatozoon and associated with cortical microtubules, (3) two bundles of parallel cortical microtubules with maximum number located in the middle part of the spermatozoon, and (4) generally two mitochondria. Moreover, H. pycnoporus shares a set of ultrastructural characteristics with the studied Aephnidiogenidae such as (1) two 9+'1' axonemes of different lengths, (2) an anterior electron-dense material, (3) mitochondrion/a, (4) an external ornamentation of the plasma membrane associated with cortical microtubules, and (5) two bundles of parallel cortical microtubules with their maximum number (around 24 microtubules) located in the middle or posterior part of the spermatozoon. In the Aephnidiogenidae, the mature spermatozoon exhibits a similar ultrastructural pattern. Some differences are observed, particularly the location of maximum number of cortical microtubules and the number of mitochondria. The presence of the anterolateral electron-dense material is the major particularity in species belonging to the Lepocreadioidea. This anterior dense material could be a synapomorphy for the superfamily and an ultrastructural argument supporting the monophyletic status of the Lepocreadioidea (Bray and Cribb, 2012).Canine tonsillar squamous cell carcinoma (TSCC) shows a higher metastasis rate than non-tonsillar oral SCC (NTSCC). The mechanisms of metastasis for TSCC have been less studied, because both TSCC and NTSCC cell lines are few. In this study, 6 cloned TSCC (TSCCLN#1-#6), which were from a metastatic lymph node, and 2 cloned NTSCC (oSCC-1 and -4) cell lines, which were from the primary lesion, were established, and their characteristics were evaluated in vitro and in vivo. Results showed that increased expression level of Vimentin in TSCC cell lines and increased expression levels of mesenchymal markers including Vimentin, Snail, and Slug in NTSCC cell lines corelated with the malignant phenotypes such as the cell growth and colony formation abilities in vitro. However, expression levels of mesenchymal markers and in vitro characteristics were unrelated to tumorigenic ability in nude mice. Additionally, the expression levels of E-cadherin and Vimentin were also evaluated by immunohistochemistry using the formalin-fixed paraffin embedded canine oral SCC tissues, and the results show that the expression level of Vimentin in TSCC was higher than in NTSCC.
