The objective of this unprecedented study was to evaluate the effects of the flour from guavira's industrial residue (GF) consumption in rats fed with hypercaloric diet (HD). During 65 days, 50 rats were separated into a control group 1%, 2%, 4% and 8% HD with GF complementation in the diet. The GF chemical composition, phenolic compounds, antioxidant capacity, serum biochemical parameters (glucose, cholesterol, HDL, non-HDL, triglycerides, AST, ALT, and oral glucose tolerance test), fat liver content, and hepatic histomorphology had been characterized. GF is mainly composed of fibres, with phenolic content of 7,391.09 mg AGE/100 g GF and relevant antioxidant capacity (IC50 2.22 and ORAC 155.68 μmol/TE g-1 ). Serum biochemical analysis did not differ statistically (except ALT reduction, p less then .05). Fat liver content was smaller on HD2%GF (p less then .0001). The control group and 1% GF showed greater diffuse microvesicular steatosis compared to the other groups when using hepatic morphological analysis (p less then .05). GF consumption attenuated NAFLD caused by a hypercaloric diet, and this effect may be related to the fibre content and bioactive compounds in GF. © 2020 Blackwell Verlag GmbH.We profiled nine pure clear cell carcinomas of the breast using massively parallel DNA and RNA sequencing (NGS), in situ hybridization (ISH), and immunohistochemistry (IHC). All cases were primary mammary clear cell carcinomas that were diagnosed in female patients (mean age 53.4 years; range 31-69 years). Based on our findings, we conclude that the majority of clear cell carcinomas are ER/PR positive and consequently amenable to anti-ER treatment modalities. A subset of clear cell carcinomas also harbored alterations in PIK3CA/PTEN/AKT pathway, particularly PTEN, indicating a potential benefit of PI3K/Akt/mTOR inhibitors. The status of I-O biomarkers in clear cell carcinomas indicates a limited therapeutic benefit of immune checkpoint inhibitors (against PD-1/PD-L1). © 2020 The Authors. The Breast Journal published by Wiley Periodicals LLC.BACKGROUND The red flour beetle, Tribolium castaneum Herbst (Coleoptera Tenebrionidae), is an important stored-product pest that is distributed worldwide and has developed resistance to many insecticides. Identifying novel and effective alternative insecticides is important for the control of T. castaneum. The volatile compound benzothiazole has been identified as having great acute toxic activity against T. castaneum. However, a comprehensive evaluation of a new insecticide should include both direct toxic effects and sublethal effects. The aim of this study was therefore to evaluate the effects of benzothiazole on the development and reproduction of T. castaneum. RESULTS Exposure of fourth-instar larvae to lethal and sublethal concentrations of benzothiazole (LC10 , LC30 and LC50 ) significantly decreased pupation rates, food intake and growth rates in T. castaneum. Larval duration was significantly reduced by approximately one day in the LC30 and LC50 treatment groups. The LC50 benzothiazole caused a significant decrease in the weight of pupae and adults, fecundity and egg hatchability. Increased and decreased nutrient (carbohydrate and lipid) contents were observed in surviving larvae and pupae, respectively. The LC30 and LC50 treatments caused the downregulation of five growth-positive regulated genes (PI3K, AKT, CyclinE, S6K1 and S6K2) and the upregulation of two growth-negative regulated genes (4EBP and FOXO). CONCLUSION Benzothiazole presented adverse effects on the development and reproduction of T. castaneum, further supporting benzothiazole as a highly active compound in stored-product protection. Flower development also provides an excellent model for understanding how cell division, expansion and differentiation are coordinated during organogenesis. In the model plant Arabidopsis thaliana, floral organogenesis requires AINTEGUMENTA (ANT) and AINTEGUMENTA-LIKE6 (AIL6)/PLETHORA3 (PLT3), two members of the Arabidopsis AINTEGUMENTA-LIKE/PLETHORA (AIL/PLT) transcription factor family. Together, ANT and AIL6/PLT3 regulate aspects of floral organogenesis including floral organ initiation, growth, identity specification, and patterning. Previously, we used RNA-Seq to identify thousands of genes with disrupted expression in ant ail6 mutant flowers, indicating that ANT and AIL6/PLT3 influence a vast transcriptional network. However, the immediate downstream targets of ANT and AIL6/PLT3 in flowers are unknown. To identify direct targets of ANT regulation, we performed a RNA-Seq time course experiment in which we induced ANT activity in transgenic plants bearing an ANT-glucocorticoid receptor fusion construct. In addition, we performed a ChIP-Seq experiment that identified ANT binding sites in developing flowers. These experiments identified 200 potential ANT target genes based on their proximity to ANT binding sites and differential expression in response to ANT. These 200 candidate target genes were involved in functions such as polarity specification, floral organ development, meristem development and auxin signaling. In addition, we identified several genes associated with lateral organ growth that may mediate ANT's role in organ size control. These results reveal new features of the ANT transcriptional network by linking ANT to previously unknown regulatory targets. This article is protected by copyright. All rights reserved.The present study was conducted with privately owned dogs and cats to investigate whether a relationship exists between the dietary AGEs and the urinary excretion of AGEs, as indication of possible effective absorption of those compounds in the intestinal tract of pet carnivores. For this purpose, data were collected from both raw fed and dry processed food (DPF) fed to dogs and cats, through spot urine sampling and questionnaires. Raw pet food (RF, low in AGE diets) was fed as a primary food source to 29 dogs and DPF to 28 dogs. Cats were categorized into 3 groups, which were RF (n = 15), DPF (n = 14) and dry and wet processed pet food (DWF, n = 25). Urinary-free carboxymethyllysine (CML), carboxyethyllysine (CEL) and lysinoalanine (LAL) were analysed using ultrahigh-performance liquid chromatography (UHPLC)-mass spectrometry, and were standardized for variable urine concentration by expressing the AGE concentrations as a ratio to urine creatinine (Ucr) concentration (µg/µmol Ucr). Urinary excretion of CML, CEL and LAL in dogs fed with DPF was 2.03, 2.14 and 3 times higher compared to dogs fed with RF (p less then .005). Similar to the dogs, a significant difference in CMLUcr, CELUcr and LALUcr between the three diet groups was observed in cats (p-overall less then 0.005, ANOVA), in which the RF fed group excreted less AGEs than the other groups. Linear regression coefficients and SE of CMLUcr, CELUcr and LALUcr showed that body weight and neuter status were significantly correlated with CML and CEL excretion, but not to LAL excretion. Our results revealed a significant correlation between dietary AGEs and urinary excretion of free CML, CEL and LAL, and also showed that endogenous formation of these AGEs occurs in both dogs and cats under physiological conditions. © 2020 The Authors. When appropriately applied it simultaneously limits harmful radiation exposures and better utilizes healthcare resources. Nonetheless, in spite of the PECARN CDR, neuroimaging of children for minor head trauma in the United States (as studied between the years 2007 and 2015) has not changed appreciably and remains at approximately 30%. All rights reserved.AIMS OF THE STUDY The ability of Yersinia enterocolitica strains to form biofilms and the capacity of different alkaloids to inhibit biofilm formation were investigated. METHODS AND RESULTS The capacity to form biofilm on polystyrene of 31 Y. enterocolitica strains was evaluated. Biofilm and quorum sensing (QS) inhibition of 17 alkaloids were assayed; furthermore, minimum biofilm inhibitory concentration (MBIC) was determined. The capacity to form biofilms among the examined strains seemed to be a strain-related feature. The best biofilm inhibitors at 100 µmol l-1 were oliverine (1), guatterine (3), liriodenine (4), oliveridine (5) and pachypodanthine (6), which showed biofilm inhibition higher than 87 %. Pachypodanthine (6) was the most effective compound with MBIC value of 12.5 µmol l-1 at sub-inhibitory concentration and also was able to inhibit QS system and reduce yenR expression at this concentration. CONCLUSION This is the first study to demonstrate that oliverine, liriodenine, and pachypodanthine are able to inhibit biofilm formation of Y.
