To describe the course of disability in patients with benign multiple sclerosis-i.e., with an expanded disability status scale score < 3 10years after disease onset-for up to 30years after disease onset. We evaluated the proportion of patients remaining in the benign state on the long term and the factor associated with this favorable outcome and determined the pattern of disability course after the loss of the benign status.

Patients were selected from the ReLSEP, a French population-based registry. We studied the probability (Kaplan-Meier method) and predictors (multivariate Cox model) of remaining < 3 after year 10, and the course of disability after score 3 according to the duration of the benign phase in patients with ≥ 30years of follow-up (graphs of the course of the mean expanded disability status scale scores in subgroups of patients).

2295/3440 patients had benign multiple sclerosis (66.7%). The probability of remaining benign at year 30 was 0.26 (95% CI 0.26-0.32). A young age at disease onset and a good recovery after the first relapse were associated with remaining benign. Graphs illustrate that those who lost their benign status between years 10 and 30 follow a two-stage course. Beyond score 3, disability accumulation is similar in all but lower disability scores at advanced age are associated with longer benign periods.

The clinical outcome of head and neck squamous cell carcinoma (HNSCC) remains poor, partly due to the presence of resistant cancer stem cells (CSCs) which are responsible of recurrences. CSCs have low EGFR expression and, conversely, overexpress the anti-apoptotic Bcl-2 protein, which is involved in resistance to apoptosis and the invasion/migration capacities of tumour cells.

The combination therapy of ABT-199, a Bcl-2 inhibitor, cetuximab an EGFR inhibitor, and radiation using an HNSCC model (SQ20B cell line) and its corresponding CSC subpopulation were evaluated in vitro (2D/3D cell proliferation; invasion/migration and apoptosis using videomicroscopy) and in vivo.

Cetuximab strongly inhibited 2D and 3D cell proliferation, as well as invasion/migration, only in non-CSC-SQ20B cells, whereas ABT-199 selectively inhibited these mechanisms in SQ20B/CSCs. The combination of irradiation + cetuximab + ABT-199 increased the inhibition of the 2D and 3D cell proliferation, invasion/migration, and resistance to apoptosis in both cell sub-populations. In addition, in a nude mouse model with heterotopic tumour xenograft, a treatment combining cetuximab + ABT-199 with fractional irradiation strongly delayed the tumour growth and increased in vivo lifespan without side effects.

Based on the present results, this triple combination therapy may represent a new opportunity for testing in clinical trials, particularly in locally advanced HNSCC.

Retrospective cumulative risk analysis conducted on 178,515 eyes operated under similar conditions in a group of refractive surgery clinics (Clínica Baviera SL) located in a relatively limited geographical area (Spain). A survival analysis was performed and the data were modelled using the Weibull regression to determine the risk over a period of 16 years and to estimate the association of different risk factors sex, age, axial length (AXL) of the eye, intraoperative posterior capsule rents (PCR), and YAG laser capsulotomies. An observational, comparative, retrospective, bi-centric study of 559 patients who had a computer tomography pelvimetry prior to delivery of a term breech presentation was conducted between August 2013 and August 2019. Patients with an attempted vaginal delivery were divided into two groups a group of asymmetric pelvis (AP) when the difference between the lengths of both oblique diameters was ≥ 1cm and a group of symmetric pelvis (SP) when the two oblique diameters differed by < 1cm. The primary outcome was the rate of vaginal delivery. Secondary outcomes were a composite variable of neonatal and maternal morbidity and mortality.

Of the 370 patients who attempted a vaginal breech delivery, 8% (n = 29) had an AP and 92% (n = 341) had a SP. In the AP group, the vaginal delivery rate was higher (93% versus 78%, p = 0.05). link3 There was no statistically significant difference in neonatal (3% versus 1% in the AP and SP groups, respectively, p = 0.4) and maternal (17% versus 23% in the AP and SP groups, respectively, p = 0.5) morbidity and mortality.

Breast cancer (BC) is the most common type of malignant tumor and the most common cause of cancer-related mortality among women. Metabolic reprogramming is considered a hallmark of cancer, and the study of BC metabolism may be the key to the development of new strategies for diagnosis and treatment. In this study, we aimed to explore thepotential metabolites and gene biomarkers for BC through the integration of metabolomics and transcriptomic data, which could further understand BC tumor biology.

Transcriptome dataset GSE139038 was downloaded to explore the differentially expressed genes (DEGs) between BC and normal control (NC) samples. Metabolomics dataset MTBLS326 was downloaded and preprocessed to obtain altered metabolites. Then, the principal component analysis (PCA) and linear models were used to reveal DEGs-metabolites relations. Finally, the pathway enrichment analysis of altered metabolites was performed.

A total of 280 DEGs and eight metabolites were explored between BC and NC samples. The liner module analysis investigated 28 DEGs-metabolites interactions including WASP family member 3 (WASF3)-lactate, ras-related protein Rab-7B (RAB7B)-lactate, and methyltransferase-like 7A (METTL7A)-pyruvate. Finally, pathways analysis showed that these metabolites (such as lactate and pyruvate) were mainly enriched in pathways like disorders of the Krebs cycle.

Combining with the transcriptomic and metabolomics data, we found that lactate, pyruvate, WASF3, RAB7B, and METTL7A might be used as novel biomarkers and potential therapeutic targets for BC. In addition, the disorders of the Krebs cycle pathway might affect the progression of BC.
