The resulting neural 'bauplan' has to account for multiple objectives simultaneously, including computational function, as well as additional factors such as robustness to environmental changes and energetic limitations. Oftentimes these objectives compete, and quantification of the relative impact of individual optimization targets is non-trivial. Pareto optimality offers a theoretical framework to decipher objectives and trade-offs between them. We, therefore, highlight Pareto theory as a useful tool for the analysis of neurobiological systems from biophysically detailed cells to large-scale network structures and behavior. The Pareto approach can help to assess optimality, identify relevant objectives and their respective impact, and formulate testable hypotheses. This study used a contingent valuation method to estimate residents' WTP for improved solid waste management in Hawassa city. The improvement in solid waste management includes the relocation of the current landfill and the switch from open donkey carts to covered tractors with a waste compactor for solid waste transportation. A method of iterative bidding was used to ask residents about their WTP, and ordinal logistic regression was used for data analysis. The result of the study indicated that the estimated average WTP of residents was Ethiopian Birr (ETB) 26.57 ($ 0.62) per month. Besides, the study findings showed that waste minimization practices such as reuse, recycle, waste separation and making compost from waste were uncommon at the household level in Hawassa city. The policy implications of the findings are that the Municipality of Hawassa should consider the residents' willingness for improved solid waste management service and adopt the solid waste management improvement project to prevent further public health risks and environmental impacts. It is generally accepted that land use and land management practices impact climate change through sequestration of carbon in soils, but modulation of surface energy budget can also be important. Using Landsat data to characterize cropland albedos in Canada's three prairie soil zones, this study estimates the atmospheric carbon equivalent drawdown of albedo radiative forcing for three management practices 1) moving from conventional tillage to no-till, 2) eliminating summer fallow in crop rotations, and 3) growing crops with higher albedos. In a 50-year time horizon, conversion from conventional tillage to no-till results in a total equivalent atmospheric CO2 (CO2-eq) drawdown of 1.0-1.5 kg m-2, and conversion from summer fallow to crops results in CO2-eq drawdown of 1.1-2.4 kg m-2. Conversion of summer fallow to crops results in different magnitudes of CO2-eq drawdown depending on specific crops. Lentils, peas, and canola have relatively higher albedo than that of spring wheat and flax; hence, a larger magnitude of CO2-eq drawdown results when they replace summer fallow in the rotation. For the management changes from 1990 to 2019 for the whole Canadian Prairies, albedo changes induced a CO2-eq drawdown of about 179.3 ± 20.9 Tg due to increased area of no-till, and 101.6 ± 9.5 Tg due to reduced area under fallow. The study shows that the magnitudes of CO2-eq drawdown due to albedo change are comparable to that due to soil carbon sequestration. Therefore, it is important to account for cropland albedo changes in assessing the potential of agricultural management practices to mitigate climate change. The introduction of invasive crayfish has led to a decline of many European native species of crayfish across their range. In this study, novel duplex assays for all crayfish occurring in Switzerland were developed. We aimed to identify the distribution of the seven species using a traditional trap surveillance method as well by collecting water samples to detect eDNA by species-specific quantitative real-time PCR. We reveal our overall experience in finding optimal field and laboratory techniques to discover the distribution and abundance of native and invasive species in order to enhance knowledge of early invasive species invasion and highlight important pockets of populations where native species remain, for implementation of conservation strategies. Using eDNA, important populations of native noble and white-clawed crayfish were revealed in multiple waters across various cantons. The successful identification of native and invasive crayfish species in Switzerland using eDNA can be applied to future nationwide projects. This method which has the ability to detect all species simultaneously across an entire country, will allow an improvement in freshwater crayfish conservation management. The classical lake internal phosphorus (P) pollution control using P-inactivating materials is typically carried out by reducing the release of soluble P from sediments to overlying water; however, particulate P loading through sediment resuspension could also cause internal P pollution for algae breeding. Therefore, based on lanthanum modified bentonite clay (Phoslock®) and drinking water treatment residues (DWTR), the effect of P inactivating materials on sediment resuspension and settlement were comprehensively investigated to assess the variations in particulate P pollution from sediment. Results showed that both materials could effectively control soluble P pollution from sediment, while both had limited effect on the supplement of particulate P to overlying water. The reason may be that hydrodynamic disturbance was the key factor regulating sediment resuspension and settlement. The disturbance induced the resuspension of different sized sediments, especially less then 8 μm fractions, while increasing ds essential to develop appropriate methods to control lake internal P pollution.A rapid upsurge in urban and industrial developments leads to increased generations of solid wastes. The most accepted technique of waste discarding around the world is landfilling. Leaching chemicals from municipal dumping grounds can pollute the groundwater source and the surrounding environment without appropriate precautionary measures. Bentonite is a low-cost constituent used as a liner material in landfills due to its low permeability, high sealing ability, high specific surface area, and the ability to hold up the impurity migration through adsorption. However, leachate interaction with bentonite may alter its properties and reduce its usefulness as a barrier material in the long term. Also, bentonite having different chemical and mineralogical compositions will behave differently due to the leachate interaction. Therefore, it is necessary to compare the performance of various bentonites in the presence of leachates. In the present investigation, two Indian bentonites of different mineralogical composi8.6 and 41.1 times increase at a void ratio of 1.2) values when infused with fly ash, sewage sludge and paper mill leachates, respectively. The study also showed that the fly ash leachate interaction causes a higher variation in bentonite behaviour than sewage sludge and paper mill leachates. The study's findings would prove beneficial to design engineers for selecting bentonite types for landfill liners.High protein diet (HPD) is an affordable and positive approach in prevention and treatment of many diseases. It is believed that transcriptional regulation is responsible for adaptation after HPD feeding and Kruppel-like factor 15 (KLF15), a zinc finger transcription factor that has been proved to perform transcriptional regulation over amino acid, lipid and glucose metabolism, is known to be involved at least in part in this HPD response. To gain more insight into molecular mechanisms by which HPD controls expressions of genes involved in amino acid metabolism in the liver, we performed RNA-seq analysis of mice fed HPD for a short period (3 days). Compared to a low protein diet, HPD feeding significantly increased hepatic expressions of enzymes involved in the breakdown of all the 20 amino acids. Moreover, using KLF15 knockout mice and in vivo Ad-luc analytical system, we were able to identify Cth (cystathionine gamma-lyase) as a new target gene of KLF15 transcription as well as Ast (aspartate aminotransferase) as an example of KLF15-independent gene despite its remarkable responsiveness to HPD. These findings provide us with a clue to elucidate the entire transcriptional regulatory mechanisms of amino acid metabolic pathways. The ASCENT trial reports impressive results with a median overall survival (OS) increased from 6.7 months to 12.1 months with sacituzumab govitecan over single-agent chemotherapy, in metastatic triple negative breast cancer (TNBC) patients in second and subsequent line of therapy. We described design features in the ASCENT trial casting doubt on the extrapolation of the reported results to real world patients. First, the open-label design may exaggerate the effect of the experimental arm. Second, the choice of progression-free-survival (PFS) as a primary endpoint, debatable in metastatic TNBC, can lead to biases early stopping rules may exaggerate efficacy results and informative censoring can bias PFS results interpretation. Third, the control arm was not a complete "physician's choice" it was restricted, preventing from using effective agents in this setting, and leading to a substandard control arm. Fourth and lastly, dose reduction and supportive care recommendations for the experimental drug were different between the trial protocol and the FDA labels, and favored the experimental arm as compared with the control arm. In conclusion, we described four design features in the ASCENT trial having the potential to favor the experimental arm or to penalize the control arm. It thus remains uncertain in which extent the reported outcomes will translate in the real world. Efforts should be made to avoid trial biases that will eventually prevent to conclude about their true impact in patients when applied broadly.Herein, we aimed to elucidate the molecular and cellular mechanism in which ubiquitin-specific protease 8 (USP8) is implicated in liver cancer progression via TRAF6-mediated signal. USP8 induces the deubiquitination of TRAF6, TAB2, TAK1, p62, and BECN1, which are pivotal roles for NF-κB activation and autophagy induction. Notably, the LIHC patient with low USP8 mRNA expression showed markedly shorter survival time, whereas there was no significant difference in the other 18-human cancers. Importantly, the TCGA data analysis on LIHC and transcriptome analysis on the USP8 knockout (USP8KO) SK-HEP-1 cells revealed a significant correlation between USP8 and TRAF6, TAB2, TAK1, p62, and BECN1, and enhanced NF-κB-dependent and autophagy-related cancer progression/metastasis-related genes in response to LPS stimulation. Furthermore, USP8KO SK-HEP-1 cells showed an increase in cancer migration and invasion by TLR4 stimulation, and a marked increase of tumorigenicity and metastasis in xenografted NSG mice. Pimicotinib nmr The results demonstrate that USP8 is negatively implicated in the LIHC progression through the regulation of TRAF6-mediated signal for the activation of NF-κB activation and autophagy induction.
