Between mountain pines and the reference species, 35 candidate genes for altitude-dependent selection were identified, including such encoding proteins responsible for photosynthesis, photorespiration and cell redox homeostasis, regulation of transcription, and mRNA processing. In comparison between two mountain pines, 75 outlier SNPs were found in proteins involved mainly in the gene expression and metabolism.Superficial amorphization and re-crystallization of silicon in orientation after irradiation by femtosecond laser pulses (790 nm, 30 fs) are studied using optical imaging and transmission electron microscopy. Spectroscopic imaging ellipsometry (SIE) allows fast data acquisition at multiple wavelengths and provides experimental data for calculating nanometric amorphous layer thickness profiles with micrometric lateral resolution based on a thin-film layer model. For a radially Gaussian laser beam and at moderate peak fluences above the melting and below the ablation thresholds, laterally parabolic amorphous layer profiles with maximum thicknesses of several tens of nanometers were quantitatively attained. The accuracy of the calculations is verified experimentally by high-resolution transmission electron microscopy (HRTEM) and energy dispersive X-ray spectroscopy (STEM-EDX). Along with topographic information obtained by atomic force microscopy (AFM), a comprehensive picture of the superficial re-solidification of silicon after local melting by femtosecond laser pulses is drawn. In genomics, perceived and personal utility have been proposed as constructs of value that include the subjective meanings and uses of genetic testing. Precisely what constitutes these constructs of utility and how they vary by stakeholder perspective remains unresolved. To advance methods for measuring the value of genetic testing in child health, we conducted a scoping review of the literature to characterize utility from the perspective of parents/caregivers. Peer reviewed literature that included empiric findings from parents/caregivers who received genetic test results for an index child and was written in English from 2016-2020 was included. Identified concepts of utility were coded according to Kohler's construct of personal utility. Of 2142 abstracts screened, 33 met inclusion criteria. Studies reflected a range of genetic test types; the majority of testing was pursued for children with developmental or neurodevelopmental concerns. Coding resulted in 15 elements of utility that mapped to Kohler's four domains of personal utility (affective, cognitive, behavioural and social) and one additional medical management domain. An adapted construct of utility for parents/caregivers may enable specific and standardized strategies for researchers to use to generate evidence of the post-test value of genetic testing. In turn, this will contribute to emerging methods for health technology assessment and policy decision making for genomics in child health. An amperometric trimethylamine N-oxide (TMAO) biosensor is reported, where TMAO reductase (TorA) and glucose oxidase (GOD) and catalase (Cat) were immobilized on the electrode surface, enabling measurements of mediated enzymatic TMAO reduction at low potential under ambient air conditions. The oxygen anti-interference membrane composed of GOD, Cat and polyvinyl alcohol (PVA) hydrogel, together with glucose concentration, was optimized until the O2 reduction current of a Clark-type electrode was completely suppressed for at least 3 h. For the preparation of the TMAO biosensor, Escherichia coli TorA was purified under anaerobic conditions and immobilized on the surface of a carbon electrode and covered by the optimized O2 scavenging membrane. Selleck IU1 The TMAO sensor operates at a potential of -0.8 V vs. Ag/AgCl (1 M KCl), where the reduction of methylviologen (MV) is recorded. The sensor signal depends linearly on TMAO concentrations between 2 µM and 15 mM, with a sensitivity of 2.75 ± 1.7 µA/mM. The developed biosensor is characterized by a response time of about 33 s and an operational stability over 3 weeks. Furthermore, measurements of TMAO concentration were performed in 10% human serum, where the lowest detectable concentration is of 10 µM TMAO.Tan spot, caused by the fungus Pyrenophoratritici-repentis (Ptr), is a severe foliar disease of wheat (Triticumaestivum L.). Improving genetic resistance is a durable strategy to reduce Ptr-related losses. Here, we dissected Ptr-infection's genetic basis in 372 European wheat varieties via single sequence repeats (SSR) in addition to 35k and 90k single nucleotide polymorphism (SNP) marker platforms. In our phenotypic data analyses, Ptr infection showed a significant genotypic variance and a significant negative correlation with plant height. Genome-wide association studies revealed a highly quantitative nature of Ptr infection and identified two quantitative trait loci (QTL), viz., QTs.ipk-7A and QTs.ipk-7B, which imparted 21.23 and 5.84% of the genotypic variance, respectively. Besides, the Rht-D1 gene showed a strong allelic influence on the resistance scores. Due to the complex genetic nature of the Ptr infection, the potential of genome-wide prediction (GP) was assessed via three different genetic models on individual and combined marker platforms. The GP results indicated that the marker density and marker platforms do not considerably impact prediction accuracy (~40-42%) and that higher-order epistatic interactions may not be highly pervasive. Our results provide a further understanding of Ptr-infection's genetic nature, serve as a resource for marker-assisted breeding, and highlight the potential of genome-wide selection for improved Ptr resistance.A novel tunable Erbium-doped fiber ring laser (EDFRL) with a cascaded-chirped long-period fiber grating (C-CLPG) as a wavelength selection filter is proposed from the viewpoint of the sensor use, in which a variable optical attenuator (VOA) is employed as an intracavity loss modulator to change the oscillation wavelength region so that the resultant tuning wavelength range is widened. In the demonstrative experiment for temperature measurements, oscillation over the wavelength range of 12.85 nm (1557.62~1570.47 nm), which is more than three times range of the previously presented laser and is equivalent to 64 °C in terms of temperature change, was achieved, while a single-wavelength oscillation was maintained. In addition, a practical technique for realizing a temperature measurement by combining with the VOA control is also discussed. The decoherence in coherent lidar becomes serious with the increase in distance. A small laser spot can suppress the decoherence of the echo light from noncooperation targets. However, it is very difficult to keep a small light spot over a long distance. In this paper, a pulsed coherent lidar with high sensitivity at the few-photon level was demonstrated. A phase plate was used to modulate the wavefront of the laser to achieve 100 m focusing which reduced the decoherence effect. Based on coherent detection and time-of-flight (TOF) measurements, long-distance laser ranging and imaging on all days was realized. A signal classification and superposition method was used to extract the echo signal submerged in noise. The system was experimentally demonstrated by ranging different noncooperation targets within 105.0 m. link2 The measurement rate was 10 k/s, and the measurement uncertainty was 1.48 cm. In addition, laser imaging was realized at ~50.0 m. The system was simple and portable as well as eye safe, and it may offer new application possibilities in automated vehicle lidar. The purpose of this study was to examine the acute effect of foam rolling and vibration foam rolling on drop jump performance. The optimal time interval between warm-up using foam rolling or vibration foam rolling and drop jump performance was identified. This study included 16 male NCAA Division I college volleyball athletes. Three interventions were performed in a randomized order the foam rolling exercise (FRE), vibration foam rolling exercise (VFRE), and static rest (control). The drop jump was performed before interventions, as well as 2 and 5 min after interventions. The FRE exhibited higher values for drop jump height (DJH) (p = 0.001; η2 = 0.382; statistical power = 0.964) and mean power generation at the hip joint (p = 0.006; η2 = 0.277; statistical power = 0.857) at 2 min compared with before intervention but not at 5 min (p > 0.05). However, the VFRE showed no significant changes in DJH (p > 0.05), and found that hippower was decreased at 5 min (p = 0.027; η2 = 0.214; statistical power = 0.680). The FRE completed in 2 min before rapid single action competition (sprint, long jump, triple jump, etc.) could increase sports performance. The purpose of the work is an experimental analysis of the behavior of reinforced concrete beams with a new, patented system of truss-shaped reinforcement. link3 Experimental tests of reinforced concrete beams with conventional reinforcement and with truss-shaped, mass equivalent reinforcement, with two different values of longitudinal reinforcement ratio, were carried out. The testing results of the load-carrying capacity and displacements of beams are presented. The cracking and failure mechanism of beams with a new truss-shaped reinforcement system was also analyzed. The test results for conventionally reinforced beams and with truss-shaped reinforcement were compared. The test results show that the use of the truss reinforcement has an influence on increasing the load-carrying capacity of beams. The amount of this increase depends on the total longitudinal reinforcement ratio and reaches as much as 95% for beams with a low reinforcement ratio and 12% for beams with a higher reinforcement ratio. Based on the investigation of the cracking mechanism, it can be concluded that the failure of the beams with transverse truss-shaped reinforcement occurs with a greater number of smaller cracks, which are more evenly distributed along the length of the cracking zone, and have a shorter range over the cross-section depth, which results in their smaller opening widths. The comparative analysis shows the effectiveness of the proposed reinforcement system, justifying the high potential possibilities of its use for the reinforcement of concrete structural elements.Although syndecan-1 (SDC1) is known to be dysregulated in various cancer types, its implication in tumorigenesis is poorly understood. Its effect may be detrimental or protective depending on the type of cancer. Our previous data suggest that SDC1 is protective against hepatocarcinogenesis. To further verify this notion, human SDC1 transgenic (hSDC1+/+) mice were generated that expressed hSDC1 specifically in the liver under the control of the albumin promoter. Hepatocarcinogenesis was induced by a single dose of diethylnitrosamine (DEN) at an age of 15 days after birth, which resulted in tumors without cirrhosis in wild-type and hSDC1+/+ mice. At the experimental endpoint, livers were examined macroscopically and histologically, as well as by immunohistochemistry, Western blot, receptor tyrosine kinase array, phosphoprotein array, and proteomic analysis. Liver-specific overexpression of hSDC1 resulted in an approximately six month delay in tumor formation via the promotion of SDC1 shedding, downregulation of lipid metabolism, inhibition of the mTOR and the β-catenin pathways, and activation of the Foxo1 and p53 transcription factors that lead to the upregulation of the cell cycle inhibitors p21 and p27.
