Five pairs of equine cadaver distal forelimbs were collected from a slaughterhouse. Sagittal sections (medial, middle and lateral) were obtained and processed with Haematoxylin Eosin Safran, Unna's Orcein, and Picrosirius red stains. Histological assessment revealed that the T-ligament was covered by the surrounding synovia of the distal interphalangeal joint, the digital flexor tendon sheath and the navicular bursa. Its collagen content was lower (30.01%±10.15) than that of the collateral sesamoidean ligament (89.48%±5.8; P = .0008) and the middle phalanx (85.72%±3.67; P = .0008). Under polarized light microscopy, it showed a slight heterogeneous pattern of birefringence, with angle-related changes. Elastic fibres were more numerous (21.76%±8.72) than in the collateral sesamoidean ligament (0.28%±0.45), or deep digital flexor tendon (0.04%±0.02); and were more densely packed. Mean cell count was higher for the T-ligament than for other tissues (P = .0007). Blood vessels were identified in the T-ligament and were penetrating the deep digital flexor tendon (8/10 limbs, 5/5 horses). In conclusion, the T-ligament looked like a vinculum for the deep digital flexor tendon, with a central elastic core, surrounding loose connective tissue and blood vessels. It is not a ligament. Its clinical relevance still needs to be determined.Conventional transdermal drug patches have been on the market since 1997 but their applicability for drug delivery is limited currently only nearly two dozen of molecules have been approved by the regulatory authorities for transdermal administration and have reached the market. The possibilities for drug delivery via the skin can be improved and expanded by using microneedle patch technologies. However, most microneedle patches focus on the delivery of low amounts of drugs that are generally very potent due to the small dimensions of the microneedle systems. In this study nanoporous microneedle arrays (npMNAs) were combined with a liquid drug reservoir. The parameters that influence the diffusion of memantine from the drug reservoir through the npMNAs in an acceptor solution were investigated. Based on these results a model was developed to predict the diffusion of low-molecular-weight drugs as a function of npMNA properties (i.e., backplate thickness and surface area) and reservoir properties (i.e., volume and drug concentration). This generated an in silico model to predict the release of low-molecular-weight drug from a drug reservoir through a microneedle array into receptor solution, showed a good correlation with the delivery of memantine in a preclinical minipig study. The drug release rates by the npMNAs can be tuned and allow for both zero and first order release kinetics. Summarizing, this work shows that the npMNA technology is a versatile drug delivery system. The npMNAs can be combined with a (seamlessly connected) external drug reservoir and this integrated drug delivery system can be used to deliver at least 9 mg of memantine over 72 h in a preclinical minipig study.The objectives of this study were to assess the ideal volume of Copan FecalSwab™ (FS) preserved stool sample to use with the BD MAX™ Enteric Bacterial Panel and the Extended Enteric Bacterial Panel (BDM GIP) and to compare the performance of FS to the recommended Meridian Para-Pak Cary-Blair medium (PP) for the BDM GIP. Three different input volumes (10, 25, and 50 μL) of stool inoculated with American Type Culture Collection strains representing the targets detected by BDM GIP were tested. Additionally, 144 unpreserved stool samples submitted for gastrointestinal (GI) testing were transferred to PP and FS media and tested by the BDM GIP using 10 μL of PP and 50 μL of FS media. A 100% agreement was observed between PP and FS results. The performance of 50 μL of FS stool preserved sample was equivalent to 10 μL of traditional Cary-Blair PP preserved specimens for GI pathogens detection using the BDM GIP.Objective Some recent studies suggest that maternal prenatal stress (MPS) increases allergic diseases in the children. However, knowledge on this issue in Asian children are lacking. We investigated the association between MPS and the risks of wheeze and asthma in Japanese infants aged 16-24 months. Methods The present subjects were 763 Japanese mother-child pairs. The first, second, and third surveys based on self-administered questionnaires were performed during pregnancy, between 2 and 9 months postpartum, and from 16 to 24 months postpartum, respectively. Data on MPS was obtained in the first survey, using the Stress Inventory (SI), which constructs 12 specific behavioral patterns as response styles to stressors. Data on wheeze and asthma was obtained in the third survey, where wheeze was based on the International Study of Asthma and Allergies in Childhood criteria and asthma was based on doctors' diagnosis. Results There were 169 infants with wheeze (22.1%) and 33 infants with asthma (4.3%), at the time of the third survey. Multiple logistic regression analyses found that a maternal behavioral pattern characterized by chronic irritation and anger was associated with the risk of childhood asthma (adjusted odds ratio [OR] = 1.56, 95% confidence interval [CI] 1.11 to 2.22), but not wheeze (adjusted OR = 1.02, 95%CI 0.88 to 1.19), while there was no appreciable association between the other SI scales and the risk of childhood wheeze or asthma. Conclusions The results partly supported the hypothesis that MPS might increase the risk of asthma in their infants in Japanese.
