The initial clinical manifestations and abdominal imaging findings of neonates with necrotising enterocolitis (NEC) and food protein-induced enterocolitis syndrome (FPIES) are sometimes similar; however, their prognosis and therapies are different. We aimed to evaluate the utility of interleukin (IL)-27 as a differentiation marker between NEC and highly suspected early onset (HSEO)-FPIES.

All samples used in this study were obtained from the neonatal diagnosis centre of Children's Hospital of Chongqing Medical University. In the case-control study, neonates with NEC (n=13), HSEO-FPIES (n=9), and jaundice (control, n=8) were enroled to determine the serum IL-27 levels using commercial enzyme-linked immunosorbent assay (ELISA) kits. In the validation cohort study, the NEC (n=87), HSEO-FPIES (n=62), and jaundice (control, n=54) groups were included to analyse the diagnostic efficiency of IL-27 for discriminating between NEC and HSEO-FPIES using a receiver operating characteristic (ROC) curve.

In the case-control study, IL-27 levels were higher in the NEC group than in the HSEO-FPIES group (p=0·005). In the cohort study, the area under the ROC curve (AUC) of IL-27 for differentiating NEC from HSEO-FPIES was 0·878, which was higher than the AUCs of IL-6 (0·761), C-reactive protein (0·800), white blood cell count (0·637), neutrophils (0·765), lymphocytes (0·782), neutrophil to lymphocyte ratio (0·781), and platelet count (0·729).

Metabolic reprogramming plays an essential role on lymphoma progression. Dysregulation of glutamine metabolism is implicated in natural-killer T-cell lymphoma (NKTCL) and tumor cell response to asparaginase-based anti-metabolic treatment.

To understand the metabolomic alterations and determine the potential therapeutic target of asparaginase, we assessed metabolomic profile using liquid chromatography-mass spectrometry in serum samples of 36 NKTCL patients, and integrated targeted metabolic analysis and RNA sequencing in tumor samples of 102 NKTCL patients. The biological function of solute carrier family 1 member 1 (SLC1A1) on metabolic flux, lymphoma cell growth, and drug sensitivity was further examined in vitro in NK-lymphoma cell line NK-92 and SNK-6, and in vivo in zebrafish xenograft models.

Schizophrenia (SCZ) is a severe psychiatric disorder that affects approximately 0.75% of the global population. Both genetic and environmental factors contribute to development of SCZ. SCZ tends to run in family while both genetic and environmental factor contribute to its etiology. Much evidence suggested that alterations in DNA methylations occurred in SCZ patients.

To investigate potential inheritable pattern of DNA methylation in SCZ family, we performed a genome-wide analysis of DNA methylation of peripheral blood samples from 106 Chinese SCZ family trios. Genome-wide DNA methylations were quantified by Agilent 1×244k Human Methylation Microarray.

In this study, we proposed a loci inheritance frequency model that allows characterization of differential methylated regions as SCZ biomarkers. Based on this model, 112 hypermethylated and 125 hypomethylated regions were identified. Additionally, 121 hypermethylated and 139 hypomethylated genes were annotated. The results of functional enrichment analysis indicated that multiple differentially methylated genes (DMGs) involved in Notch/HH/Wnt signaling, MAPK signaling, GPCR signaling, immune response signaling. Notably, a number of hypomethylated genes were significantly enriched in cerebral cortex and functionally enriched in nervous system development.

Our findings not only validated previously discovered risk genes of SCZ but also identified novel candidate DMGs in SCZ. These results may further the understanding of altered DNA methylations in SCZ.

Synovial inflammation is associated with pain severity in patients with knee osteoarthritis (OA). The aim here was to determine in a population with knee OA, whether synovial tissue from areas associated with pain exhibited different synovial fibroblast subsets, compared to synovial tissue from sites not associated with pain. A further aim was to compare differences between early and end-stage disease synovial fibroblast subsets.

Patients with early knee OA (n=29) and end-stage knee OA (n=22) were recruited. Patient reported pain was recorded by questionnaire and using an anatomical knee pain map. Proton density fat suppressed MRI axial and sagittal sequences were analysed and scored for synovitis. Synovial tissue was obtained from the medial and lateral parapatellar and suprapatellar sites. Fibroblast single cell RNA sequencing was performed using Chromium 10X and analysed using Seurat. Transcriptomes were functionally characterised using Ingenuity Pathway Analysis and the effect of fibroblast secretome n patients.

Since few in the field are likely to insist that these judgments can be made without reference to social values, environmental science can serve as fertile ground for exploring the ethical, social, and political terrain at the frontier of the science and values discourse.In the present contribution, the capability of isotopic ratio mass spectrometry (IRMS) for saffron authentication and detection of four common plant-derived adulterants (marigold flower, safflower, rubia, and saffron style) was investigated. For this purpose, 62 authentic saffron samples were analyzed by elemental analyzer-IRMS (EA-IRMS) and gas chromatography-combustion-IRMS (GC-C-IRMS). In this regard, EA-IRMS and GC-C-IRMS isotope fingerprints of carbon-13 and nitrogen-15 isotopes of saffron components were provided and then analyzed by chemometric methods. CX5461 Principal component analysis (PCA) showed two different behaviors regarding two main regions. Then, a representative saffron sample was provided to study adulteration. On this matter, binary mixtures of saffron and adulterants were prepared at five different weight percentages (5%, 10%, 15%, 25%, and 35%) and analyzed by EA-IRMS and GC-C-IRMS. Data-driven soft independent modeling of class analogy (DD-SIMCA) was used to model authentic saffron samples and find a boundary between authentic and adulterated samples with a sensitivity of 100% by GC-C-IRMS. After that, discriminant models of linear discriminant analysis (LDA), quadratic discriminant analysis (QDA), and partial least squares-discriminant analysis (PLS-DA) were tested to find the best discrimination line and also detection of the lowest level of adulterants. Among different models, the QDA model outperformed other methods and showed the ability to predict adulterants at 5% w/w level with 100% accuracy and precision. Finally, the developed QDA model was successfully used to discriminate a set of mixed samples of saffron and four adulterants as well as some commercial samples.For the first time, widespread aqueous biphasic polymer-salt extraction system was applied for selective separation of Pd(II) and Pt(IV) from chloride technological solutions by countercurrent chromatography using a rotating coiled column (an analog of centrifugal extractor). The principle novelty of the proposed approach is the possibility of multistage extraction process and the needlessness to add any specific reagents for targeted metal binding. The results showed that the use of poly(ethylene glycol)-1500 (PEG-1500) as a stationary phase allows 96-100% of both Pd(II) and Pt(IV) to be recovered from chloride solutions containing copper and nickel. To obtain individual solutions of targeted metals it is enough to change the composition of the salt-rich phase for stripping stage, in particular the salt concentration and pH value. The final purity of the targeted metal fractions obtained after their extraction from model technological solutions is ≥ 99.9%. The components of the aqueous biphasic system are recyclable, non-toxic, available, and widespread in laboratory and technology practice. A scheme of the multistage separation of platinum metals using a rotating coiled column is proposed.Understanding how Marine Protected Areas (MPAs) improve conservation outcomes across anthropogenic pressures can improve the benefits derived from them. Effects of protection for coral reefs in the western and central Indian Ocean were assessed using size-spectra analysis of fish and the relationships of trophic group biomass with human population density. Length-spectra relationships quantifying the relative abundance of small and large fish (slope) and overall productivity of the system (intercept) showed inconsistent patterns with MPA protection. The results suggest that both the slopes and intercepts were significantly higher in highly and well-protected MPAs. This indicates that effective MPAs are more productive and support higher abundances of smaller fish, relative to moderately protected MPAs. Trophic group biomass spanning piscivores and herbivores, decreased with increasing human density implying restoration of fish functional structure is needed. This would require addressing fisher needs and supporting effective MPA management to secure ecosystem benefits for coastal communities.The evaluation of anthropogenic pressures in marine environments commonly involves sediments, water and marine biota. However, the evaluation of the quality of the beach sediment and sediments from shallow areas are scarce compared to sediments from deeper areas in environmental assessment studies. In this study polycyclic aromatic hydrocarbons and linear alkylbenzenes were assessed in sediments from shallow areas of the São Sebastião channel, southeastern Brazil. The channel is part of the Marine Protected Area of the Northern Coast of São Paulo State besides holds the largest petroleum terminal in South America further than three sewage outfalls. Despite the concentrations of these contaminants in the sediments were relatively low compared to those found in the sediments from deeper areas of the channel, suggesting low toxicity of the beach sediments, the contamination reflected the pattern of the sediments of the São Sebastião channel.
