The ATP-binding cassette transporter A1 (ABCA1) is a membrane-bound exporter protein involved in regulating serum HDL level by exporting cholesterol and phospholipids to load up in lipid-poor ApoA-I and ApoE, which allows the formation of nascent HDL. Mutations in the ABCA1 gene, when presents in both alleles, disrupt the canonical function of ABCA1, which associates with many disorders related to lipid transport. Although many studies have reported the phenotypic effects of a large number of ABCA1 variants, the pathological effect of non-synonymous polymorphisms (nsSNPs) in ABCA1 remains elusive. Therefore, aiming at exploring the structural and functional consequences of nsSNPs in ABCA1, in this study, we employed an integrated computational approach consisting of nine well-known in silico tools to identify damaging SNPs and molecular dynamics (MD) simulation to get insights into the magnitudes of the damaging effects. In silico tools revealed four nsSNPs as being most deleterious, where the two SNPs (G1050V and S1067C) are identified as the highly conserved and functional disrupting mutations located in the NBD1 domain. MD simulation suggested that both SNPs, G1050V and S1067C, changed the overall structural flexibility and dynamics of NBD1, and induced substantial alteration in the structural organization of ATP binding site. Taken together, these findings direct future studies to get more insights into the role of these variants in the loss of the ABCA1 function.This study investigates the effects of axial ultrasonic vibration on the microstructure evolution, residual stresses distribution and fatigue fracture behaviour of a 7N01-T4 joint, and demonstrates that ultrasonic vibration can significantly promote the flow of plasticised metals, expand the stirred zone (SZ) width and refine the grain size. The longitudinal residual stresses of the joints are dominant, and the peak longitudinal residual stresses of the thermo-mechanically affected zone (TMAZ) on the advancing side (AS) (TMAZ-AS) in the ultrasonic-assisted friction stir welding (UAFSW) joint are 31.5 MPa lower than those in the friction stir welding (FSW) joint. Compared to that of FSW joints, the fatigue strength of UAFSW joints increases by 20 MPa at 107 cycles (stress ratio of R = 0.1). At high-stress levels, crack initiation occurs at the TMAZ-AS, and is mainly attributed to high residual stresses and second-phase particles. At low-stress levels, fatigue cracks are likely to initiate in the transition zone (TZ).Isatin (indole-2, 3-dione) is a non-peptide endogenous bioregulator exhibiting a wide spectrum of biological activity, realized in the cell via interactions with numerous isatin-binding proteins, their complexes, and (sub) interactomes. There is increasing evidence that isatin may be involved in the regulation of complex formations by modulating the affinity of the interacting protein partners. Recently, using Surface Plasmon Resonance (SPR) analysis, we have found that isatin in a concentration dependent manner increased interaction between two human mitochondrial proteins, ferrochelatase (FECH), and adrenodoxine reductase (ADR). In this study, we have investigated the affinity-enhancing effect of isatin on the FECH/ADR interaction. The SPR analysis has shown that FECH forms not only homodimers, but also FECH/ADR heterodimers. The affinity-enhancing effect of isatin on the FECH/ADR interaction was highly specific and was not reproduced by structural analogues of isatin. Bioinformatic analysis performed using three dimensional (3D) models of the interacting proteins and in silico molecular docking revealed the most probable mechanism involving FECH/isatin/ADR ternary complex formation. In this complex, isatin is targeted to the interface of interacting FECH and ADR monomers, forming hydrogen bonds with both FECH and ADR. This is a new regulatory mechanism by which isatin can modulate protein-protein interactions (PPI).Two experiments were conducted in mixed hardwood-conifer forests in the northeastern United States to test the effects of cleaning surfactant and non-surfactant treated multiple-funnel traps used to catch bark and woodboring beetles. #link# Large amounts of pollen and other debris often form a crust on the interior of traps (personal observations). Such surface deposits may provide footholds for beetles to escape capture in traps. In one experiment, we tested cleaned surfactant and non-surfactant traps against non-cleaned surfactant and non-surfactant traps. In a second experiment, we tested field cleaning of modified multiple-funnel traps as an alternative to substituting clean traps on each collection visit. There was no effect of surfactant treated traps, cleaned or not, on total beetles or individual bark beetle species captured. However, in situ cleaned traps were statistically better at capturing total beetles, total bark beetles, and several bark beetle species than non-cleaned control traps. Surfactant-treated non-modified traps and cleaned modified traps had higher species richness and abundance than other treatments at the site level. The composites were analyzed using X-ray diffraction (XRD), Fourier transforms infrared (FTIR), field emission scanning electron microscopy (FESEM), and transmission electron microscopy (TEM) of the MWCNTs. The composites were characterized for complex permittivity using the coaxial probe at 8-12 GHz range and the transmission/reflection coefficients were measured through micro strip line. The dielectric permittivity measurements carried out at X-band frequency revealed that 22 wt.% MWCNTs nanocomposite display higher dielectric constant (ε') and dielectric loss (ε″) values of 4.23 and 0.65, respectively. A maximum absorption loss of 15.2 dB was obtained for the 22 wt.% nanocomposites at 11.75 GHz. This result suggests that PLA/OPEFB/MWCNTs composites are a promising cheap and lightweight material for the effective microwave absorption in the X-band frequency range.Glioma tumors are one of the most devastating cancer types. The use of oncolytic viruses (OVs) in cancer treatment is one such immune-related therapeutic alternative. OVs have a double oncolytic action by both directly destroying the cancer cells and stimulating a tumor specific immune response to return the ability of tumors to escape the control of the immune system. OVs are one promising alternative to conventional therapies in glioma tumor treatment. Several clinical trials have proven the feasibility of using some viruses to specifically infect tumors, eluding undesired toxic effects in the patient. Here, we revisited the literature to describe the main OVs proposed up to the present moment as therapeutic alternatives in order to destroy glioma cells in vitro and trigger tumor destruction in vivo. Knowing which biological processes take place in specific tissues in a temporal manner will enable directed breeding or synthetic approaches to improve seed quantity and quality. We analyzed a genome-wide transcriptome dataset from embryo, endosperm, endothelium, epidermis, hilum, outer and inner integument and suspensor at the global, heart and cotyledon stages of soybean seed development. The tissue specificity of gene expression was greater than stage specificity, and only three genes were differentially expressed in all seed tissues. Tissues had both unique and shared enriched functional categories of tissue-specifically expressed genes associated with them. Strong spatio-temporal correlation in gene expression was identified using weighted gene co-expression network analysis, with the most co-expression occurring in one seed tissue. Transcription factors with distinct spatiotemporal gene expression programs in each seed tissue were identified as candidate regulators of expression within those tissues. Gene ontology (GO) enrichment of orthogroup clusters revealed the conserved functions and unique roles of orthogroups with similar and contrasting expression patterns in transcript abundance between soybean and Arabidopsis during embryo proper and endosperm development. link3 Key regulators in each seed tissue and hub genes connecting those networks were characterized by constructing gene regulatory networks. Our findings provide an important resource for describing the structure and function of individual soybean seed compartments during early seed development.Betel-quid (BQ) is a commonly used psychoactive substance that renders a specific cardiotoxicity. The purpose of this study was to investigate the association between BQ chewing and premature ventricular contractions (PVC) in patients with cardiopulmonary symptoms, and examine the potential influences of cardiovascular and chronic diseases on such relationship. Participants were 146 patients with cardiopulmonary symptoms who participated in 24-h Holter electrocardiogram monitoring during 2012-2018 in a hospital serving residents that lived in a BQ high prevalence area. Data on substance uses and medical histories for cardiovascular and chronic diseases were collected. Baron-Kenny method was employed to evaluate possible mediation. In patients with cardiopulmonary symptoms, 36.3% were BQ users and 63.7% were nonusers. Adjusting for covariates, BQ chewing was significantly associated with heart failure and diabetes mellitus (adjusted odds ratio (aOR) = 3.4 and 2.3, respectively), but only heart failure was significantly correlated with a low and high level of PVC.
