Streptococcus uberis is one of the leading pathogens causing mastitis worldwide. Identification of S. uberis strains that fail to respond to treatment with antibiotics is essential for better decision making and treatment selection. We demonstrate that the combination of supervised machine learning and matrix-assisted laser desorption ionization/time of flight (MALDI-TOF) mass spectrometry can discriminate strains of S. uberis causing clinical mastitis that are likely to be responsive or unresponsive to treatment. Diagnostics prediction systems trained on 90 individuals from 26 different farms achieved up to 86.2% and 71.5% in terms of accuracy and Cohen's kappa. The performance was further increased by adding metadata (parity, somatic cell count of previous lactation and count of positive mastitis cases) to encoded MALDI-TOF spectra, which increased accuracy and Cohen's kappa to 92.2% and 84.1% respectively. A computational framework integrating protein-protein networks and structural protein information to the machine learning results unveiled the molecular determinants underlying the responsive and unresponsive phenotypes.Wildlife managers design artificial structures, such as bird houses and bat boxes, to provide alternative nesting and roosting sites that aid wildlife conservation. However, artificial structures for wildlife may not be equally efficient at all sites due to varying climate or habitat characteristics influencing thermal properties. For example, bat boxes are a popular measure employed to provide compensatory or supplementary roost sites for bats and educate the public. Yet, bat boxes are often thermally unstable or too cold to fulfill reproductive females needs in northern temperate environments. To help improve the thermodynamics of bat boxes, we tested the effect of (1) three mountings, (2) four orientations, and (3) twelve bat box designs on the internal temperature of bat boxes. We recorded temperatures in bat boxes across a climate gradient at seven sites in Quebec, Canada. Bat boxes mounted on buildings had warmer microclimates at night than those on poles and those facing east warmed sooner in the morning than those facing west or south. Our best new model based on passive solar architecture (Ncube PH1) increased the time in the optimal temperature range (22-40 °C) of targeted species by up to 13% compared to the most commonly used model (Classic 4-chamber) when mounted on a building with an east orientation (other designs presented in the Supplementary Information). Based on bioenergetic models, we estimated that bats saved up to 8% of their daily energy using the Ncube PH1 compared to the Classic 4-chamber when mounted on a building with an east orientation. We demonstrate that the use of energy-saving concepts from architecture can improve the thermal performance of bat boxes and potentially other wildlife structures as well.Findings obtained on objective assessments to evaluate sensitive skin do not correlate well with the symptomatology. We utilized reflectance confocal microscopy (RCM) to compare transepidermal application of zinc in sensitive and non-sensitive skin. Thirty-six subjects participated in this study. They were divided into groups based on lactic acid sting test (LAST)'stinger' and 'non-stinger'; transepidermal water loss (TEWL) measurements; and sensitivity self-assessments 'sensitive' and 'non-sensitive'. RCM images were taken to visualize transepidermal application of topically-applied zinc. The intensity of zinc reflectance at different depths was measured by ImageJ software. Based on LAST scores, the 'stinger' group showed significantly higher reflectance of zinc at 8 µm (stratum corneum) [face (P  less then  0.001), forearm (P = 0.004)], and at 80-104 µm (dermo-epidermal junction layer) on the face. High-TEWL group showed increased zinc reflectance at 8-24 µm (tight junction layer, P  less then  0.001). There were no significant differences amongst subjects self-reporting 'sensitive' and 'non-sensitive' skin. RCM demonstrates that in sensitive skin, there is deeper and higher reflectance of zinc at multiple depths. Structural differences are also visualized. We suggest that RCM is a useful tool for evaluating skin barrier integrity.Retinitis pigmentosa (RP) is a clinically and genetically heterogeneous group of inherited retinal degenerations. The ortholog of Drosophila eyes shut/spacemaker, EYS on chromosome 6q12 is a major genetic cause of recessive RP worldwide, with prevalence of 5 to 30%. In this study, by using targeted NGS, MLPA and Sanger sequencing we uncovered the EYS gene as one of the most common genetic cause of autosomal recessive RP in northern Sweden accounting for at least 16%. The most frequent pathogenic variant was c.8648_8655del that in some patients was identified in cis with c.1155T>A, indicating Finnish ancestry. We also showed that two novel EYS variants, c.2992_2992+6delinsTG and c.3877+1G>A caused exon skipping in human embryonic kidney cells, HEK293T and in retinal pigment epithelium cells, ARPE-19 demonstrating that in vitro minigene assay is a straightforward tool for the analysis of intronic variants. We conclude, that whenever it is possible, functional testing is of great value for classification of intronic EYS variants and the following molecular testing of family members, their genetic counselling, and inclusion of RP patients to future treatment studies.In recent phantom studies low-contrast detectability was shown to be independent from variations in tube voltage in digital radiography (DR) systems. To investigate the transferability to a clinical setting, the lower extremities of human cadavers were exposed at constant detector doses with different tube voltages in a certain range, as proposed in the phantom studies. Three radiologists independently graded different aspects of image quality (IQ) in a comparative analysis. The grades show no correlation between IQ and kV, which means that the readers were not able to recognize a significant IQ difference at different kV. Signal-to-noise and contrast-to-noise ratios showed no significant differences in IQ despite the kV-setting variations. These findings were observed from a limited kV range setting. Higher kV-settings resulted in lowest patient exposure at constant IQ. These results confirm the potential of DR-systems to contribute to standardization of examination protocols comparable to computed tomography. Although this across-stimulus interference effect was unaffected by motivation, it depended on the perceptual overlap between stimuli and was more pronounced for different scene comparisons, than scene-pink noise comparisons. Intriguingly, these findings echo the pattern similarity effects from the neuroimaging literature and pinpoint a mechanism that could aid the regulation of unwanted memories.The non-equilibrium dynamics of the superspin glass state of a dense assembly of ~ 2 nm MnFe2O4 nanoparticles was investigated by means of magnetization, ac susceptibility and Mössbauer spectroscopy measurements and compared to the results of Monte Carlo simulations for a mesoscopic model that includes particles morphology and interparticle interactions. The zero-field cooled (ZFC), thermoremanent (TRM), and isothermal remanent magnetization (IRM) were recorded after specific cooling protocols and compared to those of archetypal spin glasses and their dimensionality. The system is found to display glassy magnetic features. We illustrate in detail, by a number of experiments, the dynamical properties of the low-temperature superspin glass phase. We observe that these glassy features are quite similar to those of atomic spin glasses. Some differences are observed, and interestingly, the non-atomic nature of the superspin glass is also reflected by an observed superspin dimensionality crossover. We show that autocrine actions of IGF2 are not critical for β-cell development, or for the early post-natal wave of β-cell remodelling. Additionally, adult Igf2βKO mice maintain glucose homeostasis when fed a chow diet. However, pregnant Igf2βKO females become hyperglycemic and hyperinsulinemic, and their conceptuses exhibit hyperinsulinemia and placentomegalia. Insulin resistance induced by congenital leptin deficiency also renders Igf2βKO females more hyperglycaemic compared to leptin-deficient controls. Upon high-fat diet feeding, Igf2βKO females are less susceptible to develop insulin resistance. Based on these findings, we conclude that in female mice, autocrine actions of β-cell IGF2 during early development determine their adaptive capacity in adult life.Climate change has not only exacerbated abiotic stress, but has also rendered external conditions more feasible for pests to spread and infest citrus fruit. Citrus leafminer (Phyllocnistis citrella) is a potential pest that directly feeds the newly sprouted leaves and twigs of all three spring, summer and autumn flushes. Increasing temperatures in spring and autumn, leafminer accrued more heat units or developmental degree days to accelerate the biological stages of its life-cycle, thereby increasing the pressure of infestation. Present work was conducted at three different environmental conditions in Sargodha, Toba Tek Singh (TTS) and Vehari districts of the Punjab province, Pakistan; all three experimental sites were located in different agro-ecological zones. More infestation was recorded in all three flushes at TTS and Vehari than in Sargodha. Overall, more damage was observed due to higher temperatures in TTS and Vehari than in Sargodha. After May-June heat stress, spontaneous vegetative growth continued from July to November, produced newly spouted tender leaves for feeding the leafminer larvae, and was seen more in TTS and Vehari.
