Provided that each EICR generates at least one streamer during its lifetime, the streamers will form a 3D network, some parts of which will contain hot channel segments created via the cumulative heating and/or thermal-ionizational instability. These hot channel segments will polarize, interact with each other, and cluster, forming longer conducting structures in the cloud. When the ambient potential difference bridged by such a conducting structure exceeds 3 MV, we assume that the lightning seed, capable of self-sustained bidirectional extension, is formed.Acoustic tweezing of bioparticles has distinct advantages over other manipulation methods such as electrophoresis or magnetophoresis in biotechnological applications. This manipulation method guarantees the viability of the bio-particles during and after the process. In this paper, the effects of sinusoidal boundaries of a microchannel on acoustophoretic manipulation of microparticles are studied. Our results show that while top and bottom walls are vertically actuated at the horizontal half-wave resonance frequency, a large mono-vortex appears, which is never achievable in a rectangular geometry with flat walls and one-dimensional oscillations. The drag force caused by such a vortex in combination with the tilted acoustic radiation force leads to trapping and micromixing of microparticles with diameters larger and smaller than the critical size, respectively. Simulation results in this paper show that efficient particle trapping occurs at the intermediate sinusoidal boundary amplitudes. It is also indicated that in a square-sinusoidal geometry there are two strong vortices, instead of one vortex. Sub-micrometer particles tend to be trapped dramatically faster in such a geometry than in the rectangular-sinusoidal ones.Artificial neural networks (ANN) which include deep learning neural networks (DNN) have problems such as the local minimal problem of Back propagation neural network (BPNN), the unstable problem of Radial basis function neural network (RBFNN) and the limited maximum precision problem of Convolutional neural network (CNN). Performance (training speed, precision, etc.) of BPNN, RBFNN and CNN are expected to be improved. Main works are as follows Firstly, based on existing BPNN and RBFNN, Wavelet neural network (WNN) is implemented in order to get better performance for further improving CNN. WNN adopts the network structure of BPNN in order to get faster training speed. WNN adopts the wavelet function as an activation function, whose form is similar to the radial basis function of RBFNN, in order to solve the local minimum problem. Secondly, WNN-based Convolutional wavelet neural network (CWNN) method is proposed, in which the fully connected layers (FCL) of CNN is replaced by WNN. Thirdly, comparative simulations based on MNIST and CIFAR-10 datasets among the discussed methods of BPNN, RBFNN, CNN and CWNN are implemented and analyzed. Fourthly, the wavelet-based Convolutional Neural Network (WCNN) is proposed, where the wavelet transformation is adopted as the activation function in Convolutional Pool Neural Network (CPNN) of CNN. Fifthly, simulations based on CWNN are implemented and analyzed on the MNIST dataset. Effects are as follows Firstly, WNN can solve the problems of BPNN and RBFNN and have better performance. Secondly, the proposed CWNN can reduce the mean square error and the error rate of CNN, which means CWNN has better maximum precision than CNN. Thirdly, the proposed WCNN can reduce the mean square error and the error rate of CWNN, which means WCNN has better maximum precision than CWNN.The detection of the longitudinal component of a highly focused electromagnetic beam is not a simple task. Although in recent years several methods have been reported in the literature, this measure is still not routinely performed. This paper describes a method that allows us to estimate and visualize the longitudinal component of the field in a relatively simple way. First, we measure the transverse components of the focused field in several planes normal to the optical axis. Then, we determine the complex amplitude of the two transverse field components the phase is obtained using a phase recovery algorithm, while the phase difference between the two components is determined from the Stokes parameters. Finally, the longitudinal component is estimated using the Gauss's theorem. Experimental results show an excellent agreement with theoretical predictions.Monitoring the use of anchored fish aggregating devices (AFADs) is essential for effective fisheries management. However, detecting the use of these devices is a significant challenge for fisheries management in Indonesia. These devices are continually deployed at large scales, due to large numbers of users and high failure rates, increasing the difficulty of monitoring AFADs. To address this challenge, tracking devices were attached to 34 handline fishing vessels in Indonesia over a month period each. Given there are an estimated 10,000-50,000 unlicensed AFADs in operation, Indonesian fishing grounds provided an ideal case study location to evaluate whether we could apply spatial modeling approaches to detect AFAD usage and fish catch success. We performed a spatial cluster analysis on tracking data to identify fishing grounds and determine whether AFADs were in use. Interviews with fishers were undertaken to validate these findings. We detected 139 possible AFADs, of which 72 were positively classified as AFADs. Our approach enabled us to estimate AFAD use and sharing by vessels, predict catches, and infer AFAD lifetimes. Key implications from our study include the potential to estimate AFAD densities and deployment rates, and thus compliance with Indonesia regulations, based on vessel tracking data.The gastric microbiota in Crohn's disease (CD) has not been studied. The purpose of the study was to evaluate differences of stomach microbiota between CD patients and controls. DNA was extracted from gastric mucosal and fluid samples, from 24 CD patients and 19 controls. 16S rRNA gene sequencing identified 1511 operational taxonomic units (OTUs), of which 239 passed the low abundance and low variance filters. All but one CD patients were HP negative. Fifteen bacterial phyla were identified in at least one mucosal or fluid site. Of these, Bacteroidota and Firmicutes accounted for 70% of all phyla. Proteobacteria, Actinobacteriota, and Fusobacteriota combined accounted for 27%. There was significant difference in the relative abundance of Bacteroidota, Proteobacteria, Fusobacteriota, and Campilobacterota between CD patients and controls only in gastric corpus samples. In gastric liquid, there was a significant difference only in Actinobacteriota. Pairwise comparison identified 67 differentially abundant OTUs in at least one site. Of these, 13 were present in more than one comparison, and four differentiating OTUs (Neisseriaceae, Neisseria, Absconditabacteriales, and Microbacteriaceae) were identified at all tested sites. The results reveal significant changes in gastric microbial profiles (beta diversity, phylum, and individual taxa levels) between H. pylori-negative CD patients and controls.A recently developed Phox2aCre mouse line has been shown to capture anterolateral system (ALS) projection neurons. Here, we used this line to test whether Phox2a-positive cells represent a distinct subpopulation among lamina I ALS neurons. We show that virtually all lamina I Phox2a cells can be retrogradely labelled from injections targeted on the lateral parabrachial area (LPb), and that most of those in the cervical cord also belong to the spinothalamic tract. Phox2a cells accounted for ~ 50-60% of the lamina I cells retrogradely labelled from LPb or thalamus. Phox2a was preferentially associated with smaller ALS neurons, and with those showing relatively weak neurokinin 1 receptor expression. The Phox2a cells were also less likely to project to the ipsilateral LPb. Although most Phox2a cells phosphorylated extracellular signal-regulated kinases following noxious heat stimulation, ~ 20% did not, and these were significantly smaller than the activated cells. This suggests that those ALS neurons that respond selectively to skin cooling, which have small cell bodies, may be included among the Phox2a population. Previous studies have defined neurochemical populations among the ALS cells, based on expression of Tac1 or Gpr83. However, we found that the proportions of Phox2a cells that expressed these genes were similar to the proportions reported for all lamina I ALS neurons, suggesting that Phox2a is not differentially expressed among cells belonging to these populations. see more Finally, we used a mouse line that resulted in membrane labelling of the Phox2a cells and showed that they all possess dendritic spines, although at a relatively low density. However, the distribution of the postsynaptic protein Homer revealed that dendritic spines accounted for a minority of the excitatory synapses on these cells. Our results confirm that Phox2a-positive cells in lamina I are ALS neurons, but show that the Phox2aCre line preferentially captures specific types of ALS cells.A new series of nucleosides, moieties, and Schiff bases were synthesized from sulfadimidine. Infrared (IR), 1HNMR, 13C NMR, and mass spectrometry techniques and elemental analysis were employed to elucidate the synthesized compounds. The prepared analogues were purified by different chromatographic techniques (preparative TLC and column chromatography). Molecular docking studies of synthesized compounds 3a, 4b, 6a, and 6e demonstrated the binding mode involved in the active site of DNA gyrase. Finally, all synthesized compounds were tested against selected bacterial strains. The most effective synthesized compounds against S. aureus were 3a, 4d, 4b, 3b, 3c, 4c, and 6f, which exhibited inhibition zones of inhibition of 24.33 ± 1.528, 24.67 ± 0.577, 23.67 ± 0.577, 22.33 ± 1.528, 18.67 ± 1.528 and 19.33 ± 0.577, respectively. Notably, the smallest zones were observed for 4a, 6d, 6e and 6g (6.33 ± 1.528, 11.33 ± 1.528, 11.67 ± 1.528 and 14.66 ± 1.155, respectively). Finally, 6b and 6c gave negative zone values. Kal effects were promising. Further research is required to study the in vivo antibacterial effects of these compounds and determine therapeutic doses.We present a computational case study of X-ray single-particle imaging of hydrated proteins on an example of 2-Nitrogenase-Iron protein covered with water layers of various thickness, using a start-to-end simulation platform and experimental parameters of the SPB/SFX instrument at the European X-ray Free-Electron Laser facility. The simulations identify an optimal thickness of the water layer at which the effective resolution for imaging the hydrated sample becomes significantly higher than for the non-hydrated sample. This effect is lost when the water layer becomes too thick. Even though the detailed results presented pertain to the specific sample studied, the trends which we identify should also hold in a general case. We expect these findings will guide future single-particle imaging experiments using hydrated proteins.
