A. Rich. (tuber cowpea) is an underutilized crop for consuming its tuber and mature seeds. Wild form of V. vexillata is a pan-tropical perennial herbaceous plant which has been used by local people as a food. Wild V. vexillata has also been considered as useful gene(s) source for V. unguiculata (cowpea), since it was reported to have various resistance gene(s) for insects and diseases of cowpea. To exploit the potential of V. vexillata, an SSR-based linkage map of V. vexillata was developed. A total of 874 SSR markers successfully amplified single DNA fragment in V. vexillata among 1,336 SSR markers developed from Vigna angularis (azuki bean), V. unguiculata and Phaseolus vulgaris (common bean). An F2 population of 300 plants derived from a cross between salt resistant (V1) and susceptible (V5) accessions was used for mapping. A genetic linkage map was constructed using 82 polymorphic SSR markers loci, which could be assigned to 11 linkage groups spanning 511.5 cM in length with a mean distance of 7.2 cM between adjacent markers. To develop higher density molecular linkage map and to confirm SSR markers position in a linkage map, RAD markers were developed and a combined SSR and RAD markers linkage map of V. vexillata was constructed. A total of 559 (84 SSR and 475 RAD) markers loci could be assigned to 11 linkage groups spanning 973.9 cM in length with a mean distance of 1.8 cM between adjacent markers. Linkage and genetic position of all SSR markers in an SSR linkage map were confirmed. When an SSR genetic linkage map of V. vexillata was compared with those of V. radiata and V. unguiculata, it was suggested that the structure of V. vexillata chromosome was considerably differentiated. This map is the first SSR and RAD marker-based V. vexillata linkage map which can be used for the mapping of useful traits.Nanotechnology based cancer therapeutics have rapidly advanced towards the solution of many limitations associated with other drug delivery agents such as nonspecific distribution within the body, low water solubility and non-biocompatibility. Carbon nanoparticles have demonstrated unique properties that are useful to combat with these issues, including their properties dependent on size, high stability in different solvents, compatible size for drug delivery and ease of surface modifications. Fluorescent carbon nanoparticles with good water solubility were obtained from a carbohydrate source by acid assisted ultrasonic treatment at 35kHz for 4h. This simple and economical method can be used for large scale production. Electron microscopic, spectroscopic and thermo gravimetric analysis techniques were used to characterize these carbon nanoparticles. Functionalized CNPs were further conjugated with anticancer drug-methotrexate and used as fluorescent nano-carriers. In this research work, we determined the in vitro bioactivity of CNPs-methotrexate conjugates by lactate dehydrogenase assay, cell adhesion assay and sulforhodamine B assay in human lung carcinoma cell line (H157). The CNPs showed promising biocompatibility and CNPs-MTX conjugates demonstrated potent cytotoxic effects and high anticancer activities in human lung cancer cell line.
Obesity is one of the most important link factors to coronary artery disease development mainly due to the pro-inflammatory and pro-thrombotic states favoring atherosclerosis progression. The LLLT acts in the cellular metabolism and it is highly effective to improve inflammation. The same occur in response to different kinds of exercise. However, we have not known the associate effects using LLLT therapies with aerobic plus resistance training as strategy specifically with target at human obesity control and its comorbidities.

Investigate the effects of the LLLT associated with aerobic plus resistance training on cardiometabolic risk factors in obese women.

Women aged 20-40 years (BMI ≥ 30 kg/m(2)), were divided into 2 groups Phototherapy (PHOTO) and Placebo. They were trained aerobic plus resistance exercises (in a concurrent mode), 1h, 3 times/week during 16 weeks. Phototherapy was applied after each exercise session for 16 min, with infrared laser, wavelength 808 nm, continuous output, power 100 mW, d with concurrent exercise (aerobic plus resistance training) potentiates the exercise effects of decreasing the cardiometabolic risk factors in obese woman. These results suggest the LLLT associated with exercises as a new therapeutic tool in the control of obesity and its comorbidities for obese people, targeting to optimize the strategies to control the cardiometabolic risk factors in these populations.The photoactivated adenylyl cyclase TpPAC from the spirochete bacterium Turneriella parva was synthesized and the purified recombinant protein was characterized by biochemical and optical spectroscopic methods. TpPAC consists of a BLUF domain (BLUF = Blue Light sensor Using Flavin) and an adenylyl cyclase homology domain (CHD). A light induced cAMP cyclase activity of ≈ 53.3 nmolmg(-1)min(-1) was measured while in the dark the cyclase activity was approximately a factor of 240 lower. The photo-cycling dynamics of the BLUF domain of TpPAC was studied by absorption spectra, fluorescence quantum distribution, and fluorescence lifetime measurements. The quantum efficiency of BLUF domain signaling state formation was found to be ϕs ≈ 0.59. A three-component exponential recovery of the signaling state to the receptor state was observed with the time constants τrec,1 = 4.8s, τrec,2 = 34.2s, and τrec,3 = 293s at 21.3 °C. An irreversible flavin photo-degradation was observed with quantum yield ϕD ≈ 8.7 × 10(-6).Candida species are major microorganisms isolated in denture stomatitis (DS), an inflammatory process of the mucosa underlying removable dental prostheses, and express a variety of virulence factors that can increase their pathogenicity. The potential of Photodynamic inactivation (PDI) in planktonic culture, biofilms and virulence factors of Candida strains was evaluated. A total of 48 clinical Candida isolates from individuals wearing removable maxillary prostheses with DS were included in the study. The effects of erythrosine (ER, 200 μM) and a green LED (λ 532 ± 10 nm, 237 mW/cm(2) and 42.63 J/cm(2)) in a planktonic culture were evaluated. The effect of the addition of ER at a concentration of 400 μM together with a green LED was evaluated in biofilms. The virulence factors of all of the Candida strains were evaluated before and after the PDI process in cells derived from biofilm and planktonic assays. All of the Candida species were susceptible to ER and green LED. However, the biofilm structures were more resistant to PDI than the planktonic cultures. PDI also promoted slight reductions in most of the virulence factors of C. albicans and some of the Candida tropicalis strains. These results suggest that the addition of PDI is effective for reducing yeasts and may also reduce the virulence of certain Candida species and decrease their pathogenicity.In this work there is a synthesis of new photosensitizers which is based on amide derivatives of chlorin е6 . For the disclosure of an extra ring of the initial compound - pheophorbide a 1, we used primary aliphatic amines with 4-12 carbon atoms in the alkyl chain. The reaction is carried out under mild conditions in chloroform with heating to 40 ºС. The structure of all compounds obtained was confirmed by means of electronic, IR, 1Н-NMR spectroscopy and mass-spectrometry. The photoactivity and the dark toxicity of the compounds 2b-2h were investigated on two cancer cell lines P-388 and K-562. The biological investigations revealed a good photoactivity and low dark toxicity of all compounds 2b-2f. The amide derivatives of chlorin е6 with 6 and 7 carbon atoms in the alkyl part showed the best results in our research. Thus, in this paper we propose a reliable scheme of synthesis of chlorin's photosensitizers which are promising agents for PDT.Cancer is one of the dreadest diseases once diagnosed and has severe impacts on health, social and economic global aspects. The multifunctional effects of silver and gold nanoparticles (Ag and Au NPs) have rendered them to be potent candidates for biomedical applications. The current work presents a comparative study between Au NPs and Ag NPs as possible potent photosensitizers (PS) in photodynamic therapy (PDT). Transmission electron microscopy (TEM) was used to identify and characterize the shape, size, and cellular localization of Au NPs; the absorption properties of Au NPs were determined using ultraviolet-visible spectroscopy (UV-Vis) and zeta potential was used to identify surface charge. Inverted light microscopy (LM), Trypan blue exclusion assay, adenosine triphosphate luminescence (ATP), and lactate dehydrogenase membrane integrity assays (LDH) were used for investigating the photodynamic ability of these nanost NPs with other photosensitizers to synergize its effect.Skin cancer is related to unprotected exposure to sunlight. Despite the broad expansion of sunscreen market, various researches are focused on the inefficiency and danger of sun products. This study was focused on the development of photoprotective nanoparticulate dermal preparations of the antioxidant flavonoid Rutin (RT). Nanostructured lipid carriers (NLCs) were prepared using different types of lipids. Based on particle size (PS), size distribution (PDI) and Zeta potential (Z) as well as rheological properties, NLC containing Plurol® stearique (NLC-P) and Apifil® (NLC-A) were selected and loaded with different concentrations of RT to form the medicated nanocreams. F4 (NLC-A with 2% RT) attained highest occlusive effect, drug encapsulation and release efficiencies as well as sun protective factor (SPF). Different concentrations of TiO2 were added to F4 aiming to ameliorate the sun protective effect. F7 (containing 5% TiO2) attained the highest SPF and area under the UV absorbance curve and had a critical wavelength above 370 nm, which proved its high efficiency as sunscreen. The in-vitro antioxidant effect of F7 was more than two fold that of the standard antioxidant. This study provides a suitable cosmeceutical lipidic colloidal system of Rutin to be employed as a successful photoprotective preparation.A new approach to the study of the effects on human skin of mycosporine-like amino acids (MAAs) and gadusol (Gad) incorporated in polymer gel is proposed in this work. The depth profile and photoprotector effects of Pluronic F127® gels containing each of the natural actives were evaluated by in vivo confocal Raman spectroscopy aiming at the analysis of the biochemical changes on human skin. https://www.selleckchem.com/mTOR.html Hierarchical cluster analysis (HCA) showed that the data corresponding to different depths of the skin, from surface to 4 μm, and from 6 to 16 μm, remained in the same cluster.
