efei.A combined process of anaerobic digestion (UASB), shortcut nitrification-denitrification (A/O), and semi-anoxic co-metabolism (operated by an up-flow semi-anoxic sludge bed; USSB) was constructed for the treatment of old landfill leachate (>10 years). The performance and mechanism of refractory organics degradation by the combined process (UASB-A/O-USSB) were investigated. The results showed that the semi-anoxic co-metabolism contributes 57 % of the totally degraded refractory organics. Specific microorganisms and their corresponding metabolic functions drive the degradation of refractory organics in each unit of the UASB-A/O-USSB process. In detail, organics with simple molecular structures were preferentially degraded by anaerobic digestion and shortcut denitrification, whereas those with complex structures were subsequently degraded in the oxic tanks and USSB reactor by shortcut nitrification and semi-anoxic co-metabolism. The structural equation model showed that the combined process of shortcut nitrification and semi-anoxic co-metabolism had a better effect on the degradation of recalcitrant organics than the single process. These findings provide information on how refractory organics are metabolically degraded in a combined process.Herein, an aptasensor based on target-induced strand displacement (TISD) strategy was developed for sensitive detection of T-2 toxin. Gold nanoparticles@ aminated manganese dioxide (AuNPs@NH2-MnO2) exhibited excellent electrical conductivity and provided more binding sites for aptamer (Apt). In the presence of T-2, the Apt sequence was more inclined to form an Apt-T-2 complex, and the cDNA was displaced from the Apt-cDNA duplex, while the signal tag was released, resulting in a weakened MB signal, differential pulse voltammetry (DPV) was used to record the signal change. Under optimal conditions, the signal response of the constructed electrochemical aptasensor exhibited a good linear relationship with the concentration of T-2. The detection limit was 8.74 × 10-7 ng mL-1over a wide range of concentration from 5 × 10-6 ng mL-1 to 5 ng mL-1. Furthermore, the proposed aptasensor had excellent specificity, good stability and can be well applied to the detection of real samples. It provided a new avenue for the research and development of sensitive aptasensors in food detection and analysis.This study evaluated the effects of supplementing feed with arazyme and dietary carbohydrolases derived from invertebrate gut-associated symbionts on the noxious gas emissions, gut microbiota, and host-microbiome interactions of pigs. Here, 270 and 260 growing pigs were assigned to control and treatment groups, respectively. The tested feed additives contained a mixture of arazyme (2,500,000 Unit/kg) and synergetic enzymes, xylanase (200,000 Unit/kg) and mannanase (200,000 Unit/kg), derived from insect gut-associated symbionts in a 7.511 ratio. The control group was fed a basal diet and the treatment group was fed the basal diet supplemented with 0.1 % enzyme mixture (v/v) for 2 months. Odorous gases were monitored in ventilated air from tested houses. The results demonstrated that supplementing feed with arazyme with dietary carbohydrolases effectively controls noxious gas emissions and improves health and meat quality of pigs.Improving forage productivity with lower greenhouse gas (GHG) emissions from limited grassland has been a hotspot of interest in global agricultural production. In this study, we analyzed the effects of grasses (tall fescue, smooth bromegrass), legume (alfalfa), and alfalfa-grass (alfalfa + smooth bromegrass and alfalfa + tall fescue) mixtures on GHG emissions, net global warming potential (Net GWP), yield-based greenhouse gas intensity (GHGI), soil chemical properties and forage productivity in cultivated grassland in northwest China during 2020-2021. Our results demonstrated that alfalfa-grass mixtures significantly improved forage productivity. The highest total dry matter yield (DMY) during 2020 and 2021 was obtained from alfalfa-tall fescue (11,311 and 13,338 kg ha-1) and alfalfa-smooth bromegrass mixtures (10,781 and 12,467 kg ha-1). The annual cumulative GHG emissions from mixtures were lower than alfalfa monoculture. Alfalfa-grass mixtures significantly reduced GHGI compared with the grass or alfalfa monocultures. Furthermore, results indicated that grass, alfalfa and alfalfa-grass mixtures differentially affected soil chemical properties. Lower soil pH and C/N ratio were recorded in alfalfa monoculture. Alfalfa and mixtures increased soil organic carbon (SOC) and soil total nitrogen (STN) contents. Importantly, alfalfa-grass mixtures are necessary for improving forage productivity and mitigating the GHG emissions in this region. In conclusion, the alfalfa-tall fescue mixture lowered net GWP and GHGI in cultivated grassland while maintaining high forage productivity. These advanced agricultural practices could contribute to the development of climate-sustainable grassland production in China.Ibuprofen is a nonsteroidal anti-inflammatory drug that can be found in freshwater ecosystems. Due to its current presence in aquatic ecosystems, this pharmaceutical has aroused concerns about its impact on aquatic biota. As a result, ibuprofen is the one of the most frequently studied pharmaceuticals. However, most of these studies focus on short-term observations of biomarkers and physiological endpoints. This paper presents the outcomes of whole-life-cycle observations and six-month observations of the population dynamics of Daphnia magna reared under the influence of 1 μg/L, 2 μg/L and 4 μg/L of ibuprofen. Individuals reared under the influence of ibuprofen grew slowly, matured later and lived longer. Moreover, they displayed a higher reproduction rate and carried smaller broods but delivered larger neonates. Ibuprofen in concentrations of 1 μg/L and 2 μg/L had the most significant effect on the above traits. The observed impact of ibuprofen at the individual level did not transfer to population size and dynamics. All the populations represented a typical boom and bust cycle with restricted reproduction during the periods of highest population size. This is the first study to explore the linkage between the life histories of aquatic invertebrates and the actual response of their populations to the occurrence of ibuprofen in the environment. The study emphasizes the need to apply the protocol of whole life-cycle observation in tandem with population scrutiny, since such a protocol can reveal the virtual responses of aquatic biota to the presence of chemicals in the environment.Our understanding of how anthropogenic stressors such as climate change and plastic pollution interact with biodiversity is being widened to include diversity below the species level, i.e., intraspecific variation. The emerging appreciation of the key ecological importance of intraspecific diversity and its potential loss in the Anthropocene, further highlights the need to assess the relative importance of intraspecific versus interspecific diversity. One such issue is whether a species responds as a homogenous whole to plastic pollution. Using manipulative field transplant experiments and laboratory-controlled hydrodynamic simulations, we assessed the relative effects of intraspecific and interspecific diversity on microplastic trapping in coastal biogenic habitats dominated by two key bioengineers, the brown intertidal macroalgae Fucus vesiculosus and F. guiryi. At the individual level, northern morphotypes of F. guiryi trapped more microplastics than southern individuals, and F. vesiculosus trapped more microplastics than F. guiryi. Canopy density varied among species, however, leading to reversed patterns of microplastic accumulation, with F. guiryi canopies accumulating more microplastics than those of F. vesiculosus, while no differences were observed between the canopies of F. guiryi morphotypes. We emphasize the importance of assessing the effects of intraspecific variation which, along with other crucial factors such as canopy density, flow velocity and polymer composition, modulates the extent of microplastic accumulation in coastal biogenic habitats. Our findings indicate that a realistic estimation of plastic accumulation in biogenic habitats requires an understanding of within- and between-species traits at both the individual and population levels.Plastics are one of the ubiquitous and artificial types of substrates for microbial colonization and biofilm development in the aquatic environment. Characterizing plastic-associated biofilms is key to the better understanding of organic material and mineral cycling in the "Plastisphere"-the thin layer of microbial life on plastics. In this study, we propose a new method to extract biofilms from environmental plastics, in order to evaluate the properties of biofilm-derived organic matter through stable carbon (δ13C) and nitrogen (δ15N) isotope signatures and their interactions with radionuclides especially radiocesium (137Cs). The extraction method is simple and cost-effective, requiring only an ultrasonic bath, disposable plastic syringes, and a freeze drier. After ultrasound-assisted separation from the plastics, biofilm samples were successfully collected via a sequence of syringe treatments, with less contamination from plastics and other mineral particles. Effective removal of small microplastics from the experimental suspension was satisfactorily achieved using the method with syringe treatments. Biofilm-derived organic matter samples (14.5-65.4 mg) from four river mouths in Japan showed 137Cs activity concentrations of less then 75 to 820 Bq·kg-1 biofilm (dw), providing evidence that environmental plastics, mediated by developed biofilms, serve as a carrier for 137Cs in the coastal riverine environment. Significant differences in the δ13C and δ15N signatures were also obtained for the biofilms, indicating the different sources, pathways, and development processes of biofilms on plastics. We demonstrate here a straightforward method for extracting biofilms from environmental plastics; the results obtained with this method could provide useful insights into the plastic-associated nutrient cycling in the environment.
