In order to precise the roles of AGAMOUS-LIKE 12, one of the few MADS-box genes preferentially expressed in roots, we placed its cDNA under the control of the double 35S CaMV promoter to produce transgenic walnut tree and Arabidopsis plants. In Juglans sp., transgenic somatic embryos showed significantly higher germination rates but abnormal development of their shoot apex prevented their conversion into plants. In addition, a wide range of developmental abnormalities corresponding to ectopic root-like structures affected the transgenic lines suggesting partial reorientations of the embryonic program toward root differentiation. In Arabidopsis, AtAGL12 overexpression lead to the production of faster growing plants presenting dramatically wider and shorter root phenotypes linked to increased meristematic cell numbers within the root apex. In the upper part of the roots, abnormal cell divisions patterns within the pericycle layer generated large ectopic cell masses that did not prevent plants to grow. Taken together, our results confirm in both species that AGL12 positively regulates root meristem cell division and promotes overall root vascular tissue formation. Genetic engineering of AGL12 expression levels could be useful to modulate root architecture and development.The wine with the designation of origin "Jerez-Xerez-Sherry" is one of the most produced in Spain and with a greater volume of exports abroad. This study analyzes the preferences of Sherry Wine and its different varieties, based on gender and knowledge and interest in the world of wine. Similarly, the influence of the attributes of Sherry Wine on its choice and consumption is investigated. For this purpose, 1502 participants (1407 wine consumers) were recruited; among the consumers, 58.5% are women, and 74.3% have little knowledge of wine (Mean age 22.6; SD = 3.07; range 18-30). Data collection was done through an online survey. The results reveal that among the types of wines from Sherry, the Fino and the Manzanilla are the most chosen. The young people who have the highest consumption of wine are those who have the most prior knowledge of wine. Also, these young people attribute their choice of wine to intrinsic factors, and even women and connoisseurs are more important in this type of dimension. But the importance of the attributes differs according to the types of wines (Fino/Cream-flavor, Palo Cortado/Pedro Ximénez-color, Amontillado/Manzanilla-price and Oloroso-prizes). These findings will allow establishing measures for their promotion, as well as for the design and specific sales strategies for each type of wine.A new welding method, flux bands constricting arc (FBCA) welding, is proposed to compensate for the shortage of insufficient weld width of laser welding T-joints in high steel sandwich panels. The arc behavior (arc burning position, arc shape, arc heat, and arc stability) before and after sticking the flux bands (GMAW and FBCA welding) to the ultra-narrow gap groove was tested. Results indicate that flux bands have solid-wall constricting effect (SWCE) and thermo-compression effect (TCE) on the arc and self-producing slag and gas function in FBCA welding. In ultra-narrow gap groove, the arc burning position climbing up phenomenon (APCP) occurs without flux bands. The SWCE of flux bands on the arc effectively suppresses the APCP because of the insulation of flux bands. In the FBCA welding process, the effective heating area of the arc is increased by at least 5 mm2 compared with that in GMAW. When the groove gap decreases, flux bands not only compress the arc from an inverted bell shape to a rectangular shape, but also make the 660 °C isotherm on the core-plate to increase from 3 mm to 8 mm. In the end, the proportion of unstable arc burning time is reduced by 86.85%, the fluctuation of arc voltage and welding current are also significantly reduced by the flux bands because of their SWCE on the arc.Interferon regulatory factor 7 (IRF7) is known as the master transcription factor of the type I interferon response in mammalian species along with IRF3. Yet birds only have IRF7, while they are missing IRF3, with a smaller repertoire of immune-related genes, which leads to a distinctive immune response in chickens compared to in mammals. In order to understand the functional role of IRF7 in the regulation of the antiviral response against avian influenza virus in chickens, we generated IRF7-/- chicken embryonic fibroblast (DF-1) cell lines and respective controls (IRF7wt) by utilizing the CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9) system. selleck IRF7 knockout resulted in increased viral titers of low pathogenic avian influenza viruses. Further RNA-sequencing performed on H6N2-infected IRF7-/- and IRF7wt cell lines revealed that the deletion of IRF7 resulted in the significant down-regulation of antiviral effectors and the differential expression of genes in the MAPK (mitogen-activated protein kinase) and mTOR (mechanistic target of rapamycin) signaling pathways. Dynamic gene expression profiling of the host response between the wildtype and IRF7 knockout revealed potential signaling pathways involving AP1 (activator protein 1), NF-κB (nuclear factor kappa B) and inflammatory cytokines that may complement chicken IRF7. Our findings in this study provide novel insights that have not been reported previously, and lay a solid foundation for enhancing our understanding of the host antiviral response against the avian influenza virus in chickens.
