GITR is triggered by a GITR ligand or an agonist antibody and activates CD8+ and CD4+ effector T cells, reducing tumor-infiltrating Treg numbers and resulting in activation of immune responses and tumor cell destruction by effector T cells. GITR is an attractive target for immunotherapy, especially in combination therapy with immune checkpoint inhibitors, as is being explored in clinical trials. Using H2L2 transgenic mice encoding the human immunoglobulin variable region and hybridoma technology, we generated a panel of fully human antibodies that showed excellent specific affinity and strong activation of human T cells. After conversion to fully human antibodies and engineering modification, we obtained an anti-GITR antibody hab019e2 with enhanced antitumor activity in a B-hGITR MC38 mouse model compared to Tab9H6V3, an anti-GITR antibody that activates T cells and inhibits Treg suppression from XenoMouse. As a fully human antibody with its posttranslational modification hot spot removed, the hab019e2 antibody exerted more potent therapeutic effects, and may have potential as a novel and developable antibody targeting GITR for follow-up drug studies.Human herpes viruses (HHVs) are among the most common infectious agents detected in the gastrointestinal tract that might be involved in oncogenesis and other gastrointestinal disorders. Although the link between the Epstein-Barr virus (EBV) and gastric cancer (GC) has been established, the role of the viruses in various stomach diseases remains unknown. The frequencies and viral copy number of EBV, cytomegalovirus (CMV), and human herpesvirus 6 (HHV-6) among 50 gastric cancer tumors and 105 chronic gastritis tissues were measured by quantitative real-time PCR. In the tumor specimens and the adjacent normal tissues EBV was found in 60% and 30.9%, CMV in 14% and 4.7%, and HHV-6 in 18%, and 14.2%, respectively. The detection rate of EBV and CMV was found to be significantly higher in tumor tissues relative to the adjacent normal tissues. Also, in chronic gastritis, the frequency of EBV, CMV, and HHV-6 was 19%, 12.3%, and 15.2%, respectively, compared with 16.4%, 1.1%, and 8.2% in their corresponding normal tissues. Here, the CMV frequency was found to be significantly higher in gastritis tissues relative to the adjacent normal tissues. Furthermore, viral load in both gastric cancer and gastritis groups was higher in either tumor or gastritis lesion compared with matched adjacent normal tissue. This study showed a clear association between gastric cancer with both EBV and CMV. Meanwhile, analyses revealed a strong association between the EBV, CMV, and HHV-6 viral loads with gastritis (P = 0.0026, P  less then  0.0001, and P = 0.0405, respectively). Our results suggest that these three viruses might contribute to the induction and development the gastritis and gastric cancer.Uniform magnesium (Mg) plating/stripping under high areal capacity utilization is critical for the practical application of Mg-metal anodes in rechargeable Mg batteries. However, the failure of the Mg-metal anode when cycling under a practical areal capacity (>4 mA h cm-2 ), is of rising concern. The mechanism behind these failures remains controversial. In this work, it is illustrated that the initial plating Mg can be undoubtedly uniform in a wide range of current densities (≤5 mA cm-2 ) and under a practical areal capacity (6 mA h cm-2 ). However, an unusual self-accelerating pit growth is observed in the Mg stripping side under moderate current densities (0.1-1 mA cm-2 ), which severely deteriorates the anode integrity and subsequent Mg plating uniformity, causing failure of the Mg-metal anode or short circuit of the battery. The stripping morphology depends on the applied current density, as non-uniformity versus the current density displays a volcano plot during the stripping process. Through in situ spectroscopy, it is illustrated that this current-dependent behavior is determined by the evolution of chlorine-containing complex ions near the interface. In OVA-induced asthma, administration of specific Lactiplantibacillus was confirmed to induce anti-inflammatory cytokines.
Congenital hyperinsulinism (HI) is the most common cause of persistent hypoglycemia in children. In addition to typical focal or diffuse HI, some cases with diazoxide-unresponsive congenital HI have atypical pancreatic histology termed Localized Islet Nuclear Enlargement (LINE) or mosaic HI, characterized by histologic features similar to diffuse HI, but confined to only a region of pancreas. Our objective was to characterize the phenotype and genotype of children with LINE-HI.

The phenotype and genotype features of 12 children with pancreatic histology consistent with LINE-HI were examined.

We compiled clinical features of 12 children with LINE-HI and performed next-generation sequencing on specimens of pancreas from eight of these children to look for mosaic mutations in genes known to be associated with diazoxide-unresponsive HI (ABCC8, KCNJ11, and GCK).

Children with LINE-HI had lower birth weights and later ages of presentation compared to children with typical focal or diffuse HI. Partial pancreatectomy in LINE-HI cases resulted in euglycemia in 75% of cases; no cases have developed diabetes. Low-level mosaic mutations were identified in the pancreas of six cases with LINE-HI (three in ABCC8, three in GCK). Expression studies confirmed that all novel mutations were pathogenic.

These results indicate that post-zygotic low-level mosaic mutations of known HI genes are responsible for some cases of LINE-HI that lack an identifiable germ-line mutation and that partial pancreatectomy may be curative for these cases.

Once phosphorylated, both can bind the p53 mRNA and enhance its translation; however, both recognize p53 protein and provoke its degradation under normal conditions. HDM2 has been well-recognized as an E3 ubiquitin ligase, whereas it has been reported that even with the high similarity between the RING domains of the two homologs, HDMX does not have the E3 ligase activity. Despite this, HDMX is needed for the proper p53 poly-ubiquitination. Phosphorylation at serine 395 changes the conformation of HDM2, helping to explain the switch in its activity, but no information on HDMX has been published. Here, we study the conformation of HDMX and its phospho-mimetic mutant S403D, investigate its E3 ligase activity and dissect its binding with p53. We show that phospho-mutation does not change the conformation of the protein, but HDMX is indeed an E3 ubiquitin ligase in vitro; however, in vivo, no activity was found. We speculated that HDMX is regulated by induced fit, being able to switch activity accordingly to the specific partner as p53 protein, p53 mRNA or HDM2. Our results aim to contribute to the elucidation of the contribution of the HDMX to p53 regulation.Antioxidant interaction among hydrophilic phytochemicals (caffeic acid, p-coumaric acid) and lipophilic phytochemicals (β-carotene, lycopene) in different mole ratios (n/n, 19, 37, 55, 73, 91) was evaluated. Assays performed were based on the scavenging activity of hydrogen peroxide (H2 O2 ), the inhibition of low-density lipoprotein oxidation (ox-LDL) and DNA damage in vitro, using isobological analysis, synergistic rate (SR), and combination index (CI). Results showed that groups containing higher ratios of hydrophilic phytochemicals exhibited synergism while those containing higher ratios of lipophilic phytochemicals showed antagonism. Meanwhile, groups containing caffeic acid (e.g., caffeic acidβ-carotene, 91) with more hydroxyl groups showed higher synergism (SR = 0.76 ± 0.02, CI = 0.77 ± 0.03) than groups containing p-coumaric acid (e.g., p-coumaric acidβ-carotene, 91, SR = 0.88 ± 0.04, CI = 0.82 ± 0.05) on the scavenging activity of H2 O2 . Groups that contained lycopene (caffeic acid lycopene, 91) with a higher ability of regeneration by phenolic acids showed more significant synergism (SR = 0.70 ± 0.02, CI = 0.79 ± 0.03) than groups containing β-carotene (e.g., caffeic acidβ-carotene, 91, SR = 1.00 ± 0.03, CI = 0.98 ± 0.04) on the inhibition of DNA damage. This study provided a basis for antioxidant interactions among phytochemicals against ox-LDL and DNA damage in vivo. In addition, the choice of appropriate ratios and structures of hydrophilic and lipophilic phytochemicals should be considered in the diet and formulation of functional foods.
