Studies have demonstrated a disparity in sexual minority adolescents and substance use, possibly due to factors contributing to minority stress. The objective of this study is to investigate the association between self-identifying as lesbian, gay, or bisexual and illicit substance use compared to those who identify as heterosexual among high school students in the U.S.
A secondary data analysis of a cross-sectional study was performed using data from the Youth Risk Behavior Surveillance System in 2017. The study included adolescents attending public, Catholic, or private U.S. high schools. Participants with missing data on substance use and sexual orientation were excluded. The final sample size was 12,370. The main outcome variable was illicit substance use, which included marijuana and cocaine, among others. The main exposure variable was students who identified as heterosexual, gay or lesbian, bisexual, and unsure. selleck inhibitor Binary logistic regression analysis was used to calculate unadjusted and adjusted odds ratios and 95% confidence intervals (CIs).
Participants who identified as bisexual had 1.65-fold increased odds (95% CI 1.32-2.05) of ever using illicit substance(s) compared to heterosexuals. Those who identified as "not sure" of their sexual identity had 1.37-fold increased odds of ever using illicit substance(s) compared to heterosexuals (95% CI 1.03-1.83).
The results will provide health professionals with more information about illicit substance use among lesbian, gay, or bisexual identified adolescents. An increase in the understanding of health disparities associated with sexual identity may assist in the development of interventions.

An increase in the understanding of health disparities associated with sexual identity may assist in the development of interventions.The structures of Aspergillus oryzae α-amylase were determined in a tetragonal crystal, having one molecule as asymmetric unit, and a monoclinic crystal with two molecules as asymmetric unit. Both crystal forms were obtained from trace contaminants of an old commercial lipase preparation. Structures were determined and refined to 1.65 Å and 1.43 Å resolution respectively. The latter crystal has a non-crystallographic (NCS) twofold axis within the asymmetric unit. Glycosylation at Asn197 is evident, and in the tetragonal crystal can be seen to include three, partially disordered sugar residues following the initial N-acetyl glucosamine (NAG). Superposition of the tetragonal crystal model on the α-amylases from Bacillus subtilis (PDB1BAG), pig pancreas (PDB3L2L), and barley (PDB1AMY), show a high degree of coincidence, particularly for the (β/α)8-barrel domains, and especially within the active site. Using this structural agreement between amylases, we extrapolated the binding model of a six residue, limit dextrin found in pig pancreas α-amylase to the A. oryzae enzyme model, which predicts substrate interacting amino acid residues.
The inhibition of glucocorticoid (GC) on osteoblastic differentiation of bone marrow stromal stem cells (BMSC) is an important pathway for GC to reduce bone formation. Recent studies implicated an important role of peroxisome proliferator-activated receptor-gamma (PPAR-γ) in GC-mediated cell proliferation and differentiation. Thus, our purpose is to investigate the role of PPAR-γ in regulating rat BMSC (rBMSC) osteoblastic differentiation.
The rBMSC treated with dexamethasone (Dex) was used to construct an in vitro cell model of GC-induced osteoporosis. The expressions of PPAR-γ, RUNX2, ALP, OPN and SFRP5 in cells were detected by RT-qPCR and western blot assays. Osteogenic differentiation of rBMSC was measured by Alizarin Red S (ARS) staining analysis. Lentivirus-delivered shRNA was used to knock down PPAR-γ or SFRP5, and lentivirus-delivered constructs were used to overexpress SFRP5 in rBMSC to verify the effect of PPAR-γ or SFRP5 on cell osteogenic differentiation.
Dex significantly reduced rBMSC osteoblastic differentiation. The expression of PPAR-γ was enhanced in Dex treated rBMSC. PPAR-γ down-regulation improved Dex inhibition of rBMSC osteogenic differentiation. Moreover, PPAR-γ knockdown promoted protein levels of RUNX2, ALP, OPN and Dex-decreased rBMSC osteogenic differentiation. The expression of SFRP5 was reduced while Wnt and β-catenin were increased in PPAR-γ knockdown and Dex treated rBMSC. Moreover, the up-regulation of SFRP5 reversed the osteogenic differentiation of rBMSC induced by PPAR-γ knockdown.
These data indicated that in GC-induced osteoporosis, PPAR-γ/SFRP5 affects osteogenic differentiation by regulating the Wnt/β-catenin signaling pathway.

We sequenced the IL-8 gene of 109 black and 32 white South African (SA) individuals and conducted detailed characterization of gene variation and haplotype structure. IL-8 production in phytohaemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMCs) of a subset (black N = 22; white N = 32) of these individuals was measured using ELISA. Select variants were genotyped for additional black individuals (N = 141), and data from the 1000 Genomes Project were used for haplotype analysis and comparative purposes. In white individuals, the -251A>T SNP formed part of a prevalent six-variant haplotype [haplotype frequency (HF) 61%], Hap-1C, involving the following variants -251A>T; +394T>G (rs2227307); +780C>T (rs2227306); +1240->A (rs2227541); +1635C>T (rs2227543) and +2770A>T (rs2227543).
