Flanking markers useful for identifying hybrid necrosis alleles were identified by fine mapping Ne1 and Ne2 and the distribution of the two necrosis genes was investigated in Chinese elite wheat varieties. Hybrid necrosis of wheat is caused by the interaction of two dominant complementary genes Ne1 and Ne2 present separately in normal parents and is regarded as a barrier to gene transfer in wheat breeding. However, the necrosis alleles still occur at a high frequency in modern wheat varieties. In this study, we constructed two high-density genetic maps of Ne1 and Ne2 in winter wheat. In these cultivars, Ne1 was found to be located in a span interval of 0.50 centimorgan (cM) on chromosome 5BL delimited by markers Nwu_5B_4137 and Nwu_5B_5114, while Ne2 co-segregated with markers Lseq102 and TC67744 on 2BS. Statistical analysis confirmed that the dosage effect of Ne1 and Ne2 also existed in moderate and severe hybrid necrosis systems, and the symptoms of necrosis can also be affected by the genetic background.th Ne1Ne1ne2ne2 and ne1ne1Ne2Ne2, respectively and introduced modern cultivars should directly affect the frequencies of necrosis genes in modern Chinese cultivars (lines), especially that of Ne2. Taking investigations in spring wheat together, we proposed that hybrid necrosis alleles could positively affect breeding owing to their linked excellent genes such as Lr13. Additionally, based on the pedigrees and hybridization tests, we speculated that the Ne1 and Ne2 in winter wheat may directly originate from wild emmer and introduced cultivars or hexaploid triticale, respectively.The present study was the first prospective cohort evaluated the efficacy and safety of different doses of calcitriol in XLH children. The results suggested that a dose of 40 ng/kg/day calcitriol, compared with 20 ng/kg/day, was more effective in relieving the rickets, with similar safety outcomes. Further investigations were expected to set more dose groups.
Dose recommended for calcitriol in X-linked hypophosphatemia (XLH) varies in different studies. Therefore, we aimed to compare the efficacy as well as the safety of 20ng/kg/d and 40ng/kg/d calcitriol in Chinese XLH pediatrics population.
A 2-year, randomized, open-label, prospective study recruited 68 XLH children, which were randomized to receive either 40ng/kg/day or 20ng/kg/day calcitriol. Efficacy endpoints were the total Thacher ricket severity score (RSS) change from baseline to month 12 and 24, the difference in serum TALP level, fasting serum phosphate level, body height Z-score, and frequency of dental abscess. Safety assessments were done using renal ultrasound nephrocalcinosis grades (0-4), fasting serum and 24h urine calcium level, and the occurrence of hyperparathyroidism.
The decrease in the total RSS from baseline was more significant in the high-dose group at 12 (difference 0.87, p = 0.049) and 24month (difference 1.23, p = 0.011). The serum TALP level was significantly lower in the high-dose group at 6months. Pi level, height Z-score change, frequency of dental abscess and ratio of de novo nephrocalcinosis were comparable. A lower incidence of secondary hyperparathyroidism was seen in the high-dose group (p < 0.0001).
For the first time in this prospective cohort, 40ng/kg/d calcitriol was shown to be the more effective therapy in XLH children than the 20ng/kg/d. Moreover, 40ng/kg/d calcitriol was not associated with increasing adverse events.

Eating disorders have been associated with decreased bone mineral density and increased fracture risk, but the association with fall injuries without fracture has not previously been investigated. Furthermore, fracture risk in men with eating disorders has been insufficiently studied.
In the present study, 8867 patients (9.4% men) with a diagnosed eating disorders and 88670 age-, sex-, and county-matched controls were investigated.
The mean (standard deviation) age of the patients and controls was 41.6 (13.7) years and the follow-up time 9.6 (5.2, 14.4) years (median, interquartile range) for patients and 10.1 (5.5, 14.2) years for controls. The proportions of injurious falls without fracture (17.3% vs. 9.0%) and of hip fracture (1.6% vs. 0.7%) were substantially greater in patients with an eating disorder than in th women and men with eating disorders, indicating measures to prevent both falls and fractures are important in these patients, regardless of age and sex.We collected the farmland soil and the sediment, carried out sample speciation detection and distribution analyses, explored environmental self-purification on the distribution of Cd and As of agricultural sources. There were significant differences between total Cd and total As and between speciation distribution in the orders of magnitude. Total Cd (mean content was 1.74 mg kg-1) and total As (mean content was 32.31 mg kg-1) in all the samples were higher than the background contents. Cd distribution in the farmland soil was not significantly affected by geographical locations. As cumulative effect in riverside farmland soil was stronger, river irrigation and flooding may input more As, transformation mechanism of As may be exogenous input, binding and passive release. Here, the migration of alkaline constituents and the restoration evaluation with phosphogypsum were discussed by soil column experiments to investigate the alkaline regulation in bauxite residue disposal areas (BRDAs). The pH, free alkali, exchangeable sodium in the top layer (0-25 cm depth) covered with BR and phosphogypsum mixtures were reduced from 10.89 ± 0.02, 285.45 ± 21.15 mmol/kg, 385.63 ± 30.34 mg/kg to 9.00 ± 0.50, 12.50 ± 1.50 mmol/kg, 97.00 ± 10.50 mg/kg. For the sublayers, including depths of 35, 45, 55 cm, these values dropped to 9.86, 10.06, 10.03; 38.23, 86.12, 148.00 mmol/kg; 152.90, 246.00, 305.00 mg/kg, respectively. These results indicated alkaline indicators for phosphogypsum amended BR declined dramatically, and the parameters for sublayers were also decreased due to the migration of alkaline constituents. The physicochemical properties for amended BR could meet the conditions for plant growth. This research provided a reference for alkalinity regulation in BRDAs by phosphogypsum.Urban rivers are suffering from a significant anthropogenic impact. In this study, eight trace metals were investigated in surface water along the Yangtze River in Chongqing, China. The decreasing trend of trace metals was observed in water as Sr > Li > Mo > As > Cu > Ni > Cr > Co. Multivariate statistical analysis identified two source types that accounted for 77.17% of the total variance. As, Co, Mo, and Sr were mainly originated from geological sources, while Cr, Cu, and Ni were influenced by anthropogenic activities. Moreover, the average contents of Cr, Cu, and Ni exhibited a significant increase close to the city compared to the sites that are far away from the city, which was identified by various urban rivers. This study suggested that Cr, Cu, and Ni could be regarded as anthropophile elements, which could be a promising indicator for tracing the urban activities.Long-term exposure to heavy metals from high geological background area may lead to varieties of diseases. Therefore, risk assessment from agricultural products in these areas was crucial to ensure the health of customers. However, the effects of geological background on distributions of heavy metals and their accumulation in plant remain unclear. This study aimed to determine the distributions of mercury (Hg), cadmium (Cd), arsenic (As), lead (Pb), chromium (Cr) and copper (Cu) in 1036 corn and rice samples collected from 9 locations in Guizhou province and to evaluate their health risks. The concentrations of Hg, Cd, As, Pb, Cr and Cu in these two crops were analyzed by inductively coupled plasma-mass spectrometry (ICP-MS), and their health risk were estimated by the target risk quotient approaches. Results showed that the mean concentrations of Hg, Cd, As, Pb, Cr and Cu in corn and rice were 0.01, 0.07, 0.06, 0.27, 0.56 and 1.86 mg/kg which were lower than their respective maximum allowable concentrations (MAC) of 0.02, 0.20, 0.70, 0.20, 0.50 and 10.00 mg/kg except Pb and Cr. The concentrations of Cr and Cu in corn were higher than in rice while Cd, As and Pb in rice were higher than in corn. Moreover, the distributions of Hg, Cd and Cu in corn and rice samples were mainly observed from QDN located at southeast of Guizhou province while Pb, As and Cr were most detected at ZY, QXN and BJ areas, southwest zone. The hazard indices (HIs) values for corn and rice were 0.20 and 2.61. The high HIs (> 1) in rice indicated that the health risk of heavy metals in rice was relatively high and Pb was the major component that attributed to the risk, followed by Cd. These results could provide a reference for the distributions of heavy metals in agricultural products in Guizhou province under crop cultivation conditions, and to provide scientific basis for health risk assessment and ensure food safety.
In kinematically aligned total knee arthroplasty (TKA), it is necessary to infer the pre-arthritic constitutional medial proximal tibial angle (MPTA) in advanced osteoarthritis (OA) of the knee with bone loss. The aim of this study was to investigate whether MPTA at the posterior tibial plateau represents the pre-arthritic constitutional MPTA in anterior cruciate ligament (ACL)-intact, advanced OA knees. It was hypothesized that MPTA at the posterior tibial plateau represents the pre-arthritic constitutional MPTA of ACL-intact, advanced knee OA.
One hundred varus, anterior cruciate ligament (ACL)-intact, advanced OA knees were analysed. The hip-knee-ankle (HKA) angle and MPTA were assessed on computed radiography (CR) and MPTAs at the anterior, middle, and posterior part of the tibial plateau were assessed on computed tomography (CT) images. click here The association between these parameters was also analysed.
CR images showed an HKA angle of 172.4 ± 4.1° and MPTA of 84.3 ± 2.5°. CT images showed different MPTAs in the three regions, ranging from 83.9 ± 2.4° to 85.9 ± 2.8°. The middle MPTA was the lowest at 83.9 ± 2.4°. HKA angle correlated with the middle MPTA (r = 0.3355, 95% confidence interval [CI] 0.1489-0.4991, p = 0.0006) and ΔMPTA (Middle-Posterior) (r = 0.5128, 95% CI 0.3518-0.6443, p < 0.0001).
The MPTA at the posterior tibial plateau represents the pre-arthritic constitutional MPTA in ACL-intact, advanced OA knees.
