Pediatric oncology patients (POP) have a high risk of infections due to impaired immunity. Invasive pneumococcal disease (IPD) is an important cause of severe infection in these patients and it is associated with high mortality. This study aimed to evaluate the incidence and risk factors associated with IPD at a Pediatric Oncology Center in Brazil.

This was a retrospective case-control study. All IPD cases in children with cancer from 2005 through 2016 were reviewed. Each case of IPD was matched with two controls from a cohort of patients matched for year of IPD, age and disease in order to assess risk factors. ICEC0942 The incidence density was calculated as the number of IPD per 100,000 patients-year.

A total of 51 episodes of IPD in 49 patients was identified. All pneumococci were isolated from blood cultures. The median age was five years and 67% were male; mortality rate was 7.8%. The IPD incidence density rate in POP was 311.21 per 100,000 patients-year, significantly higher than the rate in the general pediatric population. Severe neutropenia was the only risk factor associated with IPD, after multivariate conditional logistic regression analysis.

Although pneumococcal disease decreased after the introduction of 10-valent pneumococcal vaccine in the Brazilian national immunization schedule in 2010, there was no decrease in the IPD incidence rate in our cohort. A higher coverage rate of pneumococcal vaccination in children in the general population might be necessary to reduce the incidence rate in this high-risk population.

The stroma is responsible for many features of the cornea, including its strength, refractive power and transparency, so enormous efforts have been put into replicating the corneal stroma in the laboratory to find an alternative to classical corneal transplantation. Unfortunately this has not been yet accomplished due to the extreme difficulty in mimicking the highly complex ultrastructure of the corneal stroma, and none of the obtained substitutes that have been assayed has been able to replicate this complexity yet. In general, they can neither match the mechanical properties nor recreate the local nanoscale organization and thus the transparency and optical properties of a normal cornea. In this context, there is an increasing interest in cellular therapy of the corneal stroma using Induced Pluripotent Stem Cells (iPSCs) or mesenchymal stem cells (MSCs) from either ocular or extraocular sources, as they have proven to be capable of producing new collagen within the host stroma, modulate preexisting scars and enhance transparency by corneal stroma remodeling. Despite some early clinical data is already available, in the current article we will summary the available preclinical evidence about the topic corneal stroma regeneration. Both, in vitro and in vivo experiments in the animal model will be shown.Correct diagnosis of children presenting with developmental delay and intellectual disability remains challenging due to the complex and heterogeneous etiology. High throughput sequencing technologies like exome sequencing have become more commonly available and are significantly improving genetic testing. We present two siblings - a 14-year old male and an 8-year old female patient - with a similar clinical phenotype that was characterized by combined developmental delay primarily affecting speech, mild to moderate intellectual disability, behavioral abnormalities, and autism spectrum disorder, but with no congenital anomalies. The sister showed additional muscular hypotonia and more pronounced dysmorphic features compared to her brother. Both parents had psychiatric disorders and mild to moderate intellectual disability. A common genetic etiology in the siblings was suspected. Metabolic, psychological and neuroradiological examinations were complemented by basic genetic testing including chromosome analysis and array comparative genomics hybridization analysis (CGH), followed by exome sequencing and combined data analysis of the family. Exome sequencing identified two different underlying genetic conditions in the sister, a maternally inherited pathogenic variant c.1661C > T, p.Pro554Leu in SLC6A8 (NM_005629.4) was identified causing cerebral creatine deficiency syndrome 1 (MIM #300352) which was confirmed by MR spectroscopy and treated accordingly. In the brother, a paternally inherited 16p13.11 duplication was identified by exome sequencing and considered to be likely associated with his and possibly his father's phenotype. The 16p13.11 duplication had been previously identified in an array CGH but had not been prioritized due to the lack of segregation in the siblings. In conclusion, we report a case of intra-familial locus heterogeneity of developmental delay in two siblings. We advocate for the need of unbiased and comprehensive genetic testing to provide accurate diagnosis despite locus heterogeneity.Signal transducer and activator of transcription 1 (STAT1) and STAT2 are critical components of type I and type II IFNs signaling. To date, seven STAT family proteins have been identified from mammals. However, the information on STAT genes in teleost fish is still limited. In the present study, two STAT family genes (STAT1a and STAT2) were identified from Japanese eel, Anguilla japonica and designated as AjSTAT1a and AjSTAT2. link2 The open reading frames of AjSTAT1a and AjSTAT2 are 2244 bp and 2421 bp, encoding for polypeptides of 747 aa and 806 aa, respectively. Both AjSTAT1a and AjSTAT2 contain the conserved domains of STAT proteins. Phylogenetic analysis was performed based on the STATs protein sequences, and showed that AjSTAT1a and AjSTAT2 shared the closest relationship with Oncorhynchus mykiss. Quantitative real-time PCR analysis revealed that AjSTAT1a and AjSTAT2 were expressed in most examined tissues, with the highest expression both in blood. Significantly up-regulated transcripts of AjSTAT1a and AjSTAT2 were detected in response to poly IC stimulation, and Edwardsiella tarda induced increase in the expression of AjSTAT1a and AjSTAT2 genes. Subcellular localization analysis showed that in both IFNγ-stimulated and unstimulated EPC cells AjSTAT1a and AjSTAT2 were mainly distributed in the cytoplasm, but few AjSTAT1a was distributed in the nucleus. All these results suggested that AjSTAT1a and AjSTAT2 may be critical for regulating the host innate immune defense against pathogens invasion.With the advent of rapid evolution of oomycete pathogen lineages, the need for sustainable agriculture practices has become the need of the hour. The late blight of tomato caused by Phytopthora infestans, has recently emerged as one such devastating disease in India that led to huge crop losses. Hence, in the present work seed priming with mycogenic selenium nanoparticles (SeNPs) for elicitation of resistance against tomato late blight disease is investigated. It also aims to understand the defense responses triggered by SeNPs at cellular, biochemical and transcriptomic levels. Enhanced plant growth parameters were observed in bioactive SeNPs-primed tomato plants as compared to control plants. SeNPs-primed and pathogen inoculated plants exhibited a significant protection of 72.9 % against late blight disease. The primed plants also recorded a remarkable accumulation of lignin, callose and hydrogen peroxide that serve as the cellular defense over the control plants. Further, an elevated level of lipoxygenase (LOX), phenylalanine lyase (PAL), β-1,3-glucanase (GLU), superoxide dismutase (SOD) corroborated the biochemical defense in primed plants, which was also reflected in the corresponding transcriptome profiling of the genes encoding the enzymes. Thus, the present study represents an orchestrated correlation between resistance and defense responses incited by SeNPs against tomato late blight disease, which can be used as nano-biostimulant fungicide in protecting tomato plants.Selenium-containing compounds have emerged as a potentially promising treatment for viral infections and tumor development and dissemination. Selenium per se is often considered as a toxic element with little or no beneficial effects, but considerable advances have been made in the understanding of the complex biology, chemistry and drug delivery of this element, especially when it is included in bioactive molecules. Here, we summarize and critically discuss recent findings in the field of selenium-based applications for the treatment of cancer and viral infections.In recent years, food fraud is a global issue that has raised wide public concern. Mass spectrometry techniques have a significant advantage of qualitatively and quantitatively analyzing food authenticity, especially for highly processed meat products. In this work, a simple and specific, rapid resolution liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the determination of pork content in processed meat products according to internal standard (ISTD) method. To improve the efficiency of sample preparation, simplified bead-beating and enzymolysis process were investigated. In contrast with different heat-stable and specific porcine-peptides, EPITVSSDQMAK, GGPLTAAYR, HDPSLLPWTASYDPGSAK from Carbonic anhydrase 3 proved to have an excellent quantitative ability, thus obtaining good linear relationship and satisfactory recovery. This method was successfully applied to different types of meat products, thus demonstrating that complex mixtures of pork content can be accurately quantified.Calcium phosphate cements (CPCs) have been widely used during the past decades as biocompatible bone substitution in maxillofacial, oral and orthopedic surgery. CPCs are injectable and are chemically resemblant to the mineral phase of native bone. Nevertheless, their low fracture toughness and high brittleness reduce their clinical applicability to weakly loaded bones. Reinforcement of CPC matrix with polymeric fibers can overcome these mechanical drawbacks and significantly enhance their toughness and strength. Such fiber-reinforced calcium phosphate cements (FRCPCs) have the potential to act as advanced bone substitute in load-bearing anatomical sites. This work achieves integrated experimental and numerical characterization of the mechanical properties of FRCPCs under bending and tensile loading. To this end, a 3-D numerical gradient enhanced damage model combined with a dimensionally-reduced fiber model are employed to develop a computational model for material characterization and to simulate the failure process of fiber-reinforced CPC matrix based on experimental data. In addition, an advanced interfacial constitutive law, derived from micromechanical pull-out tests, is used to represent the interaction between the polymeric fiber and CPC matrix. link3 The presented computational model is successfully validated with the experimental results and offers a firm basis for further investigations on the development of numerical and experimental analysis of fiber-reinforced bone cements.
