Although upconversion photodynamic therapy (PDT) has gained extensive interests in disease treatment, the intracellular migration pathway of upconversion photosensitizers and underlying cell-particle interaction mechanism is still largely unexplored. In this work photoswitchable upconversion nanoparticles (UCNPs) are reported that can release orthogonal emissions excited by two near-infrared lights, i.e., red color of 980-nm and green color of 808-nm light excitation. Taking advantage of the dual-emissive property, a methodology based on Pearson's correlation analysis is proposed to verify the accuracy of upconversion luminescence signals under different excitation lights, which has been previously neglected. Meanwhile, we have designed a near-infrared mediated bioimaging nanoplatform that can generate reactive oxygen species (ROS) using one light and simultaneously track the location of upconversion photosensitizers using another excitation light. Our study not only depicts the migration pathway of upconversion photosensitizers, but also demonstrates the organelle escape of these upconversion nanoparticles via PCI (photochemical internalization) process. It is believed that our results inspire more efficient synergistic therapy by combining PDT with other modalities in a programmable manner.Sulfoglycolysis pathways enable the breakdown of the sulfosugar sulfoquinovose and environmental recycling of its carbon and sulfur content. The prototypical sulfoglycolytic pathway is a variant of the classical Embden-Meyerhof-Parnas (EMP) pathway that results in formation of 2,3-dihydroxypropanesulfonate and was first described in gram-negative Escherichia coli. We used enrichment cultures to discover new sulfoglycolytic bacteria from Australian soil samples. Two gram-positive Arthrobacter spp. were isolated that produced sulfolactate as the metabolic end-product. Genome sequences identified a modified sulfoglycolytic EMP gene cluster, conserved across a range of other Actinobacteria, that retained the core sulfoglycolysis genes encoding metabolic enzymes but featured the replacement of the gene encoding sulfolactaldehyde (SLA) reductase with SLA dehydrogenase, and the absence of sulfoquinovosidase and sulfoquinovose mutarotase genes. Excretion of sulfolactate by these Arthrobacter spp. is consistent with an aerobic saprophytic lifestyle. This work broadens our knowledge of the sulfo-EMP pathway to include soil bacteria.Previous studies have shown that selenium possessed chemotherapeutic effect against multiple malignant cancers, inducing diverse stress responses including apoptosis and autophagy. Selenite was previously shown to induce apoptosis and autophagy in colorectal cancer cells. However, the relationship between selenite-induced apoptosis and autophagy was not fully understood. Our results revealed a pro-survival role of selenite-induced autophagy against apoptosis in colorectal cancer cells. Real-time PCR array of autophagy-related genes showed that GABARAPL-1 was significantly upregulated in colorectal cancer cells, which was confirmed by western blot and immunofluorescence results. Knockdown of GABARAPL-1 significantly inhibited selenite-induced autophagy and enhanced apoptosis. Furthermore, we found that selenite-induced upregulation of GABARAPL-1 was caused by upregulated p-AMPK and FoxO3a level. Their interaction was correlated with involved in regulation of GABARAPL-1. We observed that activation and inhibition of AMPK influenced both autophagy and apoptosis level via FoxO3a/ GABARAPL-1 signaling, implying the pro-survival role of autophagy against apoptosis. Importantly, we corroborated these findings in a colorectal cancer xenograft animal model with immunohistochemistry and western blot results. Collectively, these results show that sodium selenite could induce ROS/AMPK/FoxO3a/GABARAPL-1-mediated autophagy and downregulate apoptosis in both colorectal cancer cells and colon xenograft model. These findings help to explore sodium selenite as a potential anti-cancer drug in clinical practices.Onchocerciasis is a disease transmitted by blackfly vector of the genus Simuliumdamnosum complex having public and socio-economic consequences. The abundance of blackfly is directly linked to fluvial ecosystems in both tropical and non-tropical countries of the world. This study assessed the ecology of blackfly breeding sites in two tributaries of River Ogun located in Olokemeji and Lisa villages, Ogun State. A total of eighteen water samples were collected for physicochemical analysis during high rainfall (May, June and July), low rainfall (August, September and October) and dry season (November and December) with mean rainfall values of 163.17, 88.03 and 8.95 mm, respectively, in both tributaries. Physicochemical parameters such as temperature (oC), electrical conductivity (EC), pH, total solids (TS), dissolved oxygen (DO), biochemical oxygen demand (BOD), total suspended solids (TSS), hardness (H), sulphate (SO4), phosphate (PO4), alkalinity, acidity, nitrate (NO3) and total dissolved solids (TDS) were anaibutary, and water samples were also viewed under an electron microscope for zooplanktons and phytoplanktons. The values of species diversity which were greater than 0.5 indicated that the species were abundant and evenly distributed. There were significant (p  less then  0.05) differences between the tributaries in all the digestive enzymes except amylase content. The regression analysis between TSS, nitrate, sulphate, phosphate, acidity, hardness, TS, pH and Simulium larvae abundance showed a positive correlation, R2 with values of 0.828, 0.994, 0.948, 0.933, 0.986, 0.997, 0.889 and 0.981, respectively. The R2 values between Simulium larvae abundance with phytoplanktons and rainfall have values of 0.868 at p  less then  0.01 and 0.911 at p  less then  0.05.The control principle implies that people should not feel guilt for outcomes beyond their control. Yet, the so-called 'agent and observer puzzles' in philosophy demonstrate that people waver in their commitment to the control principle when reflecting on accidental outcomes. In the context of car accidents involving conventional or autonomous vehicles (AVs), Study 1 established that judgments of responsibility are most strongly associated with expressions of guilt-over and above other negative emotions, such as sadness, remorse or anger. Studies 2 and 3 then confirmed that, while people generally endorse the control principle, and deny that occupants in an AV should feel guilt when involved in an accident, they nevertheless ascribe guilt to those same occupants. Study 3 also uncovered novel implications of the observer puzzle in the legal context Passengers in an AV were seen as more legally liable than either passengers in a conventional vehicle, or even their drivers-especially when participants were prompted to reflect on the passengers' affective experience of guilt. Our findings document an important conflict-in the context of AV accidents-between people's prescriptive reasoning about responsibility and guilt on one hand, and their counter-normative experience of guilt on the other, with apparent implications for liability decisions.Membrane fusion is a key step in enveloped virus infection, releasing the viral genome into the cytoplasm to initiate infection. Bombyx mori nucleopolyhedrovirus (BmNPV) is an enveloped DNA virus that mainly infects silkworms. Information about membrane fusion of BmNPV with host cells is still limited. In this study, BmN cells were pretreated with ??ammonium chloride??, and infection with BmNPV was allowed to occur naturally through endocytosis or artificially through low-pH-induced fusion with the plasma membrane, after which the cells were subjected to RNA-seq. The results indicated that a few endoplasmic reticulum-associated proteins (ERAPs) were among the common upregulated DEGs, including BiP, CRT, and HSP90, and this upregulation was confirmed by q-PCR. Knockdown of BiP, CRT, and HSP90 expression by siRNA resulted in significant inhibition of BmNPV infection. This study suggests that ERAPs may be involved in the BmNPV membrane fusion process during infection.
Sarcomatoid hepatocellular carcinoma (SHCC) is a rare subtype of primary liver malignancies and is still ill-defined and poorly understood. check details Therefore, our study was performed to have a comprehensive evaluation SHCC versus conventional hepatocellular carcinoma (HCC).

A thorough database searching was performed in PubMed, EMBASE and the Cochrane Library. RevMan5.3 and Stata 13.0 software were used for statistical analyses. The primary endpoint of our analysis is the long-term survival and the secondary endpoint is clinical and pathological features.

Four studies with a relative large cohort were finally identified. Compared with patients with pure HCC, patients with SHCC had a significantly worse overall survival (P < 0.00001) and disease-free survival (P < 0.0001). Moreover, a larger tumor size (P = 0.003), a higher incidence of node metastasis (P < 0.00001) and a higher proportion of advanced lesions (P = 0.04) were more frequently detected in patients with SHCC. Higher levels of serum ALT (P = 0.02) and TB (P = 0.005) were detected in patients with HCC rather than SHCC, while serum ALB (P = 0.02) level was relatively higher in patients with SHCC. For other measured outcomes, including concurrent viral hepatitis, liver cirrhosis, liver storage (Child A/B), multifocal tumors, vascular invasion and preoperative AFP level, the results showed no significant difference (P > 0.05).

SHCC has a worse prognosis and exhibits more aggressively than conventional HCC. Future large well-designed studies are demanded for further validation.

Defensive immobility is a common adaptive response, in rodents, elaborated by ventrolateral periaqueductal gray matter (vlPAG) when threat is unavoidable. It is associated with somatosensory and autonomic reactions such as alteration in the sensation of pain and rate of respiration. In this study, defensive immobility was assessed by chemical stimulation of vlPAG with different doses of NMDA (0.1, 0.3, and 0.6 nmol). After elicitation of defensive immobility, antinociceptive and respiratory response tests were also performed. Results revealed that defensive immobility was followed by a decrease in the nociceptive perception. Furthermore, the lowest dose of NMDA induced antinociceptive response without eliciting defensive immobility. During defensive immobility, respiratory responses were also disturbed. Interestingly, respiratory rate was increased and interspersed with prolonged expiratory phase of breathing. These findings suggest that vlPAG integrates three different defensive behavioral responses, contributing to the most effective defensive strategies during threatening situations.Ambient dose equivalent conversion coefficients (ADCRCs) for converting a radiocaesium inventory to ambient dose equivalent rates (air dose rates) depend on the vertical distribution of radiocaesium in soil. To access the validity of ADCRCs, the air dose rate at 1 m above ground and the vertical distribution of radiocaesium in the soil around the Fukushima Daiichi Nuclear Power Station (FDNPS) present between 2011 and 2019 were measured in the current study. ADCRCs were calculated using air dose rates and three different parameters representing the vertical distribution of radiocaesium in soil (1) relaxation mass depth (β), (2) effective relaxation mass depth (βeff) and (3) relaxation mass depth recommended by the International Commission on Radiation Units and Measurements before the FDNPS accident (βICRU). When ADCRCs based on β and βeff were compared to those based on β and βICRU, a positive correlation was found. To confirm the applicability of the ADCRCs based on the three types of β values, radiocaesium inventories were estimated using the air dose rates and ADCRCs, and the obtained results were compared to the radiocaesium inventory calculated using soil sample measurements.
