Sperm DNA damage is a major cause of pre- and post-implantation embryonic loss in humans. However, the factors that control how and when such DNA damage occurs in human sperm are poorly understood.

Here, I review information relating to sperm DNA damage that can be obtained from the sperm chromosome assays described in the existing literature.

The sperm chromosome assays, which consist of interspecific in vitro fertilization or intracytoplasmic sperm injection using murine oocytes and subsequent chromosome analysis, indicate that the proportion of sperm showing DNA damage is initially low and there are larger numbers of sperm with potential membrane and DNA damage that are induced after ejaculation and separation from the seminal plasma. Avasimibe purchase Other assays that directly detect sperm DNA (e.g., TUNEL assays, Comet assays, and acridine orange test) are not able to distinguish and detect the initial and potential DNA damage. Furthermore, the positive values in these direct assays are influenced by the frequency of immotile sperm and amorphous sperm populations.

The findings in the sperm chromosome assays show that further improvements in sperm preparation protocols may result in the reduction of sperm DNA damage, followed by more successful outcomes in infertility treatment.

To examine how differences in trophectoderm biopsy techniques affect the frequency of mosaic embryos and sequencing results.

We examined differences in next-generation sequencing (NGS) analysis results among operators or according to biopsy technique. Additionally, we determined the cut-off for the number of collected cells to predict the occurrence of mosaicism. We collected cells according to the cut-off value and examined whether there was a difference in the NGS analysis results between the pulling and flicking methods.

There was no difference in the NGS analysis results among the operators. Regarding re-biopsy, changes in the mosaic were observed in all specimens. The cut-off value for the number of collected cells was five, and when more than five cells were collected, there was no difference in the NGS analysis results between the two methods.

We demonstrated that if trophectoderm biopsy techniques and NGS are stable, the cell collection location has a greater effect on NGS results than the biopsy technique.

Is oral Vitamin D supplementation able to modify the intrauterine milieu in terms of cytokine/chemokine pattern?

No significant differences were detected in cytokine and chemokine levels in endometrial secretions between patients undergoing ART with or without Vitamin D supplementation.

Cytokines and chemokines secreted into the intrauterine environment are fundamental for the molecular crosstalk between the endometrium and the preimplantation embryo. Whether Vitamin D can regulate these mediators in the endometrial environment is still unclear.

This study was an analysis of a secondary outcome from the Supplementation of Vitamin D and Reproductive Outcomes-SUNDRO-clinical trial, a multicenter randomized double-blinded trial designed to explore the effects of Vitamin D replacement in women with Vitamin D levels below 30 ng/ml undergoing autologous ART cycles. Uterine fluid samples were collected from both patients supplemented with Vitamin D (n

Based on c004233-27.We created the PDX Network (PDXNet) portal (https//portal.pdxnetwork.org/) to centralize access to the National Cancer Institute-funded PDXNet consortium resources, to facilitate collaboration among researchers and to make these data easily available for research. The portal includes sections for resources, analysis results, metrics for PDXNet activities, data processing protocols and training materials for processing PDX data. Currently, the portal contains PDXNet model information and data resources from 334 new models across 33 cancer types. Tissue samples of these models were deposited in the NCI's Patient-Derived Model Repository (PDMR) for public access. These models have 2134 associated sequencing files from 873 samples across 308 patients, which are hosted on the Cancer Genomics Cloud powered by Seven Bridges and the NCI Cancer Data Service for long-term storage and access with dbGaP permissions. The portal includes results from freely available, robust, validated and standardized analysis workflows on PDXNet sequencing files and PDMR data (3857 samples from 629 patients across 85 disease types). The PDXNet portal is continuously updated with new data and is of significant utility to the cancer research community as it provides a centralized location for PDXNet resources, which support multi-agent treatment studies, determination of sensitivity and resistance mechanisms, and preclinical trials.Cell membrane deformation is an important feature that occurs during many physiological processes, and its study has been put to good use to investigate cardiomyocyte function. Several methods have been developed to extract information on cardiomyocyte contractility. However, no existing computational framework has provided, in a single platform, a straightforward approach to acquire, process, and quantify this type of cellular dynamics. For this reason, we develop CONTRACTIONWAVE, high-performance software written in Python programming language that allows the user to process large data image files and obtain contractility parameters by analyzing optical flow from images obtained with videomicroscopy. The software was validated by using neonatal, adult-, and human-induced pluripotent stem-cell-derived cardiomyocytes, treated or not with drugs known to affect contractility. Results presented indicate that CONTRACTIONWAVE is an excellent tool for examining changes to cardiac cellular contractility in animal models of disease and for pharmacological and toxicology screening during drug discovery.Engineered synthetic biomolecular devices that integrate elaborate information processing and precisely regulate living cell behavior have potential in various applications. Although devices that directly regulate key biomolecules constituting inherent biological systems exist, no devices have been developed to control intracellular membrane architecture, contributing to the spatiotemporal functions of these biomolecules. This study developed a synthetic biomolecular device, termed inducible counter mitochondrial morphology (iCMM), to manipulate mitochondrial morphology, an emerging informative property for understanding physiopathological cellular behaviors, on a minute timescale by using a chemically inducible dimerization system. Using iCMM, we determined cellular changes by altering mitochondrial morphology in an unprecedented manner. This approach serves as a platform for developing more sophisticated synthetic biomolecular devices to regulate biological systems by extending manipulation targets from conventional biomolecules to mitochondria. Furthermore, iCMM might serve as a tool for uncovering the biological significance of mitochondrial morphology in various physiopathological cellular processes.Multimodal advances in single-cell sequencing have enabled the simultaneous quantification of cell surface protein expression alongside unbiased transcriptional profiling. Here, we present LinQ-View, a toolkit designed for multimodal single-cell data visualization and analysis. LinQ-View integrates transcriptional and cell surface protein expression profiling data to reveal more accurate cell heterogeneity and proposes a quantitative metric for cluster purity assessment. Through comparison with existing multimodal methods on multiple public CITE-seq datasets, we demonstrate that LinQ-View efficiently generates accurate cell clusters, especially in CITE-seq data with routine numbers of surface protein features, by preventing variations in a single surface protein feature from affecting results. Finally, we utilized this method to integrate single-cell transcriptional and protein expression data from SARS-CoV-2-infected patients, revealing antigen-specific B cell subsets after infection. Our results suggest LinQ-View could be helpful for multimodal analysis and purity assessment of CITE-seq datasets that target specific cell populations (e.g., B cells).Consumer wearables, such as smart watches, are a promising tool for monitoring circadian health in "real world" settings. Bowman et al. demonstrate that circadian signals can be accurately captured through heart rate data obtained from wearables, opening up new possibilities for population-level studies on heart rate and circadian rhythm. With the recent events following the pandemic COVID-19, global awareness about the use of biosafety materials has been in raise. Leather industry being a major commodity-driven sector, its role in addressing the issues concerning the safe use of leather products has become inevitable for the sustainability of the industry. A significant number of researches have been conducted to fabricate bio-safe leather by incorporating different types of antimicrobial agents during leather manufacturing. Besides, the increasing diversity in the development of synthetic materials and the impact of COVID-19 outbreak on automotive industry may create more demand from customers for incorporating different functionalities in leather without losing its inherent properties. Some of the key functionalities discussed include resistance to microbial growth, self-cleaning through superhydrophobicity and photocatalysis, thermal regulation, flame retardance and scented leather. This review focusses on the fabrication of such advanced Knowledge obtained from this review on fabrication of bio-safety leather against bacteria, mold and fungi would help further to integrate the antiviral property into the same which is a global need. Also, fabrication of functionalized leather would open new avenues for leather manufactures to venture into the development of advanced leather products such as flexible electronics, radiation shielding and fire fighting garments etc.
Practice-based experiences documenting development and implementation of nutrition and health surveillance systems are needed.

To describe processes, methods, and lessons learned from developing and implementing a population-based household nutrition and health surveillance system in Guatemala.

The phases and methods for the design and implementation of the surveillance system are described. Efforts to institutionalize the system in government institutions are described, and illustrative examples describing different data uses, and lessons learned are provided.

After initial assessments of data needs and consultations with officials in government institutions and partners in the country, a population-based nutrition surveillance system prototype with complex sampling was designed and tested in 5 Guatemalan Highland departments in 2011. After dissemination of the prototype, government and partners expanded the content, and multitopic nutrition and health surveillance cycles were collected in 2013, 2015, 2016, 2017/18, and 2018/19 providing nationally representative data for households, women of reproductive age (15-49 y), and children aged 0-59 mo.
