However, its role in normal mammary gland development has not been elucidated. Here, we used a fascin knockout mouse model to assess its role in normal mammary gland morphogenesis and lactation. Fascin knockout was not embryonically lethal, and its effect on the litter size or condition at birth was minimal. However, litter survival until the weaning stage significantly depended on fascin expression solely in the nursing dams. Accordingly, pups that nursed from fascin-/- dams had smaller milk spots in their abdomen, suggesting a lactation defect in the nursing dams. Mammary gland whole-mounts of pregnant and lactating fascin-/- mice showed significantly reduced side branching and alveologenesis. Despite a typical composition of basal, luminal, and stromal subsets of mammary cells and normal ductal architecture of myoepithelial and luminal layers, the percentage of alveolar progenitors (ALDH+) in fascin-/- epithelial fraction was significantly reduced. Further in-depth analyses of fascin-/- mammary glands showed a significant reduction in the expression of Elf5, the master regulator of alveologenesis, and a decrease in the activity of its downstream target p-STAT5. In agreement, there was a significant reduction in the expression of the milk proteins, whey acidic protein (WAP), and β-casein in fascin-/- mammary glands. Collectively, our data demonstrate, for the first time, the physiological role of fascin in normal mammary gland lactogenesis, an addition that could reveal its contribution to breast cancer initiation and progression.Zinc oxide nanoparticles (ZnO NPs) were synthesized by a green method using Azadirachta indica leaf extract. The structure of the prepared ZnO (NPs) were characterized by FT-IR, XRD, SEM-EDX and TEM analyses. The biosynthesized ZnO (NPs) were then used as a catalyst for the synthesis of steroidal dihydropyrazole derivatives through a one-pot multicomponent reaction involving phenyl acetylene and hydrazine derivatives. The anticancer activity of newly synthesized compounds were evaluated against three cancer cell lines namely HeLa (human cervical carcinoma), Hep3B (human hepatocellular carcinoma) and MCF7 (human breast adenocarcinoma) by MTT assay. The tested compounds were found to be active against all cancer cell lines and less toxic towards normal peripheral blood mononuclear cells (PBMCs). Antioxidant activity have also been investigated via free radical scavenging ability using DPPH, FRAP and ABTS assay. The tested compounds were found to exhibit moderate to good antioxidant activity which increases with increase in the concentration of steroidal dihydropyrazoles. Among all the tested steroidal dihydropyrazoles, compound 17 is found to be most active.Arsenic (As) has become natural health hazard for millions of people across the world due to its distribution in the food chain. Naturally, it is present in different oxidative states of inorganic [As(V) and As(III)] and organic (DMA, MMA and TMA) forms. Among different mitigation approaches, microbe mediated mitigation of As toxicity is an effective and eco-friendly approach. The present study involves the characterization of bacterial strains containing arsenite methyltransferase (Pseudomonas oleovorans, B4.10); arsenate reductase (Sphingobacterium puteale, B4.22) and arsenite oxidase (Citrobacter sp., B5.12) activity with plant growth promoting (PGP) traits. Efficient reduction of grain As content by 61 % was observed due to inoculation of methyltransferase containing B4.10 as compared to B4.22 (47 %) and B5.12 (49 %). Reduced bioaccumulation of As in root (0.339) and shoot (0.166) in presence of B4.10 was found to be inversely related with translocation factor for Mn (3.28), Fe (0.073), and Se (1.82). Bioaccumulation of these micro elements was found to be associated with the modulated expression of different mineral transporters (OsIRT2, OsFRO2, OsTOM1, OsSultr4;1, and OsZIP2) in rice shoot. Improved dehydrogenase (407 %), and β-glucosidase (97 %) activity in presence of P. oleovorans (B4.10) as compared to arsenate reductase (198 and 50 %), and arsenite oxidase (134 and 69 %) containing bacteria was also observed. Our finding confers the potential of methyltransferase positive P. oleovorans (B4.10) for As stress amelioration. Reduced grain As uptake was found to be mediated by improved plant growth and nutrient uptake associated with enhanced soil microbial activity.Biochar aging affects the stability of soil carbon. Analyzing the effect of biochar on soil organic carbon (SOC) forms and their relations with microbial community assembly and carbon metabolism with time is helpful for soil carbon sequestration (by adapting the farm management approach). Four treatments with no, low, medium, and high biochar application rates (0 %, 1 %, 2 %, and 4 % of the total dry weight of topsoil before winter wheat planting, abbreviated as control, LB, MB, and HB, respectively) were conducted in the field. The SOC and particulate organic carbon positively correlated with the biochar application rate. Biochar decreased readily oxidizable carbon (P less then 0.05) after 8 months of application compared to the control; however, the difference disappeared with time. Biochar increased dissolved organic carbon (DOC) but had no effect on water- soluble organic carbon (WSOC); DOC and WSOC decreased with time. Furthermore, LB and HB stabilized the bacterial alpha diversities with time. Based on high-throughput sequencing, HB reduced the relative abundance of Actinobacteriota but increased that of Acidobacteria (P less then 0.05) after 12 months of biochar application. Time-wise, the bacterial community assembly was determined by deterministic processes that were significantly affected by the available nitrogen, DOC, or WSOC. Compared with the control, biochar decreased bacterial links and improved bacterial metabolism of phenolic acids and polymers with time, as evidenced by Biolog EcoPlates. Structural equation modeling revealed that the contribution of bacterial assembly processes to carbon metabolism changed with time. Microbial carbon metabolism was most positively influenced by differences in the composition of bacterial specialists. These findings reinforced that changes in soil labile organic carbon were time-dependent but not necessarilty affected by the biochar application rate.As a class of persistent organic pollutant, polybrominated diphenyl ethers (PBDEs) and their hydroxylated and methoxylated derivatives (OH-PBDEs and MeO-PBDEs) have been widely detected in soil environments. However, studies on the bioavailability and transformation of PBDEs and their derivatives in soil organisms remain scarce. In this study, a detailed kinetic investigation on the accumulation and biotransformations of BDE-47, 6-MeO-BDE-47 and 6-OH-BDE-47 in earthworms (Eisenia fetida) exposed to artificially contaminated soils was conducted. The uptake and elimination kinetics of BDE-47, 6-MeO-BDE-47 and 6-OH-BDE-47 by earthworms were in accordance with a one-compartment first-order kinetic model. The bioaccumulation factors (BAFs) followed the order 6-MeO-BDE-47 > 6-OH-BDE-47 > BDE-47. All three compounds could undergo step-by-step debromination to produce lower brominated analogs in earthworms. Both BDE-47 and 6-OH-BDE-47 could be transformed to MeO-PBDEs, whereas no transformation from 6-OH-BDE-47 or 6-MeO-BDE-47 to PBDEs or from BDE-47 and 6-MeO-BDE-47 to OH-PBDEs took place in the earthworms. Methoxylation was proposed as a potential metabolic pathway to form MeO-PBDEs in earthworms, with the metabolic rates for the methoxylation of BDE-47 and 6-OH-BDE-47 being 27.7 and 5.1 times greater, respectively, than that of the debromination metabolism. The isomers of 6-MeO-BDE-47 and 6-OH-BDE-47 were formed via the addition of methoxy/hydroxy groups or via bromine shifts on benzene ring in the earthworms. This study provides comprehensive information for a better understanding of the accumulation and biotransformation of PBDEs and their derivatives in earthworms.Due to its strategic position and direct relationship with multiple stakeholders, the HoReCa sector can be a crucial node for the promotion of sustainability in the agri-food chain. However, the sector is currently responsible for a high environmental, social, and economic load. A recent response to these impacts is the diffusion of initiatives that assess and promote sustainable practices at the catering business level. The study aims to verify, using a scientific methodology, how current sustainability programs enable the HoReCa sector to achieve the Sustainable Development Goals (SDGs) and the level of implementation of sustainable practices in the Italian context. To this end, seven currently available national and international sustainability programs were selected, analyzed, and compared. A matrix approach was adopted to understand the balance of the programs with respect to the three pillars of sustainability. A more in-depth analysis was conducted on the Italian Ristorazione Sostenibile 360 (RS 360) case study, which was analyzed both in terms of compliance with the SDGs and in terms of the sustainability performance of the 29 restaurants that participated in the pilot phase of the project. Although the programs differ from each other, they comply with the 2030 Agenda goals, with a particular focus on achieving Goal 12. Furthermore, the analysis highlighted how a balance between the social, environmental, and economic spheres is essential to ensure the sustainable growth of the sector and to bring benefits to the community. As for the case study, 76 % of the analyzed restaurants achieve at least 70 % of the maximum level of fulfillment of the SDGs achievable by the HoReCa sector. This is a symptom of the sector's potential to contribute to supply chain sustainability. Air pollution has become a significant public health concern. During exercise, many physiological factors are thought to increase the effects of air pollution. Air pollution most affects lung function and respiratory symptoms. We investigated the association between lung function, respiratory symptoms, and air pollutant concentration with meteorological factors in elite sports athletes.

A total of 59 elite sports athletes from the Korea National Sports University participated in this prospective, observational study from September 2019 to June 2020. At ten visits, lung function and respiratory symptoms were obtained after a training session. We measured six air pollutants, including SO
