This article describes development and testing of system based on fiber Bragg grating (FBG) sensors that can detect changes in strain and temperature. The first phase of the research has been focused on the development of new fiber optic sensors for the monitoring of concrete structures and their investigation in laboratory conditions. The work also shows novel applicability of the same FBG technology for glulam structures. Mechanical loading tests of the concrete beam as well as glulam beam with embedded sensors were carried out. Data measured by developed fiber optic sensors were compared with the readings from reference sensors as well as with the analytically calculated values. The achieved results proved good agreement between the measured data, analytical data and reference methods. In second phase of the research, the pilot installation of the sensors was carried out on the newly constructed prestressed-concrete bridge. The bridge was monitored throughout pre-stressing phase and monitoring continued after the completion of the construction works. Problems with the fragility of the sensors occurred during the measurements, but the obtained results provide a good basis for further improvement of the system.Long-chain fatty acyl-CoA synthetase (ACSLs) is an essential enzyme for the synthesis of fatty acyl-CoA. ACSL1 plays a key role in the synthesis of triglycerides, phospholipids, and cholesterol esters. BACKGROUND In the current study, triglyceride content did not increase after overexpression of the ACSL1 gene. METHODS RNA-seq and lipid metabolome profiling were performed to determine why triglyceride levels did not change with ACSL1 overexpression. RESULTS Fatty acyl-CoA produced by ACSL1 was determined to be involved in the diglyceride synthesis pathway, and diglyceride content significantly increased when ACSL1 was overexpressed. Moreover, the arachidonic acid (AA) content in sheep adipocytes significantly increased, and the level of cyclooxygenase 2 (COX2) expression, the downstream metabolic gene, was significantly downregulated. Knocking down the ACSL1 gene was associated with an increase in COX2 mRNA expression, as well as an increase in prostaglandin content, which is the downstream metabolite of AA. CONCLUSIONS The overexpression of the ACSL1 gene promotes the production of AA via downregulation of COX2 gene expression.Asphalt pavement consists of multiple layers of asphalt with a progressive decrease of nominal maximum aggregate size from the bottom to the top, which can be constructed by the double-decked or the conventional paving method (i.e., layer by layer). Reliable interface strength between the fine- and the coarse-grained layer of asphalt mixture is prerequisite to ensure the serviceability of the asphalt pavement. This study aims to compare the interface behavior of the asphalt pavement constructed by the conventional and the double-decked paving methods through laboratory and trial pavement tests. Laboratory test results show that the interface strength of the specimen prepared by the double-decked paving method is mainly contributed by the interlocking of the coarse- and the fine-grained asphalt mixture, fundamentally different from the conventional paving method, in which the interface strength is mainly provided by the tack coat oil. More importantly, the interface shear strength and the uniaxial tensile strength of specimens prepared by the double-decked paving method are about 1.5-1.8 times larger than that of specimens prepared by the conventional paving method. To verify the applicability of laboratory experimental findings to the practical engineering, a trial road was paved in situ using both double-decked and conventional paving methods. Cored specimens were collected from the trial road and their interface strengths are tested. Comparisons of the interface strength obtained from cored specimens further prove that the asphalt pavement constructed by the double-decked paving method has larger interface strength than that of the asphalt pavement constructed by the conventional paving method.Remote sensing is commonly performed via airborne platforms such as satellites, specialized aircraft, and unmanned aerial systems (UASs), which perform airborne photography using mounted cameras. However, they are limited by their coverage (UASs), irregular flyover frequency (aircraft), and/or low spatial resolution (satellites) due to their high altitude. In this paper, we examine the utilization of commercial flights as an airborne platform for remote sensing. Namely, we simulate a situation where all aircraft on commercial flights are equipped with a mounted camera used for airborne photography. The simulation is used to estimate coverage, the temporal and spatial resolution of aerial imagery acquired this way, as well as the storage capacity required for storing all imagery data. The results show that Europe is 83.28 percent covered with an average of one aerial photography every half an hour and a ground sampling distance of 0.96 meters per pixel. The present study focused on filling this knowledge gap by examining the roles of abscisic acid (ABA) and jasmonate in the regulation of senescence of fine, absorptive roots and leaves of Populus trichocarpa. Immunohistochemical (IHC), chromatographic, and molecular methods were utilized to achieve this objective. A transcriptomic analysis identified significant changes in gene expression that were associated with the metabolism and signal transduction of phytohormones, especially ABA and jasmonate. The increased level of these phytohormones during senescence was detected in both organs and was confirmed by IHC. Based on the obtained data, we suggest that phytohormonal regulation of senescence in roots and leaves is organ-specific. We have shown that the regulation of ABA and JA metabolism is tightly regulated during senescence processes in both leaves and roots. The results were discussed with respect to the role of ABA in cold tolerance and the role of JA in resistance to pathogens.The epidemiological research relating mortality and hospital admissions to ambient temperature is well established. However, less is known about the effect temperature has on Accident and Emergency (A&E) department attendances. Time-series regression analyses were conducted to investigate the effect of temperature for a range of cause- and age-specific attendances in Greater London (LD) between 2007 to 2012. A seasonally adjusted Poisson regression model was used to estimate the percent change in daily attendances per 1 °C increase in temperature. The risk of overall attendance increased by 1.0% (95% CI 0.8, 1.4) for all ages and 1.4% (1.2, 1.5) among 0- to 15-year-olds. A smaller but significant increase in risk was found for cardiac, respiratory, cerebrovascular and psychiatric presentations. Importantly, for fracture-related attendances, the risk rose by 1.1% (0.7, 1.5) per 1 °C increase in temperature above the identified temperature threshold of 16 °C, with the highest increase of 2.1% (1.5, 3.0) seen among 0- to 15-year-olds. There is a positive association between increasing temperatures and A&E department attendance, with the risk appearing highest in children and the most deprived areas. The performance of four machine learning algorithms-random forest, support vector machine, deep belief nets, and kernel-based extreme learning machine (KELM)-in evaluating data collected from tea leaves cultivated under different shade treatments was tested. KELM performed best with a root-mean-square error of 8.94 ± 3.05 μg cm-2 and performance to deviation values from 1.70 to 8.04 for the test data. In the present study, two 10-point augmented simplex-centroid designs were used to formulate a three-component mixture system using water, atorvastatin, and Diclofenaco-lepori or Optiben. The amoebicidal effects and in vitro-induced toxicity in a eukaryotic cell line were determined for all experiments. The optimal mixture to inhibit the parasite without inducing toxicity was established in the first plan as 30% Optiben, 63.5% atorvastatin, and 3.1% water. As for the second experimental design, the optimal mixture to inhibit Acanthamoeba with lower toxicity effect was composed of 17.6% Diclofenaco-lepori and 82.4% atorvastatin.
