Understanding factors that influence food choices in early childhood is necessary to promote healthy food choices. As parents are the main gatekeepers of children's food environment, this study aimed to explore parents/caregivers' perspectives on the factors that influence their children's food choices. Thirteen focus groups adopting a partial nominal group technique were conducted with 110 parents/caregivers of children aged 2-6 years living in an urban area, Kurunegala District, Sri Lanka. Parents/caregivers were provided with 22 potential factors and asked to rank six most influential and choose six least influential factors on their child's food decisions. Responses were discussed as a group and audio recorded discussions underwent content analysis. Child food preference was ranked as the core driving factor in food decisions of preschool children. Family income, nutritional knowledge of the family, maternal control of food choices and household food preparation facilities were ranked as the next four most influential. Most frequent choices of least influential factors were individual characteristics of child including sex and nutritional knowledge; and community (environmental) characteristics including seasonal food availability and community cultural food beliefs. Additional influential factors proposed were having a single child, a supportive family network and healthy menu suggestions by the preschool centres. Findings suggest that modifying the food preference of the child and the family characteristics need to be central for effective eating interventions. Because of their potential as novel antibiotic agents, antimicrobial peptides (AMPs) have generated considerable interest. The mechanism of bacterial toxicity of AMPs often involves the disruption and/or permeabilization of the bacterial membrane; even those that act intracellularly first have to traverse the membrane. In this work we have explored the incorporation of the fluorinated aromatic amino acids fluoro-Phe and fluoro-Tyr into the Trp- and Arg-rich AMP tritrpticin, and investigated their role in the membrane binding properties and the antimicrobial activity of the peptide. Fluorinated peptides were obtained with good yield by recombinant expression of tritrpticin as a calmodulin-fusion protein in Escherichia coli. Cells were grown in the presence of glyphosate, an inhibitor of aromatic amino acid biosynthesis, and the peptides were released by proteolysis from the purified fusion protein. By using SDS micelles, as a simplified model of the bacterial cytoplasmic membrane, we could study the peptide-membrane interactions and the preferred location of individual fluorinated residues in the micelles by 19F NMR spectroscopy. Solvent-perturbation 19F NMR measurements revealed that para-fluoro-Phe residues are embedded deeply in the hydrophobic region of the micelles. On the other hand, 3-fluoro-Tyr residues introduced in tritrpticin were located near the surface of the micelles with high solvent exposure, while 2-fluoro-Tyr sidechains were less solvent exposed. In combination with the outcome of determinations of their antimicrobial activity, our 19F NMR results indicate that the higher solvent exposure of Tyr residues correlates with a decrease of the antimicrobial potency. This different role of Tyr can likely be extended from tritrpticin to other cationic AMPs. It is widely accepted that the abnormal self-association of amyloid β-protein (Aβ) is central to the pathogenesis of Alzheimer's disease, the most common form of dementia. Accumulating evidence, both in vivo and in vitro, suggests that the binding of Aβ to gangliosides, especially monosialoganglioside GM1, plays an important role in the aggregation of Aβ. This review summarizes the molecular details of the binding of Aβ to ganglioside-containing membranes and subsequent structural changes, as revealed by liposomal and cellular studies. Furthermore, mechanisms of cytotoxicity by aggregated Aβ are also discussed. Membrane lipids are inherently highly dynamic molecules. Currently, it is difficult to probe the structures of individual lipids experimentally at the timescales corresponding to atomic motions, and consequently molecular dynamics simulations are used widely. In our previous work, we have introduced the principal component analysis (PCA) as a convenient framework for comprehensive quantitative description of lipid motions. Here, we present a newly developed open source script, PCAlipids, which automates the analysis and allows us to refine the approach and test its limitations. We use PCAlipids to determine the influence of temperature, cholesterol and curvature on individual lipids, and show that the most prominent lipid tail scissoring motion is strongly affected by these factors and allows tracking of phase transition. Addition of cholesterol affects the conformations and selectively changes the dynamics of lipid molecules, impacting the large-amplitude motions. Introduction of curvature biases the conformational ensembles towards more extended structures.
