The findings presented here offer novel molecular insights about the cell cycle regulation during the CHO cell bioprocessing and its implications for increased recombinant protein production. This data provides a background for engineering productivity-enhanced CHO cell lines for continuous manufacturing.Biotechnology has revolutionized science and health care by providing new biomolecules with biological and medical applications. However, the low stability of several life-saving bioproducts still hinders their transport, storage, and application. Hence, protein-based bioproducts instability and high costs are the main bottlenecks limiting access to biopharmaceuticals in low-income countries and communities. Aiming to improve the stability of protein-based products, researchers have studied ionic liquids (ILs) as protein stabilizers due to their unique properties and ability to enhance the solubility and stability of a wide range of biomolecules. Although different classes of ILs have the potential to improve protein stability, their effects are dependent on several variables, such as the complex and intrinsic properties of proteins, the nature and concentration of ILs, and environmental conditions (e.g., temperature, pH). For medical applications, the biocompatibility of ILs can also limit their biological use. Therefore, the current state-of-the-art on ILs applications for non-enzymatic protein stabilization was carefully analyzed and discussed, considering protein properties, ILs classes, and IL solutions concentrations. Lastly, a critical perspective regarding ILs applications as protein stabilizers was presented, highlighting the current lacunas in the field while guiding future studies to answer the existing paradigms. The diagnosis of neurological diseases is one of the biggest challenges in modern medicine, which is a major issue at the moment. Electroencephalography (EEG) recordings is usually used to identify various neurological diseases. EEG produces a large volume of multi-channel time-series data that neurologists visually analyze to identify and understand abnormalities within the brain and how they propagate. This is a time-consuming, error-prone, subjective, and exhausting process. Moreover, recent advances in EEG classification have mostly focused on classifying patients of a specific disease from healthy subjects using EEG data, which is not cost effective as it requires multiple systems for checking a subject's EEG data for different neurological disorders. This forces researchers to advance their work and create a single, unified classification framework for identifying various neurological diseases from EEG signal data. Hence, this study aims to meet this requirement by developing a machine learning (ML) baso identify various neurological abnormalities from brain signal data.
Exercise training protects against heart failure. However, the mechanism underlying the protective effect of exercise training on angiotensin II (Ang II)-induced cardiac fibrosis remains unclear.

An exercise model involving C57BL/6N mice and 6 weeks of treadmill training was used. Ang II (1.44 mg/kg/day) was administered to induce cardiac fibrosis. RNA sequencing and bioinformatic analysis were used to identify the key factors mediating the effects of exercise training on cardiac fibrosis. Primary adult mouse cardiac fibroblasts (CFs) were used in vitro. Adeno-associated virus serotype 9 was used to overexpress POU domain, class 2, transcription factor 1 (POU2F1) in vivo.

Exercise training attenuated Ang II-induced cardiac fibrosis and reversed 39 gene expression changes. The transcription factor regulating the largest number of these genes was POU2F1. Compared to controls, POU2F1 was shown to be significantly upregulated by Ang II, which is itself reduced by exercise training. In vivo, POU2F1 overexpression nullified the benefits of exercise training on cardiac fibrosis. In CFs, POU2F1 promoted cardiac fibrosis. CCAAT enhancer-binding protein β (C/EBPβ) was predicted to be the transcription factor of POU2F1 and verified using a dual-luciferase reporter assay. In vivo, exercise training activated AMP-activated protein kinase (AMPK) and alleviated the increase in C/EBPβ induced by Ang II. In CFs, AMPK agonist inhibited the increase in C/EBPβ and POU2F1 induced by Ang II, whereas AMPK inhibitor reversed this effect.

Exercise training attenuates Ang II-induced cardiac fibrosis by reducing POU2F1. Exercise training inhibits POU2F1 by activating AMPK, which is followed by the downregulation of C/EBPβ, the transcription factor of POU2F1.

Drawing on the experiences of expeditioners in the Australian Antarctic Program from 2019-21, this article provides a counter to popular media perspective by exploring how COVID-19 protocols-including quarantine and social distancing-affected expeditioners' individual well-being and their experiences of the social environment. We argue that Antarctic life during COVID-19 has not been as detached from the rest of the world nor as heroic as the popular media has suggested, but nonetheless provides important insights for survival in isolated, confined, and extreme environments (ICE) and non-ICE environments at a time of pandemic.
Microbiological analysis of body fluids (BF) provides important information for diagnosis of infection. We evaluated the analytical performance of bacterial count by UF-4000 BF mode for ascitic, cerebrospinal, pleural, synovial and continuous ambulatory peritoneal dialysis fluids compared to classical microbiological procedure (direct Gram staining and culture).

For the 1,734 BF analyzed, distribution of UF-4000 bacterial count was analyzed according to the level of growth culture and results were compared using Mann-Whitney test. ROC curves analysis allowed to define the best cut-off value to predict or exclude positive culture for each type of BF.

UF-4000 bacterial counts were significantly lower in sterile than in infected BFs (p<0.00001) and correlated with the level of growth on culture. The ROC curves of bacteria/μL and culture positivity yielded area under the curve >0.80 for each type of BF. Optimal cut-offs were chosen with excellent statistical parameters (sensitivity ranging from 0.70 to 0.86, specificity from 0.78 to 0.98, negative predictive value >0.95 and Youden index >0.55).

For BF, UF-4000 bacterial count correlate with culture results and is a discriminative method enhancing detection of microbiological etiology. It could be used as a screening method based on the cut-off values proposed in this study.

The 16p11.2 copy number variations (CNVs) are increasingly recognized as one of the most frequent genomic disorders, with a broad spectrum of phenotypes. The fetal phenotype associated with 16p11.2 CNVs is poorly described. The current study presents prenatal series of 16p11.2 CNVs and provides a better understanding of this submicroscopic imbalance in prenatal diagnosis.

Retrospective case series were extracted from a single tertiary referral center performing prenatal single nucleotide polymorphism (SNP) array from April 2017 to December 2021. The maternal demographics, indication for amniocentesis, ultrasound findings, SNP array results, inheritance of the CNVs, and pregnancy outcomes were studied.

We indentified 30 fetuses carrying 16p11.2 CNVs, representing 0.35% (30/8578) of prenatal SNP array results. The series included 17 fetuses with a proximal deletion, 7 with a distal deletion, 4 with a proximal duplication, and 2 with a distal duplication. Prenatal ultrasound anomalies were reported in 80% ts with a milder or normal phenotype. Our results underline that vertebral deformities were frequent in cases of 16p11.2 proximal deletion, and further demonstrate the incomplete penetrance of the CNVs.Genetic and phylogenetic studies indicated that Zika virus (ZIKV) has evolved into 2 major lineages, the African and Asian. However, ZIKV has been described as a single serotype. This study aimed at assessing the cross-neutralization between ZIKV African and Asian lineages strains. Sixthy-five samples collected in 2007 and 30 samples collected from the same subjects in 2011/2012 in West Africa and positive to neutralizing antibody against ZIKV MR-766 strain (African lineage) were tested against ZIKV H/PF/2013 strain (Asian lineage) by microneutralization assay. All samples showing neutralizing antibodies against MR-766 strain showed also neutralizing activity against H/PF/2013 strain, although with lower titers. This is consistent with about 120 amino acid differences between the two strains. Despite differences in the magnitude of neutralizing activity against different ZIKV strains, all samples showed neutralizing antibody titers considered to be protective.Network diffusion processes or how information spreads through networks have been widely examined in numerous disciplines such as epidemiology, physics, sociology, politics, or computer science. In this paper, we extend previous developments by considering a generalization of the diffusion by considering the possibility of differences in the speed of diffusion and reduction depending on the forces' directions. In this situation, the differential speed of diffusion produces deviations from the standard solution around the average of the initial conditions in the network. In fact, this asymmetry gives rise to non-linear dynamics in which, contrary to the symmetric case, the final solution depends on the topology of the graph as well as on the distribution of the initial values. Counter-intuitively, less central nodes in the network are able to exert a higher influence on the final solution. This behavior applies also for different simulated networks such as random, small-world, and scale-free. We show an example of this kind of asymmetric diffusion process in a real case. To do so, we use a network of US Boards of Directors, where boards are the nodes and the directors working for more than one board, are the links. Changes in the proportion of women serving on each board are influenced by the gradient between adjacent boards. We also show that there is an asymmetry the gradient is reduced at a slower (faster) rhythm if the board has less (more) women than neighboring boards. We are able to quantify the accumulated effect of this asymmetry from 2000 to 2015 in the overall proportion of women on boards, in a 4.7 percentage points (the proportion should have been an 14.61% instead of the observed 9.93% in 2015).
