Deletion of Foxa1 within the mouse mammary gland brings about abnormal accumulation regarding luminal progenitor cellular material: a link in between the reproductive system elements and ER-/TNBC breast cancer?
Thermal hydrolysis (TH) treatment has been verified to effectively reduce antibiotic resistance genes (ARGs) in waste activated sludge. This study focused on the effect of TH parameters on ARGs reduction, and the optimal conditions basing on tetracycline (tet) resistance genes reduction rate in sludge phase were pH 3, temperature 160 °C and reaction time 2 h. The pH was found to play a critical role in tet genes reduction behavior. Additionally, the tet genes distributions in TH supernatant are considered as well because genes will release from solid to liquid phase as sludge cell breaking. The lowest genes content in TH supernatant was also observed at pH 3. Further investigation revealed that contents of DNA and organics released from sludge phase to liquid phase under pH 3 were less than that in neutral and alkaline condition; moreover, the organics in TH supernatant was found to protect DNA. Therefore, under the acidic condition, the genes had less releasing from the sludge phase and the DNA protection effect of organics was weaker, resulting in the lowest tet genes content in TH supernatant. Moreover, bacterial community structure after TH was closely related with the ARGs content. The bacterial on phylum and genus level showed various responses to TH pH values. Although the Firmicutes phylum and Escherichia-Shigella genus exhibited the stronger resistance and had higher accumulation in TH acidic condition, more possible total tet genes hosts were destroyed in acidic condition, causing less ARGs remaining in the sludge phase under pH 3.It is little known about the lncRNA-PVT1 effect on occupational pulmonary fibrosis, although researches show it plays an essential role in cancer. Studies reveal that lung fibroblast activation is one of the key events in silica-induced fibrosis. Here, we found that lncRNA-PVT1 promoted the proliferation, activation, and migration of lung fibroblasts. The isolation of cytoplasmic and nuclear RNA assay and fluorescence in situ hybridization experiment showed that lncRNA-PVT1 was abundantly expressed in the cytoplasm. Luciferase reporter gene assay and RNA pull-down experiment indicated that the cytoplasmic-localized lncRNA-PVT1 could competitively bind miR-497-5p. MiR-497-5p was further observed to attenuate silica-induced pulmonary fibrosis by targeting Smad3 and Bcl2. Moreover, the transcription factor FOXM1 acted as a profibrotic factor by elevating lncRNA-PVT1 transcription in lung fibroblasts. Inhibition of FOXM1 expression with thiostrepton alleviated silica-induced pulmonary fibrosis in vivo. Collectively, we revealed that FOXM1-facilitated lncRNA-PVT1 activates lung fibroblasts via miR-497-5p during silica-induced pulmonary fibrosis, which may provide potential therapeutic targets for pulmonary fibrosis.Awareness of risks posed by widespread presence of nanoplastics (NPs) and bioavailability and potential to interact with organic pollutants has been increasing. Inhalation is one of the more important pathways of exposure of humans to NPs. In this study, combined toxicity of concentrations of polystyrene NPs and various phthalate esters (PAEs), some of the most common plasticizers, including dibutyl phthalate (DBP) and di-(2-ethyl hexyl) phthalate (DEHP) on human lung epithelial A549 cells were investigated. When co-exposed, 20 μg NPs/mL increased viabilities of cells exposed to either DBP or DEHP and the modulation of toxic potency of DEHP was greater than that of DBP, while the 200 μg NPs/mL resulted in lesser viability of cells. PAEs sorbed to NPs decreased free phase concentrations (Cfree) of PAEs, which resulted in a corresponding lesser bioavailability and joint toxicity at the lesser concentration of NPs. The opposite effect was observed at the greater concentration of NPs, which may result from the dominated role of NPs in the combined toxicity. Furthermore, our data showed that oxidative stress and inflammatory reactions were mechanisms for combined cytotoxicities of PAEs and NPs on A549 cells. Results of this study emphasized the combined toxic effects and mechanisms on human lung cells, which are helpful for assessing the risk of the co-exposure of NPs and organic contaminants in humans.Mosquito larvicides are an effective tool for reducing numbers of adult females that bite and potentially spread pathogenic organisms. Methionine, an essential amino acid in humans, has been previously demonstrated to be a highly effective larvicide against four (4) mosquito species in three (3) genera, Anopheles, Culex and Aedes. The aim of the present study was to determine the potential impact on non-target aquatic organisms of methionine applied as a mosquito larvicide. DL-methionine concentrations ranging from 0.06% to 1.00% were used; wherein the highest concentration of 1.00% would result in 100% mortality within 48 h in mosquitoes. Acute toxicity assays were conducted in accordance with the US Environmental Protection Agency (US EPA) guidelines for the water flea (Daphnia magna Straus; Cladocera Daphniidae) and the fathead minnow (Pimephales promelas Rafinesque; Cypriniformes Cyprinidae). Tocilizumab datasheet Water fleas and fish were placed directly into the solutions in glass containers and tanks for 48-hours and 96-hours, respectively. When applied within the above-mentioned range of effective mosquito larvicide concentrations, DL-methionine meets US EPA criteria as a "practically non-toxic" pesticide for both species. These results suggest that methionine is a viable alternative to current mosquito larvicide options, which are typically classified as moderately to highly toxic and may be a valuable addition to a mosquito integrated pest management program. In this study, we have evaluated the role of karrikin (KAR1) against the absorption and translocation of a persistent organic pollutant (POP), 2,4,4'-Tribromodiphenyl ether (BDE-28) in plants, in the presence of two other stressors, cadmium (Cd) and high temperature. Furthermore, it correlates the physiological damages of Brassica alboglabra with the three stresssors separately. The results revealed that the post-germination application of KAR1 successfully augmented the growth (200%) and pertinent physiochemical parameters of B. alboglabra. KAR1 hindered air absorption of BDE-28 in plant tissues, and reduced its translocation coefficient (TF). Moreover, BDE-28 was the most negatively correlated (-0.9) stressor with chlorophyll contents, while the maximum mitigation by KAR1 was also achieved agaist BDE-28. The effect of temperature was more severe on soluble sugars (0.51), antioxidative machinery (-0.43), and osmoregulators (0.24). Cd exhibited a stronger inverse interrelation with the enzymatic antioxidant cascade.
